瘤胃微生物区系的影响因素及其调控措施

摘要: 瘤胃中存在着大量微生物,这些微生物在反刍动物的消化过程中起着十分重要的作用。近年来研究发现瘤胃微生物区系受饲粮及外界环境等多种因素的影响,基于这些因素,通过调控瘤胃微生物区系可提高瘤胃对饲粮的利用率,促进生产性能的发挥。本文综述了瘤胃微生物区系的组成及影响因素,重点介绍了瘤胃微生物区系调控在养牛业中的应用,旨在为奶牛和肉牛研究中瘤胃微生物区系的调控提供参考。

Abstract: There is a large number of microorganisms in the rumen, which play an important role in the digestion of ruminants. Recently, studies have found that the rumen microbial flora was influenced by many factors such as diet and external environment. Based on these factors, the rumen microbial flora can be regulated to improve the ruminal utilization rate of diets and promote the performance. In order to provide references for the regulation of rumen microbial flora in beef and dairy cow study, this paper summarized the composition and influencing factors of rumen microbial flora, and emphatically introduced the application of rumen microbial flora regulation in cattle industry.

随着科技的发展,对反刍动物瘤胃的研究除了挥发性脂肪酸(VFA)及氨态氮(NH3-N)等常规发酵指标的研究外,对瘤胃中的微生物区系也进行了系统的研究。瘤胃作为反刍动物的特殊器官,存在着大量的微生物,在这些微生物的作用下,饲粮中的植物纤维性物质可以被降解成反刍动物可以利用的营养物质,生产出人类所需要的奶和肉制品[1]。瘤胃微生物通过发酵可为宿主提供70%左右的能量需要,瘤胃微生物区系的变化与动物生产效率密切相关[2-3]。瘤胃微生物区系的稳定有利于宿主自身健康,维持生产性能的发挥,相反,瘤胃微生物区系的紊乱会导致宿主免疫力降低。因此,维持瘤胃微生物区系的稳定具有重要意义。

在生产中,由于饲粮及生理状态的变化,会导致动物瘤胃微生物群落结构发生变化,进而影响影响机体的健康状况[4-5]。有研究证实,人为地对消化道微生物区系进行调控,可以实现对微生物宿主的代谢进行调控[6]。通过人为手段调控瘤胃微生物区系来影响宿主的方法已经得到了越来越多的研究证实,现有研究认为通过改变饲粮组成及添加添加剂等方法均可以调控瘤胃微生物区系。同时,随着微生物移植在人医上的开展,近年来,微生物移植调控瘤胃微生物区系也取得了一定的研究成果。及时了解瘤胃微生物区系组成的变化,是调控瘤胃微生物区系的前提。本文介绍了瘤胃微生物区系的组成及影响因素,重点阐述了瘤胃微生物区系调控在养牛业中的应用,旨在为今后的相关研究提供参考。

1 瘤胃微生物区系的组成

瘤胃微生物区系并不是一出生就存在的,而是随着动物对母乳的采食、饲粮的转变以及外界环境的变化,微生物才逐步定植、存活进而繁殖扩增后而形成的[7]。动物幼龄阶段的瘤胃微生物区系组成与成熟动物差异明显,一般情况下,在动物保持健康以及外界环境稳定时,将4周岁以后反刍动物的瘤胃微生物区系定义为成熟的瘤胃微生物区系[8]。另有研究认为,在2周岁以前反刍动物瘤胃微生物都有可能发生较大变化[9]。现阶段针对如何定义成熟的瘤胃微生物区系存在争议,但不可否认的是,幼龄反刍动物瘤胃微生物区系还处在不稳定的状态。近年来,随着测序技术的发展,针对瘤胃微生物区系组成的变化过程进行了较为系统的研究,本文在查阅了相关文献[8-10]后,对瘤胃微生物区系组成随着日龄的变化情况进行了总结(表 1)。从表 1可以看出,在任何年龄段,瘤胃微生物区系中的优势菌均为变形菌门、拟杆菌门和厚壁菌门,但是这3种细菌在不同年龄段所占比例不同,幼龄动物变形菌门的丰度最高,成熟动物则变形菌门丰度明显降低。此外,Henderson等[11]对来自世界各国的742头反刍动物的瘤胃微生物区系进行了研究,结果发现,虽然分布的地理位置不同,但核心菌群是这些反刍动物所共有的,微生物在科、目和属水平上的优势菌分别为瘤胃球菌科和毛螺菌科,梭菌目和拟杆菌目,丁酸弧菌属、普氏菌属和瘤胃球菌属。对不同生存环境动物的瘤胃微生物区系进行研究,例如生活在高原的牦牛[12],也许可以从瘤胃微生物区系的角度进一步揭示动物对环境的适应机制。

2 瘤胃微生物区系的影响因素 2.1 动物的品种、年龄和生理状况

动物的品种和年龄是影响瘤胃微生物区系的直接因素,Cersosimo等[13]研究了荷斯坦牛、娟姗牛及杂交牛的瘤胃微生物区系组成,发现荷斯坦牛(30.7%)和杂交牛(30.3%)的产甲烷菌共存甲烷短杆菌(Methanobrevibacter thaueri)的丰度高于娟姗牛(26.7%)。而关于年龄对瘤胃微生物区系的影响在上文已经提到,此处不再赘述。动物所处的生理状况也是影响瘤胃微生物区系的重要因素。围产期是奶牛生理状况比较敏感的一个时期,瘤胃微生物区系在围产前期和后期具有差异,软壁菌门、绿弯菌门及疣微菌门在产前丰度较高,而放线菌门在产后丰度较高,此外,研究还发现,产后随着采食量的增加,瘤胃微生物区系在个体间的差异增加[14]。对奶牛围产期瘤胃优势菌进行分析,发现奶牛从围产期进入到泌乳阶段后,瘤胃微生物区系中拟杆菌门:厚壁菌门从6:1变为12:1[15]。奶牛围产期瘤胃微生物区系组成的变化是为了让机体更好地适应饲粮等因素的变化。相似的研究在国内也有报道,蒋涛[16]比较了产后7和50 d的瘤胃微生物区系组成,发现产后7 d的瘤胃微生物群落α多样性显著低于产后50 d。此外,泌乳期奶牛瘤胃微生物区系组成也不相同,泌乳中期的瘤胃微生物群落多样性显著高于泌乳前期[2]。这些研究表明动物的生理阶段对瘤胃微生物区系有较大影响,但同时这些变化也为我们更好地调控瘤胃微生物区系,进而调控机体功能的紊乱提供了可能。

2.2 饲粮

饲粮是瘤胃微生物区系的最大影响因素之一,饲粮中的营养成分不仅可以提供营养物质,还可以影响瘤胃微生物区系组成。通过查阅相关文献[17-21],本文总结了不同饲粮模式对瘤胃微生物区系组成的影响(表 2),众多研究证实了饲粮对瘤胃微生物区系组成的影响。此外,Zhao等[22]指出,随着奶牛饲粮中粗饲料的变化,瘤胃微生物区系结构也会发生变化,进而影响瘤胃微生物代谢,改变微生物蛋白和VFA的产量。生产中饲粮合理的使用是保证瘤胃微生物区系稳定的前提,对维持动物机体的健康及生产性能的发挥具有重要意义。

2.3 添加剂

生产实践中,在奶牛或肉牛饲粮中补充一些添加剂可以起到提高生产效率的作用,同时,这些添加剂可以通过改善动物瘤胃微生物群落结构进而对机体起到积极有益的作用。在查阅相关度比较高的文献[23-27]基础上,本文概括了一些添加剂对瘤胃微生物区系组成的影响(表 3)。

瘤胃微生物区系组成除受上述因素影响之外,动物所处的外界环境也会对瘤胃微生物区系产生影响,本文主要介绍以上几个因素。但是在生产实践中必须要综合考虑各个因素,力争使养殖效益达到最大。

3 瘤胃微生物区系的调控方法

通过前文综述可以看出,很多因素都可以对瘤胃微生物区系产生影响,基于这些因素,针对瘤胃微生物区系的调控,现有研究主要集中在饲粮和添加剂上,同时瘤胃微生物移植也在逐步开展,并且展现出良好的应用效果。因此,在这一部分将简单介绍饲粮及添加剂对瘤胃微生物区系的调控,重点阐述瘤胃微生物移植的研究进展。

3.1 饲粮及添加剂

瘤胃微生物区系最易受饲粮变化的影响,瘤胃微生物区系的组成与饲粮组成密切相关[28],因此,通过改变饲粮组成可以对瘤胃微生物区系进行有效调控。Chen等[29]研究表明,不同的饲粮结构会改变肉牛瘤胃微生物区系组成,进而使宿主更好地消化不同类型的饲粮。粗饲料的不同搭配在调控瘤胃微生物区系方面展现了一定的效果,相比于高比例的苜蓿青贮饲粮,高比例的玉米青贮饲粮可以增加瘤胃总细菌数,降低原虫数,同时降低甲烷菌的丰度,从而实现降低甲烷排放量[30]。在动物不同的生理阶段,通过改变饲粮结构可以调控瘤胃微生物区系,进而维持机体的健康或生产性能的发挥。此外,在饲粮中补充的一些添加剂对瘤胃微生物区系也可以起到调控作用。在犊牛上的研究发现,在断奶犊牛饲粮中添加乳酸菌,可以增加瘤胃中拟杆菌的数量,并降低瘤胃NH3-N的含量[31]。Hu等[32]指出,茶皂素可以起到抑制瘤胃原虫生长的作用。Torok等[33]研究表明,在饲粮中添加二十二碳六烯酸可以调控瘤胃菌群结构,降低产甲烷菌数量,并可以改善泌乳性能。在饲粮中添加自溶酵母和植物提取物,可以调控高精料条件下导致的瘤胃微生物紊乱,自溶酵母提高了瘤胃纤维降解菌(瘤胃球菌)的数量,而植物提取物降低了拟杆菌门的丰度,增加了丁酸的含量[34]。通过饲粮因素来调控瘤胃微生物区系已经被大量研究证实,上文简单介绍了相关研究,在今后仍需要更多的关注饲粮组成对瘤胃微生物区系及宿主健康的影响;此外,能调控靶向瘤胃微生物区系的饲粮值得更加深入的研究。

3.2 瘤胃微生物移植

瘤胃微生物移植是指将供体动物瘤胃内的微生物群落作为一个整体移植到受体动物瘤胃内,以达到重塑受体瘤胃微生物群落结构的目的。相对于人及单胃动物上的粪菌移植,瘤胃微生物移植起步较晚,目前,瘤胃微生物移植改善动物机体性能的机制仍然不清楚,操作程序也没有固定的标准。但是,不可否认的是,近年来的研究已经证实瘤胃微生物移植具有改善动物机体健康、促进生产性能发挥的作用。

3.2.1 瘤胃微生物移植条件

用于移植的瘤胃液或瘤胃内容物主要通过4种方式获取:1)从健康动物嘴里获得反刍食团[35];2)从屠宰后动物的瘤胃内获取[23];3)通过瘤胃液采样胃管获取[11];4)经永久性瘤胃瘘管获取[14]。由于通过从健康动物嘴里获取反刍食团和通过胃管获取瘤胃液量较少,并且对动物刺激较大,而屠宰动物则成本较高,目前较为常用的是经瘤胃瘘管获得移植供体。此外,用于移植的瘤胃液或瘤胃内容物要满足以下要求[16]:1)收集后的瘤胃液或瘤胃内容物应该尽快进行移植,最好在30 min内完成;2)用于移植的瘤胃液pH最好在6以上,低pH会降低原虫数量;3)收集的瘤胃液应过滤掉大分子物质;4)对泡沫较多的瘤胃液应进行原虫检测;5)用于犊牛移植所需瘤胃液体积约为1 L,而成年牛在8~16 L较为合适。

3.2.2 瘤胃微生物移植研究现状

由于瘤胃液采集方便,在研究中被广泛应用,但是瘤胃液是否能够代表瘤胃微生物区系值得思考。对此,有研究针对瘤胃液、瘤胃食糜及残渣的微生物区系进行了比较,研究表明,采样部位对瘤胃微生物区系的影响要小于饲粮,虽然瘤胃食糜在分析微生物区系组成方面更具代表性,但瘤胃食糜、瘤胃液和残渣在微生物多样性及组成结构方面具有高度相似性,因此,瘤胃液基本承载了瘤胃微生物群落结构,可以作为微生物移植的有效供体[36]

瘤胃微生物移植在调控动物生长性能和消化代谢方面展现了良好的应用前景。饲粮的变化通常会降低奶牛采食量,通过瘤胃微生物移植可以使奶牛的采食量在较短时间内得以恢复[37]。Ribeiro等[38]用野牛的瘤胃内容物交换70%的肉牛瘤胃内容物,结果发现,移植后肉牛的干物质采食量(DMI)显著升高,改变了肉牛瘤胃微生物的组成结构,改善了瘤胃发酵指标,提高了VFA含量,并提高了蛋白质消化率及氮利用率。供体瘤胃内容物中含有VFA及蛋白质等营养素,可能会对接受瘤胃微生物移植的受体有利。此外,有研究指出,刺激位于瘤胃和网胃壁的机械受体可促进反刍和瘤胃蠕动[39],瘤胃微生物移植可能会刺激胃壁机械受体,这值得在今后做进一步研究。

在其他方面,瘤胃微生物移植也表现出了可喜的效果。Ji等[40]以奶牛为试验动物,研究了瘤胃微生物移植对抗生素导致的瘤胃微生物紊乱的调控作用,发现灌注瘤胃液可以通过提高微生物多样性而改善抗生素导致的瘤胃微生物紊乱。围产期是奶牛非常重要的时期,在这一时期,采食量的降低导致无法满足机体对能量的需求,有学者尝试用瘤胃微生物移植解决这一问题。在奶牛产后第1、2和3天移植奶牛产后50 d的瘤胃液,研究发现,移植瘤胃液组奶牛的DMI显著升高,且瘤胃微生物群落结构接近供体奶牛[16],这一研究对解决围产期奶牛能量负平衡问题提供了新的思路。此外,有学者通过瘤胃微生物移植研究了真胃移位手术后的恢复治疗,发现移植组奶牛的采食量和产奶量显著高于对照组[41]。Leo-Penu等[42]分析了瘤胃微生物移植在缓解运输应激方面的作用,研究指出,瘤胃微生物移植可以增加运输后肉牛的采食量。通过以上研究可以看出,对瘤胃微生物区系的调控得到了很大的发展,尤其在瘤胃微生物移植方面取得了阶段性的成果。但是我们也要认识到,瘤胃微生物移植研究目前还处于起步阶段;此外,不同的个体在瘤胃微生物移植后展现出不同的群落重建模式,并且不同种属微生物对移植的贡献程度存在差异[43],如何针对这些微生物进行更加系统的研究,将对提高移植效率有着重要意义;同时,在瘤胃微生物移植过程中也应考虑供体和受体动物内环境的变化,避免影响移植效果。

4 小结

瘤胃微生物在营养物质消化和维持机体健康中起着重要的作用,同时受很多外界因素的影响,对瘤胃微生物区系进行有效地调控,可以更好地发挥动物的生产性能,维持动物健康。在瘤胃微生物移植取得阶段性成果的同时,应该加强对这些有效微生物体外培养的研究,争取早日将瘤胃微生物移植用于生产实践。同时,组学技术的进步将有助于我们研究瘤胃微生物区系中某一类别微生物的代谢及作用机制,这对瘤胃微生物区系的调控研究具有重要意义。
