低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪生长性能、胴体性状、肉品质及氮排放的影响

摘要: 本试验旨在研究低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪生长性能、胴体性状、肉品质及氮排放的影响。试验1:选取健康、初始体重[(78.19±6.69)kg]相近的"杜×长×大"肥育猪108头,随机分为6组,每组6个重复,每个重复3头猪。组1为正常蛋白质水平饲粮组,即12.5%粗蛋白质(CP)+0.73%标准回肠可消化赖氨酸(SID-Lys);组2、3、4、5和6为低蛋白质水平饲粮(10.5% CP)不同SID-Lys组,SID-Lys水平分别为0.58%、0.66%、0.73%、0.80%和0.88%。试验期28 d。试验2:选取初始体重为(90.56±3.28)kg的"杜×长×大"肥育猪18头,随机分为3组,每组6个重复,每个重复1头猪。3组分别饲喂试验1中组1、4和5的饲粮,试验期共7 d,其中预试期3 d,氮平衡试验期4 d。结果表明:1)降低饲粮蛋白质水平对肥育猪的生长性能无显著影响(P>0.05)。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪的生长性能无显著影响(P>0.05)。2)降低饲粮蛋白质水平对肥育猪胴体性状无显著影响(P>0.05),但显著影响肥育猪背最长肌的红度值(P < 0.05)。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪胴体性状无显著影响(P>0.05),但显著影响肥育猪背最长肌的滴水损失(P < 0.05)。3)降低饲粮蛋白质水平显著影响了肥育猪的粪氮排放、尿氮排放和总氮排放(P < 0.05)。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪粪氮排放、尿氮排放和总氮排放均无显著影响(P>0.05)。由此可见,降低饲粮蛋白质水平对肥育猪的生长性能和胴体性状无显著影响,且可改善肉色,降低总氮排放;低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪生长性能、胴体性状和氮排放无显著影响,但可改善肉品质。

Effects of Different Supplemental Levels of Lysine in Low Protein Diets on Performance, Carcass Traits, Meat Quality and Nitrogen Excretion of Finishing Pigs

Abstract: This study was conducted to investigate the effect of different supplemental levels of lysine in low protein diets on performance, carcass traits, meat quality and nitrogen excretion of finishing pigs. In experiment 1, a total of 108 healthy "Durco×Landrace×Large White" finishing pigs with average initial body weight of (78.19±6.69) kg were assigned into 6 groups with 6 replicates per group and 3 pigs per replicate. Group 1 was normal protein level diet group, which was 12.5% crude protein (CP) and 0.73% standardized ileal digestible lysine (SID-Lys); groups 2, 3, 4, 5 and 6 were low protein level diet (10.5% CP) and different SID-Lys groups with the SID-Lys levels were 0.58%, 0.66%, 0.73%, 0.80% and 0.88%, respectively. The experiment lasted for 28 days. In experiment 2, eighteen "Durco×Landrace×Large White" finishing pigs with average initial body weight of (90.56±3.28) kg were assigned into 3 groups with 6 replicates per group and 1 pig per replicate. Pigs in the 3 groups were fed the same diets of groups 1, 4 and 5 in experiment 1, respectively. The experiment lasted for 7 days include 3 days preliminary experiment period and 4 days nitrogen balance experiment period. The results showed as follows:1) reducing dietary protein level had no significant effects on growth performance of finishing pigs (P>0.05). Different supplemental levels of lysine in low protein diets had no significant effects on growth performance of finishing pigs (P>0.05). 2) Reducing dietary protein level had no significant effects on carcass traits of finishing pigs (P>0.05), but significant affected the Longissimus dorsi redness value of finishing pigs (P < 0.05). Different supplemental levels of lysine in low protein diets had no significant effects on carcass traits of finishing pigs (P>0.05), but significant affected the Longissimus dorsi drip loss of finishing pigs (P < 0.05). 3) Reducing dietary protein level significant affected the fecal nitrogen excretion, urine nitrogen excretion and total nitrogen excretion of finishing pigs (P < 0.05). Different supplemental levels of lysine in low protein diets had no significant effects on fecal nitrogen excretion, urine nitrogen excretion and total nitrogen excretion of finishing pigs (P>0.05). Collectively, reducing dietary protein level have no significant effects on growth performance and carcass traits of finishing pigs, but improve the meat color and decrease the nitrogen excretion. Different supplemental levels of lysine in low protein diets have no significant effects on growth performance, carcass traits and nitrogen excretion of finishing pigs, but improve the meat quality.

猪对蛋白质的需要实际是对氨基酸的需要,在必需氨基酸和氮满足供给时,猪饲粮蛋白质水平可适当降低[1]。饲喂高蛋白质水平饲粮直接导致必需氨基酸和氮通过粪、尿排出而产生浪费,使得氨基酸的氮利用率下降。饲粮蛋白质水平在NRC(1998)基础上降低2~4个百分点同时添加晶体氨基酸对猪生长性能无显著影响,且可减少氮排放,降低生产成本,促进肠道健康[2-5]。因此,低蛋白质水平饲粮是缓解我国饲用蛋白质资源短缺及降低养猪业氮排放的重要技术措施[6]。目前,低蛋白质水平饲粮在肥育猪上的研究多集中在蛋白质水平的降低程度及氨基酸种类的平衡程度上[7],而关于低蛋白质水平饲粮添加不同水平氨基酸在肥育猪上作用效果的研究则鲜见报道。赖氨酸(Lys)是猪饲粮的第一限制性氨基酸,其供给过量或缺乏都将影响猪的生长性能、氮沉积、胴体性状及营养物质的利用率[8-10]。本实验室前期研究表明,低蛋白质水平饲粮添加不同水平赖氨酸显著影响断奶仔猪的生长性能和腹泻率[11]。进一步研究发现,供给高于NRC(2012)推荐水平的赖氨酸时,断奶仔猪的氮沉积更多、营养物质消化率更高,同时可促进小肠对氨基酸的转运吸收,且利于后肠有益菌的生成及减少有害代谢物的产生,利于仔猪肠道健康[11-12]。因此,本研究以生长性能、胴体性状、肉品质及氮排放为效应指标,旨在探明低蛋白质水平饲粮添加不同水平赖氨酸在肥育猪上的饲喂效果,为低蛋白质水平饲粮的应用提供更多的技术支撑。

1 材料与方法 1.1 试验设计与饲粮

试验1:采用单因子试验设计,选取遗传背景相同、健康、初始体重为(78.19±6.69) kg的“杜×长×大”肥育猪108头,按照体重相近、性别比例一致的原则随机分为6个组,每组6个重复,每个重复3头猪。组1为正常蛋白质水平饲粮组:12.5%粗蛋白质(CP)+0.73%标准回肠可消化赖氨酸(SID-Lys)[0.73% SID-Lys为100% NRC(2012)推荐值];组2、3、4、5和6为低蛋白质水平饲粮(CP 10.5%)不同水平SID-Lys组,SID-Lys水平分别为0.58%、0.66%、0.73%、0.80%和0.88%[SID-Lys水平分别为NRC(2012)推荐值的80%、90%、100%、110%和120%]。试验2:选取体重相近[(90.56±3.28) kg]的杜×长×大肥育猪18头,随机分为3组,每组6个重复,每个重复1头猪,3组分别饲喂试验1中组1、4和5的饲粮。

试验饲粮为玉米-豆粕型饲粮,参照NRC(2012)75~100 kg阶段猪营养需要配制。通过添加L-赖氨酸盐酸盐(L-Lys·HCl)满足更高水平的赖氨酸供给,其他必需氨基酸的添加按照氨基酸模型[赖氨酸100%、含硫氨基酸(SAA)65%、蛋氨酸(Met)36%、苏氨酸(Thr)70%、色氨酸(Trp)19%、异亮氨酸(Ile)55%、缬氨酸(Val)68%、亮氨酸(Leu)100%、精氨酸(Arg)32%、组氨酸(His)32%和苯丙氨酸(Phe)60%](AMINO Pig ® 1.0, Evonik Industries, Hanau-Wolfgang,德国)平衡供给,随赖氨酸供给水平的提高而增加。所有饲粮按净能体系等能配制,各组饲粮其他营养物质(维生素、矿物质等)的供给满足或超过NRC(2012)75~100 kg阶段猪的推荐需要。试验饲粮组成及营养水平见表 1

1.2 饲养管理

本试验在四川农业大学动物营养研究所科研基地开展,试验1肥育猪按重复分圈饲养,试验2肥育猪按重复在代谢笼中单独饲养,自由采食和饮水。每天晚上结算当天每个重复的余料并估计浪费料量,记录每天采食量。整个试验过程严格按照动物营养研究所科研基地饲养管理制度和四川农业大学动物福利相关标准执行。

1.3 样品采集

试验1结束时,从每个重复中选取1头与平均体重相近的肥育猪屠宰。电击致晕,然后放血,按常规屠宰法去头、蹄,剥皮,开膛。取样品测定胴体性状和肉品质。试验2严格按照全收粪、尿的程序收集4 d排出的所有粪便、尿液,按鲜粪重量、尿液体积的10%滴加10%稀硫酸固氮,数滴甲苯防腐。收集4 d的粪样称重并混合,取样、烘干、粉碎,待测粪氮含量。收集4 d的尿样计量体积,混合均匀后取样,用于测定尿氮含量。

1.4 指标测定与方法 1.4.1 生长性能测定

于试验1开始和结束时08:00空腹个体称重,以重复为单位记录耗料量,计算平均日增重(ADG)、平均日采食量(ADFI)和料重比(F/G)。

1.4.2 胴体性状和肉品质的测定

参照瘦肉型猪胴体性状测定技术规范(NY/T 825—2004)进行胴体分割,测定胴体重,计算屠宰率。用游标卡尺测量肩部最厚处、胸腰椎结合处和腰荐椎结合处的平均背膘厚,同时在最后肋处垂直切断背最长肌,测量眼肌面积。参照猪肌肉品质测定技术规范(NY/T 821—2004),用pH计测定胸腰结合处背最长肌的pH;用色差计测定肉色;取第1~2腰椎处背最长肌,修剪成约5 cm×4 cm×3 cm的肉块,将肉样用金属钩吊起,测定24和48 h滴水损失;屠宰后72 h测定背最长肌第1~2腰椎处的剪切力。

1.4.3 粪、尿中氮含量的测定

粪、尿中的氮含量采用GB/T 6432—2018的方法测定。

1.5 数据统计分析

试验1和试验2的数据先采用Excel 2010进行整理,通过SAS 8.0软件的GLM模型对数据进行单因素方差分析,其中生长性能以每个圈为统计单位,其他指标以每头猪为统计单位,差异显著时进行Duncan氏多重比较,同时对试验1中组1和组4的数据进行t检验,以P < 0.05为差异显著。

2 结果 2.1 低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪生长性能的影响

表 2可见,降低饲粮蛋白质水平对肥育猪的ADFI、ADG、F/G均无显著影响(P>0.05)。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪的ADFI、ADG、F/G均无显著影响(P>0.05)。

2.2 低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪胴体性状和肉品质的影响

表 3表 4可见,降低饲粮蛋白质水平对肥育猪胴体性状无显著影响(P>0.05),但显著影响肥育猪背最长肌的红度值(P < 0.05)。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪胴体性状无显著影响(P>0.05),但显著影响肥育猪的24和48 h滴水损失(P < 0.05),且以组5的滴水损失最低。

2.3 低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪氮排放的影响

表 5可见,降低饲粮蛋白质水平显著影响了肥育猪的粪氮排放、尿氮排放和总氮排放(P < 0.05),饲粮蛋白质水平每降低1%,肥育猪总氮排放可减少13.7%。低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪粪氮排放、尿氮排放和总氮排放均无显著影响(P>0.05)。

3 讨论

近年来许多报道指出,降低饲粮蛋白质水平同时按照理想氨基酸模型补充合成氨基酸对肥育猪生长性能无显著影响[13-15]。本研究表明,饲粮蛋白质水平降低2个百分点不影响肥育猪的ADFI、ADG和F/G,这与上述报道相一致;在低蛋白质水平饲粮下,供给不同水平赖氨酸对75~100 kg肥育猪生长性能无显著影响,在相似体重阶段朱立鑫[16]也得到了相似的结果。这表明75~100 kg肥育猪饲喂低蛋白质水平饲粮时对赖氨酸的需要量在NRC(2012)基础上可能无需进一步提高。然而朱立鑫[16]在50~80 kg肥育猪上的研究则发现,低蛋白质水平饲粮下随着赖氨酸供给水平的提高,肥育猪生长性能呈提高趋势;Xie等[17]的研究也得到了相似的结果。其可能的原因是猪的生长阶段不同,瘦肉沉积对赖氨酸的需求不同,即饲喂低蛋白质水平饲粮时肥育猪在育肥前期对赖氨酸的需要量可能高于NRC(2012)推荐值。因此,关于低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪生长性能的影响还需开展更多的研究。

前人报道指出,给肥育猪饲喂低蛋白质水平饲粮时,其屠宰率降低,背膘厚显著升高[18-19]。其可能原因是饲粮能量水平有差异。配制低蛋白质水平饲粮,减少豆粕用量的同时提高了玉米等能量饲料的比例,按照消化能或代谢能体系,低蛋白质水平饲粮的消化能和代谢能与正常蛋白质水平饲粮相近,但净能却高于正常蛋白质水平饲粮。而本试验得到,当采用净能体系配制饲粮时,低蛋白质水平饲粮组肥育猪的胴体性状与正常蛋白质水平饲粮组相比无显著差异。Xue等[20]的试验结果也发现,相同净能水平下,低蛋白质水平饲粮组和高蛋白质水平饲粮组相比,肥育猪的第10肋背膘厚和眼肌面积差异均不显著。同时本研究发现,降低饲粮蛋白质水平显著增加了肥育猪背最长肌的红度值,这一结果与前人的报道[21-22]相同。此外,低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪胴体性状无显著影响,表明胴体脂肪的沉积可能主要与能量水平有关,与赖氨酸供给水平无显著相关。然而,低蛋白质水平饲粮添加不同水平赖氨酸会显著影响肥育猪的滴水损失,且0.80% SID-Lys时滴水损失最低,表明饲喂低蛋白质水平饲粮时适宜提高赖氨酸的供给水平有利于降低肥育猪的滴水损失。其可能原因是饲喂含0.80% SID-Lys饲粮时可促进肥育猪肌肉蛋白质沉积,提高了对水分子的吸附力,进而增加了肌肉系水力,改善了肉品质。

猪通过饲粮摄入蛋白质的数量与其本身的需要量越吻合,氮的利用率越高,氮排放量越低[23]。许多研究结果表明,降低猪饲粮蛋白质水平能够显著降低氮的排放量,饲粮蛋白质水平每降低1%,总氮排放可降低8%~10%[24-25]。本研究结果表明,降低饲粮蛋白质水平可显著降低肥育猪的氮排放,饲粮蛋白质水平每降低1%,肥育猪总氮排放降低了13.7%。此外,饲喂低蛋白质水平饲粮时,提高赖氨酸的供给水平对肥育猪的氮排放无显著影响,表明添加高于NRC(2012)推荐水平的赖氨酸不能提高肥育猪的氮利用率。

4 结论

① 饲粮蛋白质水平在12.5%基础上降低2%,同时平衡10种必需氨基酸,对75~100 kg肥育猪的生长性能和胴体性状无显著影响,但可改善肉色,同时降低总氮排放。

② 低蛋白质水平饲粮添加不同水平赖氨酸对肥育猪的生长性能和胴体性状无显著影响,但显著影响滴水损失,0.80% SID-Lys时肥育猪的滴水损失最低,改善了肉品质。
