饲粮能量水平对德州驴生长性能、营养物质消化率及血清生化指标的影响

摘要: 本研究旨在探讨饲粮能量水平对德州驴生长性能、营养物质消化率及血清生化指标的影响。试验选用健康的7~8月龄德州公驴36头,体重(123.9±5.0)kg,随机分为3个组,每组3个重复,每个重复4头驴。各组饲粮消化能水平分别为10.43(低能组)、11.03(中能组)和11.90 MJ/kg(高能组),粗蛋白质水平相近。预试期20 d,正试期40 d。结果表明:1)中能组和高能组的平均日增重显著高于低能组(P < 0.05),且高能组的平均日增重显著高于中能组(P < 0.05)。中能组和高能组的料重比显著低于低能组(P < 0.05),且高能组的料重比显著低于中能组(P < 0.05)。2)中能组和高能组的干物质、有机物和碳水化合物消化率显著高于低能组(P < 0.05),且高能组的干物质、有机物和碳水化合物消化率显著高于中能组(P < 0.05)。3)高能组的血清总蛋白、甘油三酯含量显著高于中能组和低能组(P < 0.05)。中能组和高能组的血清白蛋白含量显著高于低能组(P < 0.05),且高能组的血清白蛋白含量显著高于中能组(P < 0.05)。中能组和高能组的血清乳酸脱氢酶活性显著低于低能组(P < 0.05),且高能组的血清乳酸脱氢酶活性显著低于中能组(P < 0.05)。由此可见,在本试验饲粮能量水平范围内(10.43~11.90 MJ/kg),饲喂高能量水平(11.90 MJ/kg)饲粮能够较好地提高德州驴的平均日增重,同时提高干物质、有机物和碳水化合物消化率,降低料重比。

Effects of Dietary Energy Level on Growth Performance, Nutrient Digestibility and Serum Biochemical Indices of Dezhou Donkeys

Abstract: The object of this study was to investigate the effects of dietary energy level on growth performance, nutrient digestibility and serum biochemical indices of Dezhou donkeys. Thirty-six healthy Dezhou male donkeys aged from 7 to 8 months with similar body weight of (123.9±5.0) kg were selected and randomly divided into 3 groups with 3 replicates in each group and 4 donkeys in each replicate. Dietary digestible energy levels of 3 groups were 10.43 (low energy group), 11.03 (mid energy group) and 11.90 MJ/kg (high energy group), respectively, and the crude protein levels were similar. The adaptation period was 20 days and the experimental period was 40 days. The results showed as follows: 1) the average daily gain of mid energy group and high energy group was significantly higher than that of low energy group (P < 0.05), and the average daily gain of high energy group was significantly higher than that of mid energy group (P < 0.05). The feed to gain ratio of mid energy group and high energy group was significantly lower than that of low energy group (P < 0.05), and the feed to gain ratio of high energy group was significantly lower than that of mid energy group (P < 0.05). 2) The dry matter, organic matter and carbohydrate digestibility of mid energy group and high energy group was significantly higher than that of low energy group (P < 0.05), and the dry matter, organic matter and carbohydrate digestibility of high energy group was significantly higher than that of mid energy group (P < 0.05). 3) The serum total protein and triglyceride contents of high energy group were significantly higher than those of mid energy group and low energy group (P < 0.05). The serum albumin content of mid energy group and high energy group was significantly higher than that of low energy group (P < 0.05), and the serum albumin content of high energy group was significantly higher than that of mid energy group (P < 0.05). The serum lactate dehydrogenase activity of mid energy group and high energy group was significantly lower than that of low energy group (P < 0.05), and the serum lactate dehydrogenase activity of high energy group was significantly lower than that of mid energy group (P < 0.05). In conclusion, in the range of dietary energy level in this experiment (10.43 to 11.90 MJ/kg), feeding high energy level diet can better improve the average daily gain of Dezhou donkeys, improve the dry matter, organic matter and carbohydrate digestibility, and decrease the feed to gain ratio.

德州驴是我国优良的草食动物家畜,具有较高的生产力[1]。驴肉被认为是一种优质的食物,脂肪和胆固醇含量较低,但不饱和脂肪酸和蛋白质含量丰富[2]。此外,阿胶是一种传统的功能食品,具有提高免疫力和抗病能力等多种功能。目前,毛驴养殖已成为我国畜牧业的重要产业。然而,目前世界范围内并无统一的驴饲养标准,与其他家畜相比,驴营养研究还处于相对初级的阶段。能量作为饲粮的重要组成部分,对动物的生长性能和饲料利用效率起着重要作用[3-4]。淀粉是饲粮中主要的碳水化合物来源[5]和能量提供者[6],通过微生物消化纤维素产生的短链脂肪酸(SCFA)也提供了驴生长发育所需的很大一部分能量。合适的饲粮能量水平可以较好地满足动物的需求并且提高生长性能。然而,关于饲粮能量水平对驴营养物质消化率的研究甚少[7],特别是在生长前期的驴驹上的研究鲜有报道。在适宜的饲粮能量水平范围内,可以通过提高饲粮能量水平来提高驴驹的生长性能,并结合饲粮蛋白质水平,保持适宜的能量和蛋白质比例来提高营养物质消化率。因此,本试验通过研究饲粮能量水平对德州驴生长性能、营养物质消化率及血清生化指标的影响,确定德州驴早期育肥的适宜饲粮能量水平,对提高德州驴的生产效率具有重要意义。

1 材料与方法 1.1 试验设计

试验选用健康的7~8月龄德州公驴36头,体重(123.9±5.0) kg,随机分为3个组,每组3个重复,每个重复4头驴。各组饲粮消化能水平分别为10.43(低能组)、11.03(中能组)和11.90 MJ/kg(高能组),粗蛋白质水平相近。预试期20 d,正试期40 d。使用酸不溶木质素(ADL)作为内源指示剂进行消化试验。试验时间为2019年4月26日至2019年6月25日,在山东无棣进行。

试验饲粮配制参考山东省地方标准[8],将饲粮制成直径6~10 mm的颗粒型全混合日粮(TMR),试验饲粮组成及营养水平见表 1

1.2 饲养管理与样品采集

试验驴室内栏饲养殖,试验期间自由采食和饮水,预试期以适应环境和饲粮,正试期分别于每天06:00和18:00进行饲喂。正试期间每个组随机选取6头驴(第20天和第40天)分别采用随排随捡拾的方式收集取样新鲜粪便,然后粉碎过1 mm孔筛[9],混合后制样置于广口瓶中进行各营养成分的测定。

1.3 测定指标及方法 1.3.1 生长性能

正试期开始前和结束时所有驴驹于早晨空腹称重,记为初始体重和结束体重。试验期间,以重复为单位,并每天准确记录采食量。

1.3.2 营养物质消化率

样品中的水分、粗灰分、粗蛋白质、粗脂肪、钙、磷、中性洗涤纤维和酸性洗涤纤维含量的测定均采用国标法[10-15]。准确记录每日采食量并混合均匀后取样,通过内源指示剂ADL法测定营养物质消化率[16]

1.3.3 血清生化指标

于正试期(第40天)使用真空促凝管和抗凝管各采集血液5 mL,每组随机选取4头。将真空促凝管于37 ℃水浴静置10 min,3 000 r/min离心10 min,分离血清,-20 ℃保存待测,采用全自动生化分析仪(COBASMI RA Plus,罗士公司,瑞士)测定血清中总蛋白、白蛋白、甘油三酯、总胆固醇、高密度脂蛋白和低密度脂蛋白含量及谷丙转氨酶、谷草转氨酶、碱性磷酸酶和乳酸脱氢酶活性。

1.4 统计分析

试验数据采用Excel 2016和SPSS 23.0进行统计分析和显著性检验,结果平均值和均值标准误(SEM)表示,P < 0.05为差异显著。

2 结果 2.1 饲粮能量水平对德州驴生长性能的影响

饲粮能量水平对德州驴生长性能的影响见表 2。各组之间的初始体重和平均日采食量无显著差异(P>0.05)。中能组和高能组的结束体重显著高于低能组(P<0.05)。中能组和高能组的平均日增重显著高于低能组(P<0.05),且高能组的平均日增重显著高于中能组(P<0.05)。中能组和高能组的料重比显著低于低能组(P<0.05),且高能组的料重比显著低于中能组(P<0.05)。

2.2 饲粮能量水平对德州驴营养物质消化率的影响

饲粮能量水平对德州驴营养物质消化率的影响见表 3。中能组和高能组的干物质、有机物和碳水化合物消化率显著高于低能组(P<0.05),且高能组的干物质、有机物和碳水化合物消化率显著高于中能组(P<0.05)。各组之间的粗蛋白质、粗脂肪、酸性洗涤纤维和中性洗涤纤维消化率无显著差异(P>0.05)。

2.3 饲粮能量水平对德州驴血清生化指标的影响

饲粮能量水平对德州驴血清生化指标的影响见表 4。高能组的血清总蛋白、甘油三酯含量显著高于中能组和低能组(P<0.05)。中能组和高能组的血清白蛋白含量显著高于低能组(P<0.05),且高能组的血清白蛋白含量显著高于中能组(P<0.05)。中能组和高能组的血清乳酸脱氢酶活性显著低于低能组(P<0.05),且高能组的血清乳酸脱氢酶活性显著低于中能组(P<0.05)。各组之间的血清总胆固醇、高密度脂蛋白、低密度脂蛋白含量和谷丙转氨酶、谷草转氨酶、碱性磷酸酶活性无显著差异(P>0.05)。

3 讨论 3.1 饲粮能量水平对德州驴生长性能的影响

饲粮能量水平对草食动物的生长具有重要作用[17],适宜的饲粮能量水平是提高平均日增重的决定性因素,李文强[18]研究发现,随着饲粮能量水平的提高,公驴的平均日增重显著提高,其中11.70 MJ/kg时增重效果最好。周艳等[19]研究表明,肉驴育肥前期饲粮能量水平在11.97 MJ/kg时育肥效果最好,料重比最低。本研究结果表明,提高饲粮能量水平能够提高驴驹的平均日增重和饲料利用率,在饲粮粗蛋白质水平为15%左右时,饲粮能量水平为11.90 MJ/kg组驴驹平均日增重显著高于其他各组,可能是提高饲粮能量水平会促进更多的能量用于机体的生长,从而增重效果较好。

3.2 饲粮能量水平对德州驴营养物质消化率的影响

营养物质消化率是评价饲粮营养价值的重要指标,反映营养物质的消化吸收状况,也可以反映饲粮营养是否平衡。在本试验的饲粮能量水平范围内,随着饲粮能量水平的升高,干物质、有机物和碳水化合物消化率显著提高。周艳[20]研究表明,随着饲粮能量水平的升高,肉驴的营养物质消化率也相应上升,可能通过影响消化酶活性来影响营养物质的消化吸收。当饲粮中提供足够的蛋白质时,适宜的能量水平可以使能量和蛋白质的供应更加平衡[21-22],可能是由于适宜的能量水平有利于肠道健康发育及有益微生物菌群的生长[23],从而更好地吸收营养物质。然而,不同的饲粮粗蛋白质水平以及蛋白质组成均会影响肉驴的生长育肥性能和饲粮营养物质的消化利用率。本研究是在保持饲粮粗蛋白质水平基本不变的情况下对不同能量水平之间进行的探索,因此,有必要针对不同育肥时期和不同的饲粮配比进行探索,得出最佳饲粮能量和粗蛋白质水平。

3.3 饲粮能量水平对德州驴血清生化指标的影响

血清白蛋白含量可以反映机体对摄入蛋白质的消化利用情况。本试验中,随着饲粮能量水平的提高,血清总蛋白和白蛋白含量均提高,表明机体蛋白质代谢增强。动物采食量降低、供能不足等均可引起血清乳酸脱氢酶活性升高,参与丙酮酸转化为乳酸的糖酵解过程。本试验中,血清乳酸脱氢酶活性随着饲粮能量水平的提高而显著降低,其活性降低可能使得有氧作用加强,提高饲粮的能量利用效率和机体的消化吸收能力,从而提高了平均日增重和饲料利用率。

甘油三酯是反映脂类代谢的指标,在本试验饲粮能量水平范围中,提高饲粮能量水平其相应的血清甘油三酯含量也提高,可能是由于饲粮能量水平的提高导致驴驹对脂肪酸和甘油的合成增强导致的,而血清总胆固醇含量无显著差异,其中高密度脂蛋白和低密度脂蛋白都是其运载介质,各组之间的血清高密度脂蛋白和低密度脂蛋白含量也无显著差异。

4 结论

在本试验饲粮能量水平范围内(10.43~11.90 MJ/kg),饲喂高能量水平(11.90 MJ/kg)饲粮能够较好地提高德州驴的平均日增重,同时提高干物质、有机物和碳水化合物消化率,降低料重比。
