Effects of Different Inositol Levels on β-Catenin and Hormone-Sensitive Triglayceride Lipase Expression in Skin of Rex Rabbits

Abstract:The aim of the present study was to investigate the effects of different inositol levels in diets on the expression of β-catenin and hormone-sensitive triglayceride lipase (HSL) in skin of Rex rabbits. A total of 120 (40±1)-day-old Rex rabbits (half male and half female) were randomly divided into 4 groups with 30 rabbits per group. Rex rabbits in 4 groups were fed diets with different levels of inositol (0, 25, 50 and 75 mg/kg, respectively), and the experiment lasted for 3 months. The skin of abdomen, dorsocentral region, hip were collected on days 30 (2-month-old), 60 (3-month-old), 90 (4-month-old), respectively, and the protein expression and location of β-catenin and HSL were studied by immunohistochemistry and Western blot methods. The results showed that the β-catenin was widely expressed in hair follicle, and the hair root sheath cells and hair papilla all had brown positive reaction cells. HSL was showed a very obvious dark brown strong positive expression in hair root sheath cells, especially in inner root sheath cells. The addition of 50 mg/kg inositol could extremely significantly increase the mean gray value of β-catenin and HSL positive cells in the hair follicles of the dorsocentral region skin of 2- to 4-month-old Rex rabbits (P<0.01), extremely significantly increase the mean gray value of β-catenin and HSL positive cells in the hair follicles of the dorsocentral region, abdomen and hip skin of 4-month-old Rex rabbits (P<0.01). The results suggest that inositol can promote the growth of hair follicle by increasing the expresstion of β-catenin and HSL of 4-month-old Rex rabbits, and the effect is the best when dietary inositol level is 50 mg/kg in this experiment.
