本试验旨在研究黄芪多糖(Astragalus polysaccharides,APS)对黄颡鱼(Pelteobagrus fulvidraco)免疫细胞活性的影响。540尾鱼被随机分成6组,每组3个重复,每个重复30尾鱼,分别投喂添加0、300、600、900、1 200、1 500 mg/kg APS的饲料,饲养期为8周。采用密度梯度离心技术分离纯化黄颡鱼头肾巨噬细胞与外周血液白细胞;分别采用硝基蓝四氮唑(NBT)法、格里斯氏(Griesse)试剂法和噻唑蓝(MTT)比色法测定头肾巨噬细胞氧、氮呼吸爆发活性和外周血白细胞的增殖能力。结果表明:各APS组的黄颡鱼头肾巨噬细胞氧、氮呼吸爆发活性均高于空白对照,但仅1 200 mg/kg组鱼巨噬细胞氧呼吸爆发活性显著提高(P<0.05);1 200和1 500 mg/kg APS组氮呼吸爆发活性极显著提高(P<0.01);投喂黄芪多糖能显著提高外周血白细胞的增殖能力(P<0.05),且1 200和1 500 mg/kg组增殖效果尤为显著(P<0.01)。结果提示,APS可以有效地提高黄颡鱼免疫细胞的活性,最佳剂量为1 200 mg/kg。
The trial was conducted to investigate the effects of Astragalus polysaccharides (APS) on the activities of immunological cells of yellow catfish (Pelteobagrus fulvidraco). Five hundred and forty yellow catfish were randomly divided into 6 groups with 3 replicates, and 30 individuals in each replicate in an 8-week trial. Fish were fed diets with 0, 300, 600, 900, 1 200 and 1 500 mg/kg APS, respectively. The head-kidney macrophages and peripheral blood leukocytes of yellow catfish were separated and purified by continuous density gradient centrifugation; the respiratory burst activity of oxygen and nitrogen of head-kidney macrophages and the proliferation of peripheral blood leukocytes were measured by nitroblue tetrazolium method, Griesse reagent method and thiazolyl blue colorimetric assay, respectively. The results showed as follows: the oxygen and nitrogen respiratory burst activities of head-kidney macrophages in each APS groups were higher than those of blank control, however, oxygen respiratory burst activity of head-kidney macrophages in 1 200 mg/kg APS group was increased significantly (P<0.05); the nitrogen respiratory burst activities in 1 200 and 1 500 mg/kg APS groups were extremely higher than those of blank control (P<0.01); the proliferation of peripheral blood leukocytes of yellow catfish in each APS groups was significantly enhanced (P<0.05), and that in 1 200 and 1 500 mg/kg APS groups was extremely enhanced (P<0.01). The results indicate that APS can efficiently enhance the activities of immunological cells of yellow catfish, and the proper level is 1 200 mg/kg.[Chinese Journal of Animal Nutrition, 2011, 23(5):887 -892]
