摄食对动物的生长和生产至关重要,摄食量过低不能满足动物生长的需要而降低养殖效应,摄食量过高则可能使饲料转化效率低而增加养殖成本。因此,在养殖过程中控制动物摄食十分重要。大量的研究显示,动物摄食行为受许多因素的影响,其中包括许多由中枢神经系统和外周组织分泌的激素和小肽,如神经肽Y(NPY)、阿片黑素促皮质素原(POMC)、刺鼠基因相关蛋白(AgRP)、可卡因安非他明调节转录因子(CART)、胃饥饿素(Ghrelin)、肽YY(PYY)、胰高血糖素样肽-1(GLP-1)和瘦素(Leptin)等。其中,神经介素U(neuromedin U,NMU)与神经介素B、C、K、L、N、S组成神经介素家族,其是一种重要的厌食欲因子,广泛分布于动物的中枢神经系统和外周组织中。NMU主要有25肽(NMU-25)和8肽(NMU-8)2种形式。NMU主要通过其受体神经介素受体1(neuromedin U receptor 1,NMUR1)和神经介素受体2(neuromedin U receptor 2,NMUR2)发挥多种生物学功能,如调控摄食[1-2]、能量稳态[3-4]、睡眠[5]、肠道炎症[6]、成骨细胞分化[7]、肿瘤[8]、哮喘[9]和热应激[10]等。本文试图归纳总结NMU的发现、结构、组织分布及其调控摄食的作用及机制,以期为肥胖、厌食症的治疗和动物摄食调控提供部分参考。
1 NMU的发现与结构在1985年日本学者Minamino等[11]从猪脊髓中分离出一种新型肽,因其能刺激子宫平滑肌收缩,故将其命名为NMU。此后,学者们相继在人[12]、大鼠[13]、小鼠[14]、猪[15]和兔[16]等哺乳动物,以及鸡[17]、蟾蜍[18]和鱼类[19-22]等非哺乳动物中克隆获得NMU基因。有趣的是,尽管哺乳动物NMU基因只有1种,但是对部分鱼类的研究显示NMU基因存在多种亚型[19-20, 22]。这种现象可能与鱼类经过了多重基因组复制事件相关。此外,不同亚型NMU基因发挥的功能是否存在差异有待研究。
各物种NMU基因编码的氨基酸一致性较高,尤其是靠近C末端的结构域,可裂解为多种形式的NMU多肽。除了NMU-25和NMU-8这2种普遍存在于哺乳动物的NMU[19, 23],还从其他物种上鉴定出其他形式的NMU多肽,如大鼠(NMU-23)[13]、豚鼠和鸡(NMU-9)[17, 24]、蟾蜍(NMU-17)[18]、金鱼(NMU-21、NMU-38)[19](图 1)。各物种NMU靠近C端的4肽(Arg-Pro-Arg-Asn-NH2)氨基酸序列均一致,而哺乳动物NMU靠近C端的7肽(Phe-Leu-Phe-Arg-Pro-Arg-Asn-NH2)完全相同,C端天冬酰胺(Asn)的酰胺化使其具有活性。体外研究证实NMU小肽的活性可能存在差异,如大鼠NMU-23的活性比猪NMU-25活性高2倍、比蟾蜍NMU-17活性高3倍。因此,对不同物种NMU基因编码的氨基酸序列和裂解位点进行分析,选择适宜形式的NMU小肽开展功能研究显得十分必要。
越来越多的研究证实,NMU广泛分布于中枢神经系统和外周组织中。学者们通过放射免疫分析法、免疫组化法和实时荧光定量等方法检测NMU的免疫活性或mRNA表达,已经发现NMU主要在外周组织尤其是胃肠道中高表达,在中枢神经系统如垂体前叶、纹状体、下丘脑、延髓、扣带回和额叶内侧回也有分布[25-29]。与中枢神经系统和外周组织中NMU的含量相比,血液中NMU的含量非常低。以上研究提示NMU可能主要以自分泌或旁分泌的方式在局部组织中发挥作用,而不是作为一种循环激素。
此外,各物种NMU的组织表达模式存在差异。值得注意的是,部分鱼类NMU存在亚型,而这些亚型的组织表达模式存在差异。例如,有学者通过实时荧光PCR(RT-PCR)测定NMU mRNA的表达情况,发现金鱼NMU-1和NMU-2 mRNA在肠道高表达,NMU-3仅在脑中表达,NMU-4在脑中高表达[19]。红鳍东方鲀NMU-1、NMU-2和NMU-3 mRNA在肠道、鳃、头肾、肝脏和皮肤中均有表达,NMU-2 mRNA在肌肉和脾脏中也有表达,NMU-3 mRNA还在脾脏中表达[22]。
总的来说,NMU广泛分布于动物中枢神经系统和外周组织中,提示NMU可能具有多种生物学功能。
3 NMU调控动物摄食关于NMU对动物摄食的影响,主要集中于啮齿动物和鱼类上,在鸟类和其他动物上鲜有报道。在啮齿动物、鸟类和鱼类上,许多研究已证实了NMU具有降低动物摄食量的作用。在饲粮诱导的肥胖雄性小鼠上,单次皮下注射人类NMU-25(1.0、3.0、10 mg/kg)显著降低2和18 h摄食量[30]。皮下注射0.3 μmol/kg的NMU类似物(GUB07-007)也能显著降低6 h摄食量[31]。与天然NMU-25相比,人工修饰后的NMU-25类似物抑制食欲的效应更强且持久。在大鼠上,腹腔注射NMU-23能抑制摄食,天然NMU-8对采食量无显著影响[32]。在蛋鸡和肉鸡上,脑室注射大鼠NMU-23能显著降低30 min、1 h和2 h的摄食量[17, 33]。在金鱼上,除了之前发现脑室注射NMU-21能抑制摄食外,最近,有学者还发现脑室注射人工合成的金鱼NMU-9和NMU-25(100 pmol/g)也能显著降低1 h摄食量,NMU-25和NMU-21对摄食量的抑制作用类似,而NMU-9的抑制作用相对较弱[2]。由上可知,NMU多肽的厌食欲作用可能存在种属特异,不经修饰的NMU-25小肽的厌食欲作用已被普遍证实,而经人工修饰的NMU类似物或NMU小肽的厌食欲作用似乎更强。
4 NMU调控动物摄食的机制越来越多的研究显示,NMU可能主要通过作用于中枢神经系统调控动物摄食。下丘脑是调控动物摄食的关键区域,能整合从外周和其他脑区传递的机体能量和代谢状态信息。在大鼠上,持续3 d脑室注射NMU于下丘脑,24 h的累积摄食量均显著降低[34]。此外,脑室注射NMU于下丘脑室旁核(PVN)和背侧缝核(DRN)也能抑制摄食,Fos样免疫反应结果显示室旁核和背侧缝核是NMU的作用位点[35]。最近,学者发现脑室注射NMU(125 ng)于前腹侧被盖区(aVTA)时小鼠4 h采食量显著降低,注射于外侧被盖核(LDTg)对摄食量有抑制的趋势,而注射于伏隔核(NAc)对采食量无显著影响[1]。由上可知,在啮齿动物上,不同大脑区域中的NMU信号能选择性地调节摄食行为,室旁核、背侧缝核、前腹侧被盖区和外侧被盖核可能是NMU发挥厌食作用的关键作用位点。此外,尽管有研究发现持续注射NMU能抑制胃肠排空速度,但是该研究是通过检测血浆中乙酰氨基酚含量的变化来间接反映胃排空速度的[36],尚不清楚NMU是否通过作用于外周组织(如胃肠道)调控摄食。
在中枢神经系统中,NMU抑制摄食的作用可能主要受NMUR2的介导。NMUR2作为NMU的高亲和力受体,主要分布于中枢神经系统,尤其是下丘脑室旁核。在大鼠上,有学者通过腺病毒介导的RNAi选择性地敲降室旁核中的NMUR2基因,虽然对饲喂普通饲粮大鼠的摄食量没有显著影响,但是能显著提高饲喂高脂饲粮大鼠的摄食量[37]。类似地,注射NMUR2激动剂(NMU-7005)能抑制肥胖小鼠的摄食,刺激小鼠下丘脑和延髓中c-Fos基因表达[38]。NMUR2介导NMU的厌食欲作用可能与动物肥胖程度相关。最近,有学者发现当敲降室旁核NMUR2时,长期剥夺高脂饮食的大鼠对高脂食物的认知反应和渴望程度被明显抑制[39]。NMUR2对高脂食物摄入的调节作用较复杂,不仅能调节偏好高脂食物的神经回路,还影响渴望摄食的相关回路。前者主要受中脑边缘多巴胺系统调节,而后者可能受前额叶皮层的调节。此外,在小鼠上,有学者发现当敲除NMUR1时,皮下注射NMU-25或NMU-23的抑制食欲作用被阻断[30]。外周NMU抑制摄食可能受在外周组织中丰富表达的NMUR1介导。由上可知,NMUR1和NMUR2均可能参与NMU的摄食调控作用,NMUR1可能主要参与外周NMU调控的非致肥的摄食行为,而室旁核中NMUR2信号可能与高脂食物的偏好、认知和渴望相关。尽管目前已鉴定NMUR1和NMUR2分别参与NMU中枢神经系统和外周组织中的调控摄食作用,但是NMU是否还可能通过其他受体来调控摄食尚不清楚。
进一步的研究显示,NMU能与下丘脑核团和外周分泌的部分食欲因子相互作用调控摄食,如NPY、POMC和促肾上腺皮质激素释放激素(CRH)。在下丘脑弓状核(ARC)中,增食欲因子NPY和抑制食欲因子POMC扮演着重要的角色。在DIO小鼠上,皮下注射NMU-25(10 mg/kg)显著降低下丘脑NPY mRNA表达[30]。在点带石斑鱼中,腹腔注射NMU-21能显著降低下丘脑NPY mRNA表达[21]。Nakahara等[40]研究显示脑室注射NMU可刺激下丘脑弓状核POMC mRNA的表达,体外研究结果显示NMU能刺激下丘脑弓状核α-黑色素细胞刺激素(α-MSH)的分泌。由上可知,在下丘脑弓状核中,NMU可能通过抑制NPY表达,促进POMC表达,从而抑制动物摄食。
NMU的厌食欲作用也与厌食欲因子CRH相关,CRH主要在下丘脑室旁核产生。室旁核能进一步整合和调节来自中枢神经系统各个区域和胃肠神经系统的神经信号。在小鼠上,当敲除CRH基因时,NMU不能抑制食物摄入[41]。Nakahara等[40]研究显示脑室注射NMU可刺激室旁核中CRH mRNA的表达。在金鱼中,脑室注射NMU-21能显著上调CRH mRNA的表达,并且脑室注射CRH受体拮抗剂(α-helical CRH9-41)能阻断NMU-21的厌食欲效应[42]。然而,在点带石斑鱼上,腹腔注射NMU-21不影响下丘脑CRH基因的表达[21]。以上研究显示,在中枢神经系统中,NMU也能通过作用于下丘脑室旁核,与CRH及其受体相互作用来调控动物摄食。但是,外周NMU抑制摄食的作用可能与CRH信号无关。
此外,NMU厌食欲作用也可能与Leptin信号有关。Leptin是一种主要由脂肪细胞分泌的蛋白质,具有抑制动物摄食的作用。在大鼠上,脑室注射NMU抗体能部分削弱Leptin抑制摄食的作用[43]。类似的,在小鼠上,腹腔注射NMU抑制摄食的作用不受Leptin缺失的影响[30]。由上可知,NMU厌食欲作用可能与Leptin有关,且NMU可能作为Leptin的下游因子发挥厌食欲作用。此外,学者也发现部分因子与NMU的厌食欲作用不相关,如GLP-1[30]、PYY[30]、黑素皮质素受体4(MC4R)[42]和神经降压素受体1(NTSR1)[44]。
综上,NMU抑制动物摄食的作用十分保守,尤其是通过NMUR2在中枢神经系统中发挥厌食欲作用。在中枢神经系统中,NMU能通过NMUR2作用于NPY、POMC和CRH等食欲信号调节动物摄食。在外周组织中,NMU调控摄食的机制尚不清楚,有待研究。NMU调控摄食机制如图 2所示。
NMU在物种间十分保守,其广泛分布于动物的中枢神经系统和外周组织中,提示NMU功能的多样性。目前,国内外关于NMU调控摄食的研究主要集中于人类、啮齿动物和鸡上,在鱼类上仅见鲤形目的金鱼和鲈形目的点带石斑鱼,许多调控机制尚未阐明。因此,继续深入开展调控机制研究十分必要。此外,NMU类似物的开发有利于治疗全球流行的肥胖等疾病;筛选并鉴定天然NMU受体拮抗剂将有利于提高动物摄食量,促进动物生长,提高养殖经济效益。
