天然和合成腐植酸对热应激蛋鸡生产性能、血清促性腺激素浓度、肝脏抗氧化能力和肠道形态结构的影响

摘要: 本试验旨在探讨饲粮中添加天然和合成腐植酸对热应激蛋鸡生产性能、血清促性腺激素浓度、肝脏抗氧化能力和肠道形态结构的影响。选用60周龄、体重[(1.32±0.24)kg]相近、产蛋率[(80.16±0.21)%]一致的健康海兰白蛋鸡540只,随机分成4个组,每组9个重复,每个重复15只鸡。对照组1在常温[(25±1)℃]条件下饲养,饲喂基础饲粮;其他组在高温[(35±1)℃]条件下饲养,分别饲喂基础饲粮(对照组2)及在基础饲粮中添加1 000 mg/kg天然腐植酸(腐植酸A组)和1 000 mg/kg合成腐植酸(腐植酸B组)的试验饲粮。试验期4周。结果表明:1)与对照组1相比,对照组2蛋鸡的产蛋率(LR)、平均日采食量(ADFI)、血清促黄体激素(LH)浓度、空肠绒毛高度(V)、绒毛高度/隐窝深度(V/C)和回肠V极显著降低(P < 0.01),不合格蛋率(UER)、死亡率、肝脏丙二醛(MDA)含量和过氧化氢酶(CAT)活性极显著增加(P < 0.01);与对照组2相比,饲粮中添加天然腐植酸极显著提高蛋鸡的LR、血清LH浓度、空肠V、V/C和回肠V(P < 0.01),极显著降低不合格蛋率、肝脏MDA含量和CAT活性(P < 0.01)。综上所述,饲粮中添加腐植酸可提高蛋鸡的抗氧化能力,调节血清促性腺激素浓度,改善生产性能,减轻热应激对肠道的损伤,且在本试验条件下天然腐植酸效果更佳。

Effects of Natural and Synthetic Humic Acids on Performance, Serum Gonadotropin Concentrations, Liver Antioxidant Capacity and Intestinal Morphological Structure of Laying Hens under Heat Stress

Abstract: This study was conducted to explore the effects of dietary natural and synthetic humic acids (HAs) on performance, serum gonadotropin concentrations, liver antioxidant capacity and intestinal morphological structure of laying hens under heat stress. Five hundred and forty healthy Hy-Line White laying hens of 60-week-old with similar body weight of[(1.32±0.24) kg] and consistent laying rate of[(80.16±0.21)%] were randomly divided into 4 groups with 9 replicates per group and 15 hens per replicate. Laying hens in control group 1 were fed a basal diet at normal temperature[(25±1)℃], and the others were fed a basal diet (control group 2) and the basal diets supplemented with 1 000 mg/kg natural humic acids (group humic acids A) and 1 000 mg/kg synthetic humic acids (group humic acids B) at high temperature[(35±1)℃]. The experiment lasted for 4 weeks. The results show as follows:1) compared with control group 1, laying rate (LR), average daily feed intake (ADFI), serum luteinizing hormone (LH) concentration, jejunum villus height (V) and the ratio of villus height to crypt depth (V/C) and ileum V of laying hens in control group 2 were significantly decreased (P < 0.01), and unqualified eggs rate (UER), mortality, liver malondialdehyde (MDA) content and catalase (CAT) activity were significantly increased (P < 0.01). Compared with control group 2, dietary natural humic acids significantly increased LR, serum LH concentration, jejunum V and V/C and ileum V of haying hens (P < 0.01), and significantly decreased UER, liver MDA content and CAT activity (P < 0.01). In conclusion, dietary humic acids can increase antioxidant capacity, regulate serum gonadotropin concentrations, improve performance and alleviate intestine injury caused by heat stress of laying hens, and the effects of natural humic acids is better in this experiment.

由于全球气候变暖,热应激逐渐成为影响畜牧业发展的重要因素之一[1]。在美国,由于畜禽在温度超出热舒适区域的地方和季节饲养而导致了畜牧业的巨大经济损失[2]。现代家禽养殖业对高温较为敏感,在一定的高温条件下,禽类机体内易产生活性氧(ROS),从而促使机体进入氧化应激阶段[3],为了保护自身免受ROS的有害损伤,机体内会发生抗氧化反应,最终造成热应激[4]。热应激对家禽生理行为、免疫应答以及食品质量和安全等方面都有不利的影响[4-5]。此外,胃肠道对热应激亦高度敏感[6],热应激导致的胃肠道结构改变和完整性受损均可能导致肠道损伤,甚至对家禽的健康造成伤害[7]

研究表明,饲粮中添加饲料添加剂如维生素E、维生素C[8-9]和维生素A[10]等可通过其自身良好的抗氧化应激能力来减缓禽类的热应激状况。腐植酸(humic acids,HAs)也具有减缓氧化应激的作用,其抗氧化能力已得到学术界的广泛关注与重视[11]。腐植酸可作为有机化合物氧化的电子传递者[12],在氧化还原反应中起重要作用,已有研究表明在腐植酸的分子结构中存在与辅酶Q相似的物质,其可能会影响机体的氧化还原系统[13]。腐植酸分子结构中存在羟基、羧基、羰基和巯基等官能团,其可通过去除机体ROS等物质来提高机体的抗氧化能力[14-15]。腐植酸有抗菌、抗炎和抗病毒作用[16-17],可促进动物的生长发育,提高机体的免疫能力[13],但有关腐植酸对动物热应激的缓解作用鲜有报道。根据来源可将腐植酸分为天然腐植酸和合成腐植酸,不同来源的腐植酸其有效成分和含量有所差异,这可能导致腐植酸在畜禽养殖上的作用效果不同。因此,本试验主要研究饲粮中添加天然和合成腐植酸对热应激蛋鸡生产性能、血清促性腺激素浓度、肝脏抗氧化能力和肠道形态结构的影响,旨在探究腐植酸对蛋鸡热应激的减缓作用。

1 材料与方法 1.1 试验材料

天然腐植酸:由水体、土壤和煤炭等物质在自然条件下形成的,其有机有效成分含量(风干基础)为62.88%,主要成分为高分子有机弱酸和少量未知营养因子;合成腐植酸:在人为控制条件下通过化学合成、生物发酵等形成的,其有机有效成分含量(风干基础)为60.51%,主要成分为高分子有机弱酸混合物。

1.2 试验设计

采用单因子试验设计,选用60周龄、体重[(1.32±0.24) kg]相近、产蛋率[(80.16±0.21)%]一致的健康海兰白蛋鸡540只,随机分成4个组,每组9个重复,每个重复15只鸡。对照组1(CON1组)在常温[(25±1) ℃]条件下饲养,饲喂基础饲粮;其他组在高温[(35±1) ℃]条件下饲养,分别饲喂基础饲粮(对照组2,CON2组)及在基础饲粮中添加1 000 mg/kg天然腐植酸(腐植酸A组,HAA组)和1 000 mg/kg合成腐植酸(腐植酸B组,HAB组)的试验饲粮。试验期4周。基础饲粮参照NRC(1994)和《海兰白蛋鸡饲养手册》配制成粉料,其组成及营养水平见表 1

1.3 饲养管理

饲养试验于农业农村部华东动物营养与饲料重点实验室研究试验基地的鸡舍进行,采用上、中、下3层阶梯式笼养,每层5个鸡笼(45 cm×30 cm×30 cm),每笼3只鸡。按蛋鸡场常规方法进行饲养,采用不锈钢料槽和乳头饮水器,每天早晚定时2次喂料和清粪,自由采食、饮水,鸡舍采用人工光照16L : 8D,光照强度为20 lx。每周对鸡舍进行1次消毒。

1.4 样品采集与制备

于试验结束时,每个重复随机选取1只蛋鸡(每组9只),禁食12 h后采血5 mL于促凝管中,37 ℃水浴5 min,3 000 r/min离心10 min制成血清,上清液分装于1.5 mL Eppendorf管中,置于-80 ℃保存待用。然后进行屠宰,采集蛋鸡的肝脏、十二指肠、空肠和回肠组织,做好标记,用锡箔纸包好,于-80 ℃保存待用。

1.5 测定指标与方法 1.5.1 生产性能

以重复为单位,每天记录蛋鸡的产蛋量、总蛋重、鸡只死亡数和不合格蛋(破壳蛋、软壳蛋等)数量,每周进行饲料消耗量的记录。基于记录的生产数据计算产蛋率(LR)、不合格蛋率(UER)、平均日采食量(ADFI)、平均蛋重(AEW)、料蛋比(F/E)和死亡率。

1.5.2 血清促性腺激素浓度

采用酶联免疫吸附试验(ELISA)法测定血清促性腺激素浓度,包括促卵泡素(FSH)和促黄体素(LH),严格遵循ELISA试剂盒说明书进行操作,试剂盒购买于南京建成生物工程研究所。

1.5.3 肝脏抗氧化指标

将采集的肝脏组织用4 ℃的生理盐水制备成10%的肝脏上清液,测定肝脏组织的抗氧化指标,包括丙二醛(MDA)含量与总超氧化物歧化酶(T-SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性,严格按照试剂盒说明书进行操作,试剂盒购买于南京建成生物工程研究所。

1.5.4 肠道绒毛形态结构

分别轻轻挤出采集好的十二指肠、空肠和回肠中的内容物,将肠段切成约1 cm长的片段,用生理盐水冲洗干净肠道内的残留物,拭去残留的水分,放入10%的甲醛溶液中固定,用石蜡切片,苏木精-伊红(HE)染色,参照Taklimi等[18]的研究方法,使用Image Pro-Plus 7.0图像分析软件(Media Cybernetics,美国)观察肠道形态结构,测量绒毛高度(V)和隐窝深度(C),计算V/C。

1.6 数据统计分析

数据采用SPSS 20.0统计软件进行单因素方差分析(one-way ANOVA),用Duncan氏法进行多重比较检验。P < 0.01表示差异极显著,P < 0.05表示差异显著。

2 结果 2.1 天然和合成腐植酸对热应激蛋鸡生产性能的影响

表 2可知,与对照组1相比,对照组2蛋鸡的LR和ADFI极显著降低(P < 0.01),不合格蛋率和死亡率极显著增加(P < 0.01)。与对照组2相比,饲粮中添加天然腐植酸极显著增加蛋鸡的LR(P < 0.01),极显著降低不合格蛋率(P < 0.01);饲粮中添加天然和合成腐植酸对ADFI和F/E无显著影响(P>0.05)。各组蛋鸡的AEW无显著差异(P>0.05)。

2.2 天然和合成腐植酸对热应激蛋鸡血清促性腺激素浓度的影响

表 3可知,与对照组1相比,对照组2蛋鸡的血清LH浓度极显著降低(P < 0.01)。与对照组2相比,饲粮中添加天然腐植酸极显著增加蛋鸡的血清LH浓度(P < 0.01)。各组蛋鸡的血清FSH浓度无显著差异(P>0.05)。

2.3 天然和合成腐植酸对热应激蛋鸡肝脏抗氧化指标的影响

表 4可知,与对照组1相比,对照组2蛋鸡的肝脏MDA含量和CAT活性极显著增加(P < 0.01)。与对照组2相比,饲粮中添加天然腐植酸极显著降低蛋鸡的肝脏MDA含量和CAT活性(P < 0.01)。各组蛋鸡的肝脏T-SOD和GSH-Px活性无显著差异(P>0.05)。

2.4 天然和合成腐植酸对热应激蛋鸡肠道形态结构的影响

图 1可知,对照组1蛋鸡的十二指肠、空肠和回肠并未发现明显的组织病理学损伤,而对照组2的3段肠段中绒毛结构均有损伤折断且分层明显;饲粮中添加天然和合成腐植酸除十二指肠外均可在一定程度上减少绒毛结构的折断分层现象,有效缓解热应激引起的肠道绒毛损伤。

表 5可知,与对照组1相比,对照组2蛋鸡的空肠V、V/C和回肠V极显著降低(P < 0.01)。与对照组2相比,饲粮中添加天然和合成腐植酸均极显著增加蛋鸡的空肠V、V/C和回肠V(P < 0.01)。各组蛋鸡的十二指肠V、C和V/C无显著差异(P>0.05)。

3 讨论 3.1 天然和合成腐植酸对热应激蛋鸡生产性能的影响

热应激会对产蛋母鸡的生产性能产生不利影响,例如降低LR和饲料转化率,提高死亡率和不合格蛋率等[19-21]。本试验中,热应激导致的蛋鸡LR的降低和不合格蛋率的增加可通过在饲粮中添加腐植酸来改善。由此可知,在高温下添加腐植酸对LR和不合格蛋率具有与在正常温度下添加腐植酸相似的效果[22-24]。热应激时,饲粮中添加腐植酸不同程度增加了蛋鸡的血清LH浓度,进而提高了蛋鸡的生产性能,其中天然腐植酸表现出的效果更佳,可能的原因是天然腐植酸中存在的未知营养因子具有提高生产性能和促性腺激素浓度的作用,具体的作用机制还需对天然腐植酸进行进一步的研究。

3.2 天然和合成腐植酸对热应激蛋鸡血清促性腺激素浓度的影响

促性腺激素浓度的变化可能会影响蛋鸡的产蛋性能。研究报道,热应激蛋鸡的血清LH浓度可能会降低[25-26],这与本试验结果相一致。本试验中,蛋鸡的血清LH浓度在热应激时降低,但在饲粮中添加腐植酸后增加。一般来说,当排卵频率最高时机体的LH浓度会激增,而卵巢周期黄体期时LH浓度则降到最低[27]。Tanabe等[28]研究表明,对蛋鸡禁食7 d可减少产蛋,显著降低血浆LH浓度。当母鸡恢复产蛋时,血浆LH浓度则开始升高[29]。由此可推断,在家禽养殖中血清LH浓度的增加可以促进排卵的增加,从而提高LR并降低破壳蛋和软壳蛋的产生。蛋鸡生产性能与血清LH浓度之间可能存在正相关关系,但需要进一步的研究来探索产蛋量与血清LH浓度之间的关系。

3.3 天然和合成腐植酸对热应激蛋鸡肝脏抗氧化能力的影响

随着蛋鸡周龄的增加,更多的脂肪会积聚在肝脏中。脂肪肝与产蛋性能下降有关,过度脂肪肝还与产蛋母鸡的死亡率有关[30]。在脂肪肝中会同时存在抗氧化能力降低和蛋白质氧化[31],抗氧化能力表明体内自由基和活性氧的状况,肝脏抗氧化能力的提高可以减轻脂肪肝的形成。因此,肝脏的抗氧化指标是反映蛋鸡健康状况的重要指标。热应激可能损害线粒体功能并诱导身体不同部位的氧化损伤[32]。本试验中,热应激使蛋鸡的肝脏MDA含量和CAT活性均有所增加,但通过在饲粮中添加天然腐植酸而有所降低。CAT是重要的抗氧化酶,可以分解体内的过氧化氢[33],CAT活性增加可能是因为体内过氧化氢羟自由基含量升高,机体为了维持体内平衡从而导致CAT活性增加。MDA则是体内脂质过氧化的产物,间接反映体内过氧化程度[34]。Ganesan等[35]研究发现,应激会显著增加骨骼肌中的MDA含量,这与本试验研究结果相一致。本试验研究表明,饲粮中添加天然腐植酸通过一定程度的抗氧化反应刺激可减少由热应激引起的氧化损伤。

3.4 天然和合成腐植酸对热应激蛋鸡肠道形态结构的影响

正常的肠道结构可以防止细菌移位,促进营养物质吸收和能量消耗[36-37];当肠道形态结构受损时则会影响机体的健康状况。本试验中,热应激时可观察到明显受损的蛋鸡肠道绒毛结构和V显著缩短的空肠和回肠,这与Deng等[7]在34 ℃时对蛋鸡进行的为期12 d的试验结果一致,表明热应激已影响到了蛋鸡的肠道结构和功能。热应激可能在肠道中产生了毒素,毒素的存在与V的缩短和C的增加有关,导致机体营养吸收不良和生产性能降低[38],进而影响到蛋鸡的正常机能。本试验还发现热应激对肠道的负面影响从大到小依次为空肠、回肠和十二指肠,因此热应激主要影响的肠道可能是空肠。本试验在饲粮中添加腐植酸观察到相对正常的绒毛形态结构,可能是由于腐植酸对热应激蛋鸡的肠道形态产生了保护作用,但腐植酸对肠道具体的保护机制还有待进一步研究。

综上所述,热应激时饲粮中添加腐植酸可提高机体的抗氧化能力,改善体内促性腺激素浓度,缓减机体肠道形态结构损伤,提高生产性能,从而缓解蛋鸡的热应激状况。

4 结论

① 高温条件使蛋鸡表现出热应激症状:生产性能下降,体内促性腺激素紊乱,同时增加了肝脏氧化应激和肠道结构损伤。

② 在本试验条件下,饲粮中添加腐植酸可缓解蛋鸡的热应激状况,提高抗氧化能力,减少肠道结构损伤,且天然腐植酸还可恢复生产机能和促性腺激素分泌,对热应激的缓减效果更佳。
