胆固醇在甲壳动物中的研究进展

摘要: 胆固醇是动物体内一种重要的固醇类物质, 是动物细胞膜的重要组成成分, 还是类固醇激素形成的前体物质, 协调参与机体内糖类、脂肪和蛋白质等营养物质的代谢活动。甲壳动物不能由乙酸从头合成胆固醇, 需要依靠饮食补充机体对于胆固醇的需求。本文从胆固醇对于甲壳动物的生理作用以及胆固醇在甲壳动物中的吸收转运机制为切入点, 综述了甲壳动物对胆固醇的需求量, 并探讨了不同饲料原料及饲料营养组成对于甲壳动物胆固醇需求量的影响, 以及饲料胆固醇含量对甲壳动物生长、体成分、健康和繁殖的影响, 以期为深入开展胆固醇在甲壳动物中的作用机理研究及开发甲壳动物饲料胆固醇添加剂提供科学依据。

Abstract: Cholesterol is an important steroid substance in the animal body, an important component of animal cell membranes, and a precursor substance for the formation of steroid hormones. It coordinates and participates in the metabolism of nutrients such as sugar, fat and protein in the body. Crustaceans cannot synthesize cholesterol from acetic acid and need to rely on diet to supplement the body's demand for cholesterol. Starting from the physiological effects of cholesterol on crustaceans and the absorption and transport mechanism of cholesterol in crustaceans, this paper summarized the requirements of crustaceans for cholesterol, and discussed the effects of different feed ingredients and dietary nutritional composition on crustacean cholesterol requirements, and the effects of dietary cholesterol content on the growth, body composition, health and reproduction of crustaceans, in order to provide scientific basis for in-depth research on the mechanism of cholesterol in crustaceans and the development of crustacean feed cholesterol additives.

脂类是动物六大营养素之一,胆固醇作为参与细胞膜形成的一种类脂[1],与动物细胞的生命活动息息相关。胆固醇除了参与细胞膜的形成以及影响细胞膜对脂类物质的通透性外,还参与了类固醇激素、胆汁酸盐以及维生素D3的合成,协调机体内包括糖、蛋白质和脂肪等物质的代谢,并间接促进神经系统的正常运转。人类以及绝大多数的动物都具有从头合成或者利用其他甾醇合成胆固醇的能力,并利用合成的胆固醇保证生命活动的进行。但体内胆固醇含量过高会对机体造成损伤,例如,人体血清中胆固醇含量过高易引起动脉粥硬化,造成血管堵塞,对人体健康产生威胁[2]

随着水产养殖规模的不断扩增,对甲壳动物的研究日益增多,人们逐渐发现甲壳动物并不能从乙酸从头合成胆固醇,甚至大部分的甲壳动物也不能利用其他甾醇合成胆固醇,只能依靠外源胆固醇的摄入来维持生命活动[3-6]。有关胆固醇对甲壳动物影响的研究主要集中于胆固醇在饲料中的适宜添加量。研究结果显示,不同物种对饲料中胆固醇的需求量并不一致,且同一物种的不同发育时期需求量也不一致[7-8]。本文主要对甲壳动物对胆固醇的需求量、胆固醇的来源与消化吸收机制以及饲料中添加胆固醇对甲壳动物体成分、健康以及蜕壳与繁殖的影响进行综述,以期为深入开展胆固醇在甲壳动物中的作用机理研究及开发胆固醇饲料添加剂提供科学依据。

1 胆固醇的理化性质

胆固醇,又名胆甾醇,是一种含有环戊烷多氢菲骨架的类固醇及甾醇,化学式为C27H46O,在固态时是白色、类白色结晶或结晶性粉末,放置在空气中会被氧化变黄。胆固醇几乎不溶于水,易溶于有机溶剂,在工业生产中通常采取对动物大脑使用丙酮浸提和对羊毛脂皂化和抽提来提取胆固醇[9]。胆固醇广泛存在于动物体的细胞膜中,是构成细胞膜的重要组分,同时也是几种重要的激素和胆汁酸合成的前体物质[10-12]。在血清中,胆固醇以2种不同的形式存在,即游离胆固醇(free cholesterol,FC)和与脂肪酸结合的胆固醇酯(cholesterol ester,CE)[13-14],二者合称为总胆固醇(total cholesterol,TC)。根据运输胆固醇的脂蛋白(lipoprotein,LPs)不同,胆固醇在机体中的运输形式亦可分为2类,即低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)与高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)。

2 甲壳动物胆固醇的来源、吸收与转运 2.1 甲壳动物胆固醇的来源

胆固醇在机体内合成的主要场所是肝脏,其生物合成可以分为5个阶段:1)由乙酸合成甲羟戊酸;2)由甲羟戊酸形成异戊酰焦磷酸和二甲烯丙基焦磷酸;3)角鲨烯的合成;4)角鲨烯环合形成羊毛固醇;5)由羊毛固醇转化为胆固醇。大多数动物都可以在体内合成胆固醇。虾蟹类等甲壳动物可能因为其体内法尼焦磷酸到鲨烯这一中间反应受到阻滞[15],故不能由乙酸从头合成胆固醇[11, 16-19]

Zandee[4]对欧洲龙虾(Homarus gammarus)施用乙酸盐—1-14C后,在其体内并没有发现胆固醇的合成。对日本对虾(Penaeus japonicus)和三疣梭子蟹(Portunus trituberculatus)施用乙酸盐—1-14C后亦未发现这2种甲壳动物将乙酸盐—1-14C掺入到胆固醇中[5]。甲羟戊酸需由乙酸合成,直接将甲羟戊酸-2-14C注入到黄道蟹(Cancer pagurus)中[20]也得到了相同的结果。有研究者推断,整个甲壳纲动物都需要从食物中获得胆固醇[3]。故对于甲壳动物饲料原料的选择,通常会利用鱼油、牛脂以及猪油等胆固醇含量较高的原料,提高饲料总体的胆固醇含量。在某些甲壳动物中,例如斑节对虾(Penaeus monodon),当饲料原料含有足够高的胆固醇时,可以不用在饲料中额外添加胆固醇[17]

近年来,由于经济以及环境因素,饲料原料中鱼粉以及鱼油的替代成为一个亟需解决的问题[21]。不少研究者将目光转向了植物性蛋白质,例如大豆蛋白和毕氏海蓬子的种子等[22]。但植物性蛋白质通常不含胆固醇且常常含有抗营养成分,如单宁、蛋白酶抑制剂、生物碱等[23-25],以高植物性蛋白质源饲料进行饲养时,饲料中不仅会缺乏胆固醇以及牛磺酸等微量成分,其中的植物性蛋白质源还可能会降低胆固醇利用率[1, 26],出现血液中胆固醇含量降低的情况,而在以植物性蛋白质为主的饲料中添加适量的胆固醇可以有效地预防此现象的发生[27-29]。故在以豆粕代替鱼粉作为蛋白质源或者高豆粕低鱼粉为代表的高植物性蛋白质源饲料中,胆固醇的最适添加量会比以鱼粉为蛋白质源的饲料高,如在骆源等[30]的研究中,豆粕代替鱼粉的饲料中胆固醇添加量为0.6%时,凡纳滨对虾(Litopenaeus vannamei)幼虾(0.35 g)的生长性能和饲料效率较高,而在Zhang等[8]的研究中凡纳滨对虾幼虾(0.61 g)胆固醇的需求量只有0.16%,凡纳滨对虾亚成体(4.25 g)甚至不需要额外在饲料中补充胆固醇。

2.2 甲壳动物胆固醇的吸收与转运

胃肠道是脊椎动物胆固醇消化吸收的主要场所,其中的胆固醇也是脊椎动物除自身合成外体内胆固醇的主要来源。胃肠道中的胆固醇是由食物中的胆固醇、胆汁胆固醇和脱落细胞中的胆固醇组成[31],胆汁胆固醇可以直接由小肠进行吸收。食物中的甘油三酯(triglyceride,TG)和磷脂经胰酶、胃蛋白酶等消化酶消化后,释放出胆固醇,酯化的胆固醇无法穿过小肠上皮细胞进入细胞内,需由胆汁酸乳化成胆固醇。胆固醇进入小肠细胞内后在内质网上催化形成CE,并同TG、磷脂和少部分FC一起组装成乳糜微粒(chylomicron,CM),经基底膜分泌进入淋巴循环[32-33]

在机体吸收足够的胆固醇后,胆固醇可在肝细胞中转化为胆汁酸排出体外或者转化为CE储存起来,这是哺乳动物或者鱼类等脊椎动物体内胆固醇代谢的主要去路。而虾蟹类甲壳动物,不仅没有合成胆固醇的能力,而且由于没有胆囊,无法分泌胆汁,更不能将体内多余的胆固醇转化为胆汁酸[13]。对黄道蟹进行解剖,在其胃液中没有分离到胆汁酸,表明其确实无法通过合成胆汁酸降解胆固醇[20]。在甲壳动物中,通过食物获得的外源胆固醇通常是被其他乳化剂乳化溶解后在小肠中吸收。甲壳动物肝胰腺(中肠腺)合成并分泌到肠道的N-牛磺酸(N-十二烷基肌醇)能够有效地溶解胆固醇,从而起到促进胆固醇在小肠内吸收的作用[34-35]。除此之外,卵磷脂也被报道能够促进胆固醇在甲壳动物肠道中的溶解和运输[34],并且在饲料中添加卵磷脂时,所需要添加的胆固醇的量也会降低,可能是卵磷脂可以催化卵磷脂胆固醇脂酰转移酶(lecithin-cholesterol acyltransferase,LCAT)的激活,使胆固醇以LPs的形式进行体内运输,提高胆固醇的利用率[36]

胆固醇是甲壳动物的基本营养物质,在甲壳动物中,胆固醇主要由小肠吸收之后转运到全身各处参与各项生命活动的调节。除极端情况下,胆固醇不太可能被用于能量供给[14]。在饥饿等极端情况下,首先被调用的是富含能量的脂肪。随着脂库的消耗,蛋白质的分解代谢增加。饥饿最后阶段的能量来源于对结构组织(如细胞膜)中脂质的消耗,是机体濒临死亡的阶段[14, 37]。脂质不溶于水,需要特殊的机制才能在动物血浆中有效运输[38]。在脊椎动物和甲壳动物中,脂质(包含CE)被包装成LPs,然后运输到整个身体。甲壳动物将脂质从肠道装载到LPs,然后在血淋巴中循环,直到达到细胞膜上的特定受体[39]。在斑节对虾中,各组织中均含有CE,肝胰腺中CE的含量最高[40],说明胆固醇在体内主要以CE的形式储存,且肝胰腺是脂类营养储存的重要组织。

Guary等[41]给处于不同蜕壳时期的日本对虾注射14C胆固醇,分析各组织中的放射性强度,放射性物质主要集中在性腺和真皮组织。Kanazawa等[42]给日本对虾注射14C胆固醇后,在被注射对虾的肝胰腺、卵巢及血淋巴中分离得到含放射性物质的类固醇激素。这说明胆固醇与甲壳动物的蜕壳和生殖相关,且胆固醇在被吸收后主要被转运到皮下组织、性腺和肝胰腺中。

3 甲壳动物对胆固醇需求量的研究进展

胆固醇是动物体维持生命活动的重要活性物质之一,在生理功能上,它是构成细胞膜的重要组分,有助于细胞膜的流动性和刚性,以抵御环境突变[43-44]。除此之外,甲壳动物对胆固醇的需求更多体现在其作为激素合成的前体物质上。在如克氏原螯虾(Procambarus clarkii)及红螯螯虾(Cherax quadricarinatus)等甲壳动物中,其生长发育总是伴随着蜕壳活动[45]。自然状态下,在生存环境达到蜕壳条件时,甲壳动物会产生相应的蜕壳激素诱导蜕壳活动的发生,如20-羟基蜕皮酮(β-蜕皮酮)[46]。在缺乏胆固醇的生存条件下,甲壳动物会因不能合成蜕壳激素导致无法蜕壳,出现蜕壳死亡现象[47]。甲壳动物中的胆固醇主要通过外源性食物获取,食物中的胆固醇由小肠进行消化吸收后被包装成CM经血淋巴运输到全身各处,参与各项生命活动。在饲料中添加有利于胆固醇进行转运的物质,似乎能够起到提高转运效率,增强机体对胆固醇的利用率,使机体对胆固醇的需求量减少[12]

3.1 饲料胆固醇对甲壳动物生存和生长的影响

从20世纪70年代开始,研究者在发现甲壳动物不能合成胆固醇后,便对甲壳动物的胆固醇需求进行探索,主要依据等氮饲料中胆固醇的不同添加量对甲壳动物的存活率、增重率以及蜕壳次数来确定饲料中胆固醇的最适添加量。饲料中添加胆固醇的日本对虾与饲料中不添加胆固醇的日本对虾相比,生存率会更高[48]。在对凡纳滨对虾的研究中发现,不含鱼粉的植物性蛋白质源饲料中的胆固醇并不足以促进幼年凡纳滨对虾的生长[49]

在不同的研究中,基础饲料的组成并不相同,得出的胆固醇最适添加量也不尽相同。有关日本对虾和凡纳滨对虾的胆固醇需求量的研究相对较多,在饲料中使用6%的脂肪时,确定日本对虾幼虾的胆固醇需求量为2.1%[50],而在饲料中使用8%的脂肪时,得出胆固醇的需求量分别为0.5%与0.12%[48, 51],显著低于低脂肪水平饲料下的需求量。Morris等[49]以豆粕为主要蛋白质源的饲料饲喂凡纳滨对虾时,饲喂添加0.06%胆固醇的饲料(饲料TC水平为0.125%),对虾的生长性能有显著改善,与Gong等[7]利用植物性蛋白质源饲料饲喂幼年凡纳滨对虾得到的胆固醇需求量(0.14%)基本一致。以酪蛋白(33.7%)和鱼粉(4%)为蛋白质源的养殖试验中,饲料中胆固醇含量在0.066%以上的对虾生物量、平均体重和增重率均显著高于饲喂基础饲料(胆固醇含量为0.045%)的对虾,且投喂基础饲料的对虾存活率最低(68%)[52],说明低鱼粉饲料中的胆固醇亦不能满足凡纳滨对虾的生存需求。

除脂肪含量与蛋白质源等基础饲料组成的差别之外,当饲喂水平不同时,饲料胆固醇的最适添加水平也会不同。在饲喂水平为体重的3%、5%和7%时,日本对虾饲料中胆固醇的适宜添加水平分别为0.5%~0.6%、0.36%~0.40%和0.26%~0.29%[53],若根据经济成本进行选择,在饲料中添加0.4%的胆固醇以5%的饲喂水平进行饲喂最为合适。

另外,一些甲壳动物对胆固醇似乎没有特殊的需求。饲喂以脱脂鱿鱼粉为主要蛋白质源的饲料对锯缘青蟹(Scylla serrata)胆固醇与卵磷脂需求量影响的研究中,发现锯缘青蟹对于胆固醇与卵磷脂的需求量具有交互作用,且当卵磷脂的添加量足够多(39.7~44.1 g/kg)时,饲料中胆固醇添加量的多少对其存活率的影响并不显著[12]。也就是说当饲料中补充足够多的卵磷脂时,锯缘青蟹对于饲料中胆固醇的需求量可以降低,从而降低经济成本。这种交互作用被认为是卵磷脂促进了胆固醇的溶解与转运,促进胆固醇在甲壳动物机体内的利用[36]。Smith等[17]曾报道大部分补充卵磷脂的商业虾饲料中,胆固醇的含量是高于亚成年斑节对虾的最佳胆固醇需求量(1.7 g/kg),所以他们认为不需要在商业虾饲料中补充胆固醇。Thongrod等[54]曾报道,在初重0.11 g的墨吉对虾(Penaeus merguiensis)幼虾饲料中添加0.5%和1.0%胆固醇时与对照组(不添加胆固醇)的增重率无显著差异,这有可能是该研究中的饲料已经提供了足够水平的胆固醇。

本文对一些甲壳动物的胆固醇需求量做了总结,发现大部分的甲壳动物对于胆固醇的需求量为饲料水平的0.5%,详见表 1

在这些不同的研究中,基础饲料的组成各不相同,直接从非胆固醇成分引入的胆固醇水平也可能不同。所有这些研究都表明,不同的物种和发育阶段以及饮食状况不同,对胆固醇的需求量不尽相同。大部分的甲壳动物对饲料中胆固醇的需求量一般为0.2%~1.0%[54],摄食含有较高水平的膳食类固醇的饲料,会抑制美洲龙虾(Homarus americanus)、日本对虾和锯缘青蟹等甲壳动物的生长[6, 11, 48, 72]。若在饲料配方的设计上充分考虑到不同的影响因素,并更精确地定义不同甲壳动物对胆固醇的需求量,则可以在确保甲壳动物最适的生长性能下减少胆固醇的补充量,节约经济成本。

3.2 饲料胆固醇对甲壳动物体成分的影响

甲壳动物虽然不能由乙酸从头合成胆固醇,但其体内胆固醇含量较高,且在肝胰腺中含量最高[13]。研究发现,在饲料中添加一定量的胆固醇可以提高甲壳动物体内胆固醇的含量[72]。在凡纳滨对虾中,随着饲料中胆固醇含量的升高,肝胰腺中胆固醇、脂肪以及TG含量都显著提高[7],Guo等[52]也曾报道,对虾全虾胆固醇和胆固醇沉降率与饲料胆固醇含量呈剂量-反应关系。在日本对虾中还发现饲料中添加胆固醇能增加对虾体内脂肪的沉积,提高肌肉中总脂肪的含量,投喂无胆固醇饲料的日本对虾组织胆固醇含量相对低于投喂含胆固醇饲料的日本对虾幼虾[48, 74],在美洲龙虾中也得到了相似结果[6]。这可能与胆固醇在机体内的吸收转运机制相关,小肠细胞内的CE与TG以及磷脂和少量胆固醇形成CM进入淋巴循环,胆固醇含量的增加,使机体内的脂肪与TG含量都得到提高。

在饲料中添加0.5%的胆固醇除了使幼年印度对虾(Penaeus indicus)生长率以及胆固醇含量显著提高外,其蛋白质保留率也显著提高,体内蛋白质沉积增加[64]。骆源等[30]也报道称,在以豆粕替代鱼粉的饲料中,随着胆固醇添加水平的升高,全虾粗蛋白质和粗脂肪含量呈上升趋势。在其他研究中还发现饲料中补充胆固醇能促进日本对虾中虾青素的沉积[56]

有意思的是,被报道不需要在饲料中额外补充胆固醇的甲壳动物,胆固醇含量的改变就如对于其生存率和生长率无显著影响一般,对其总脂的含量也没有显著影响,但游离脂肪酸含量升高[54],可能是对虾体内存在某种在摄入高胆固醇时将酯化固醇游离化的机制,并将游离的固醇排出机体,维持机体的总脂平衡,具体的机制值得进一步的探索。

与胆固醇添加水平过高抑制生长相似,在斑节对虾中,当胆固醇的添加水平过高时,组织中胆固醇含量会降低[11]。Castell等[6]也曾报道,摄取2%胆固醇的美洲龙虾比摄取0.5%和1.0%胆固醇的美洲龙虾保留的胆固醇更少。这表明,过量的甾醇在美洲龙虾体内诱导了一种胆固醇清除机制,导致这种甾醇的体内含量低于最佳饲料胆固醇含量的龙虾。胆固醇含量过高,除了可能降低体内沉积胆固醇含量,粗脂肪含量也可能降低。在凡纳滨对虾幼虾中,随着饲料胆固醇含量的升高,其粗脂肪含量呈下降趋势。在凡纳滨对虾亚成体中,随着饲料胆固醇含量的升高,粗脂肪含量也呈先升后降的趋势[8]

饲料中添加胆固醇对甲壳动物体成分影响的研究主要集中在对胆固醇沉积以及粗脂肪、粗蛋白质含量的影响,研究表明,在适宜的饲料胆固醇含量下,胆固醇可以促进这些成分在甲壳动物体内的沉积,但具体的影响机制还有待进一步研究阐明。

3.3 饲料胆固醇对甲壳动物健康的影响

甲壳动物特异性免疫能力较弱,一般认为甲壳动物依靠非特异性免疫抵御外界侵染[75]。通常可通过对先天免疫相关的酶活性或免疫通路中基因表达的水平来评价机体免疫和健康状况[76-77]

Gu等[78]研究发现,饲料中添加胆固醇能上调日本沼虾总抗氧化能力,以及谷胱甘肽转移酶(glutathione transferase,GST)、过氧化氢酶(catalase,CAT)、碱性磷酸酶(alkaline phosphatase,AKP)和酸性磷酸酶(acid phosphatase,ACP)等非特异性免疫相关酶的活性,提高日本沼虾非特异性免疫能力,这与在三疣梭子蟹幼蟹中得到的结果[79]相似。在一定的添加范围内,三疣梭子蟹血清中的溶菌酶活性也随饲料中胆固醇添加水平的增加而增强[80]。闫明磊等[81]曾报道,以添加胆固醇的饲料饲养凡纳滨对虾,并在8周后注射副溶血弧菌悬液,对虾免疫相关基因表达量上升,表明其具有良好的抗细菌感染能力。

除了免疫指标的变化外,适量的胆固醇可以使甲壳动物抵抗不利环境的能力增强。Paibulkichakul等[61]发现,饲喂含有1%胆固醇饲料的斑节对虾,应激能力增加,在面对低盐胁迫时,存活率显著高于对照组。王鑫磊等[82]研究发现,在饲料中添加0.2%的胆固醇(饲料中胆固醇含量为2.45 g/kg)时,凡纳滨对虾的免疫灵敏性有显著提高,获得最佳的抗弧菌能力和理想的抗亚硝态氮胁迫能力。胆固醇添加量为0.1~0.3 g/kg,即饲料胆固醇含量在1.57~3.43 g/kg时,凡纳滨对虾表现出较好的抗低温胁迫能力。

但胆固醇的添加量也不是越高越好,当添加量过高时会影响生长[6, 64, 72]。Zhang等[8]的研究发现,随着饲料中胆固醇添加量的增加,幼虾的血清谷丙转氨酶(glutamic-pyruvic transaminase,ALT)活性和亚成体的谷草转氨酶(glutamic oxalacetic transaminase,AST)活性均呈先降低后升高的趋势。这一现象可能是由肝胰腺中胆固醇过多引起的,这可能会导致肝脏功能障碍。目前,已有相关报道表明饲料中添加胆固醇能提高甲壳动物的免疫力,但提高免疫力与促进生长的添加量并不一定一致[81],且添加过多反倒会影响生长,不同甲壳动物的胆固醇适宜添加量还需要进一步探索。

3.4 饲料胆固醇对甲壳动物蜕壳、繁殖的影响

虾蟹类胆固醇放射性示踪的研究发现,胆固醇可转换为烃黄体酮、睾丸素及11-酮睾丸素等类固醇激素,主要集中在性腺和真皮组织,表明胆固醇与虾蟹类的生殖、蜕壳有关[5]。甲壳动物的生长总是伴随着蜕壳活动。甲壳动物在蜕壳时死亡,或者蜕壳不久后死亡的现象被称为蜕壳死亡综合征[83]。胆固醇是与蜕壳活动相关的蜕壳激素形成的前体物质[72, 84],在甲壳动物饲料中补充适量固醇,可以减少蜕壳死亡综合征的发生。Han等[79]报道,在三疣梭子蟹幼蟹饲料中添加1%的胆固醇,可以减少其蜕壳死亡综合征的发生。在饮食需求之外加入胆固醇也已被认为是减少凡纳滨对虾蜕壳死亡综合征发生的一种潜在的方法[85]。除此之外,Kanazawa等[48]曾报道添加胆固醇的饲料能增加日本对虾的蜕壳次数,Tao等[84]对中华绒螯蟹(Sinensis meggalopae)投喂含胆固醇的饲料时,蟹的蜕壳频率随胆固醇添加量的增加而上升。克氏原螯虾饲料中添加0.5%的胆固醇时,蜕壳次数达到最大[68]。眼柄中X器官产生的蜕壳抑制激素(molting inhibitory hormone,MIH)可能对Y器官中合成的促蜕壳激素具有负调控作用,并延长蜕壳周期[86-89]。饲料中添加适量胆固醇可能在一定程度上减轻这种抑制效果,促进蜕壳激素的合成,从而促进蜕壳,缩短蜕壳时间,并减少蜕壳死亡综合征的发生。此外,Gong等[90]曾报道,眼柄中表达的蜕壳激素受体(ecdysone receptor,EcR)可能对MIH的分泌具有抑制作用。Zheng等[91]也发现在中华绒螯蟹饲料中添加适量(1.1%)的胆固醇可以增加EcR的表达量。但具体机制还需进一步探索。

蜕壳频率的提高可以对甲壳动物的生长起到促进作用,但对于蜕壳频率也不能一味提高。朱雅珠等[92]研究发现,在蟹的饲料中添加过高比例的胆固醇可造成蜕壳频率过快,性早熟的比例增加,不利于优势蟹种的培育。由于胆固醇还是甲壳动物性激素合成的前体物质[13],其对甲壳动物的性成熟和繁殖也起到了重要的调控作用[93]。Gehring[94]发现,桃红美对虾(Farfantepenaeus duorarum)卵巢胆固醇随着性腺发育成熟而逐渐增多。Teshima等[95]分析发现,游离态固醇是虾类卵巢脂肪的重要组分。这可能都是在为了合成性激素而做准备。甘信辉等[70]通过检测与生殖器官发育相关的AKP活性以及与精子代谢相关的乳酸脱氢酶(lactate dehydrogenase,LDH)活性[96],表明饲料中添加胆固醇还能够促进红鳌螯虾(Cherax quadricarinatus)精巢的发育。

饲料中胆固醇含量增加,促进蜕壳激素的合成,使蜕壳时间缩短,蜕壳频率加快,性腺发育加快,性早熟也明显升高。因而,在促进生长保证正常蜕壳与避免性早熟比例增高之间寻找适宜的胆固醇添加量尤为重要。

4 小结与展望

胆固醇是甲壳动物中多种激素形成的前体物质,对于甲壳动物的生存具有重要意义。由于甲壳动物不能从乙酸和甲羟戊酸合成胆固醇,外源食物中的胆固醇成为甲壳动物胆固醇的唯一来源。但胆固醇对甲壳动物的影响是复杂的,其对甲壳动物的生存、生长、体成分、健康以及繁殖都有不同程度的影响,且每种影响所需要的最适胆固醇添加量并不完全一致。在今后的研究中,以下几点研究内容值得深入探究:1)针对不同发育时期、不同生长环境以及不同养殖目的(繁殖或增重)的甲壳动物精准配制饲料,满足甲壳动物需求;2)胆固醇在甲壳动物中的消化机制研究;3)在以植物性蛋白质代替鱼粉作为饲料蛋白质源以及植物甾醇部分替代胆固醇的背景下,针对不同蛋白质源与脂肪源的饲料,对甲壳动物饲料中胆固醇的最适添加量进行一次系统地评估,并对植物甾醇的作用以及机理进行探究。
