无氮饲粮对家兔肠道消化酶活性和盲肠微生物区系的影响

摘要 本试验旨在通过研究无氮饲粮对家兔肠道消化酶活性和盲肠微生物区系的影响,为家兔内源氮代谢的研究提供参考。选取1.5岁、健康状况良好的獭兔、肉兔、毛兔各20只(公母各占1/2),将每个种类的家兔按照性别随机分成2组,分别饲喂基础饲粮和无氮饲粮,每组10个重复,每个重复1只。预试期和正试期各7 d。结果显示:1)3种家兔无氮饲粮组肠道淀粉酶和纤维素酶活性均高于基础饲粮组,其中獭兔、肉兔和毛兔分别提高了8.00%、4.08%和20.69%以及12.45%、12.19%和9.47%,但均无显著差异(P>0.05)。3种家兔无氮饲粮组肠道胰蛋白酶和脂肪酶活性均低于基础饲粮组,其中獭兔、肉兔和毛兔分别降低了5.47%、8.20%和9.98%以及4.34%、0.96%和8.97%,但均无显著差异(P>0.05)。2)在盲肠菌群结构方面,无氮饲粮组獭兔拟杆菌属(Bacteroides)、瘤胃球菌科NK4A214群(Ruminococcaceae_NK4A214_group)和艾克曼菌属(Akkermansia)的相对丰度均高于基础饲粮组,瘤胃球菌属1(Ruminococcus_1)、毛螺菌科NK4A136群(Lachnospiraceae_NK4A136_group)的相对丰度均低于基础饲粮组。无氮饲粮组肉兔拟杆菌属和艾克曼菌属的相对丰度均高于基础饲粮组,瘤胃球菌属1、毛螺菌科NK4A136群和不可培养紫单胞菌科(uncultured_bacterium_f_Porphyromonadaceae)的相对丰度均低于基础饲粮组。无氮饲粮组毛兔拟杆菌属、艾克曼菌属和Lachnoclostridium的相对丰度均高于基础饲粮组,不可培养拟杆菌目S24-7群(uncultured_bacterium_f_Bacteroidales_S24-7_group)和瘤胃球菌属1的相对丰度均低于基础饲粮组。综上所述,无氮饲粮对家兔的肠道消化酶活性和盲肠微生物区系均有一定影响,应用无氮饲粮法研究家兔内源氮代谢时应综合考虑这些影响。

Abstract: This experiment was to investigate the effects of nitrogen-free diets on intestinal digestive enzyme activities and cecal microflora of rabbits, and to provide a reference for the study of endogenous nitrogen metabolism in rabbits. Twenty 1.5-year-old healthy Rex rabbits, meat rabbits and wool rabbits were selected, and the rabbits in each variety were randomly assigned to 2 groups based on gender. The rabbits in one group were fed a basal diet, while the rabbits in the other group were fed a nitrogen-free diet, each group had 10 replicates and each replicate had 1 rabbit. Both pretrial and formal trial periods lasted for 7 days. The results showed as follows:1) the intestinal amylase and cellulase activities of 3 kinds of rabbits in nitrogen-free diet group were higher than those in the basal diet group, and those of Rex rabbits, meat rabbits and wool rabbits increased by 8.00%, 4.08%, 20.69% and 12.45%, 12.19%, 9.47%, respectively, but the differences were not significant(P>0.05). The intestinal trypsin and lipase activities of 3 kinds of rabbits in nitrogen-free diet group were lower than those in the basal diet group, and those of Rex rabbits, meat rabbits and wool rabbits were 5.47%, 8.20%, 9.98% and 4.33%, 0.96%, 8.97%, respectively, but the differences were not significant (P>0.05). 2) Compared with the basal diet group, the cecal microbe diversity of Rex rabbits and meat rabbits trended to decrease, while that of wool rabbits trend to increase. For cecal microflora structure, the relative abundances of Bacteroides, Ruminococcaceae_NK4A214_group and Akkermansia of Rex rabbits in the nitrogen-free diet group were higher than those in the basal diet group, while the relative abundances of Ruminococcus_1, Lachnospiraceae_NK4A136_group were lower than those in the basal diet group. The relative abundances of Bacteroides and Akkermansia of meat rabbits in the nitrogen-free diet group were higher than those in the basal diet group, while the relative abundances of Ruminococcus_1, Lachnospiraceae_NK4A136_group, uncultured_bacterium_f_Porphyromonadaceae were lower than those in the basal diet group. The relative abundances of Bacteroides, Akkermansia and Lachnoclostridium of wool rabbits in the nitrogen-free diet group were higher than those in the basal diet group, while the relative abundances of uncultured_bacterium_f_Bacteroidales_S24-7_group and Ruminococcus_1 were lower than those in the basal diet group. In summary, nitrogen-free diet has some effects on intestinal digestive enzyme activities and cecal microflora of rabbits. These effects should be considered comprehensively when using the nitrogen-free diet method to study endogenous nitrogen metabolism in rabbits.
