菜籽粕是一种优良的蛋白质饲料资源,其脱毒利用受到广泛的关注。作为菜籽粕脱毒微生物之一,黑曲霉能有效降低菜籽粕中的植酸和硫甙含量。本研究旨在通过超声破碎技术从筛选到的1株具硫甙降解能力的黑曲霉(Aspergillas niger)H23中提取胞内芥子酶,建立微生物酶测定菜籽粕中硫甙含量的方法。此法首次在硫甙测定中使用微生物酶,操作过程简单且标准化。通过稳定性分析,证明该方法稳定可靠,其稳定性高于内源酶法,为硫甙测定方法的研究及微生物芥子酶的利用提供新思路。
Rapeseed meal is an excellent resource of vegetable protein, whose detoxication and application have acquired extensive attention. Aspergillas niger is one of the microbes to detoxicate rapeseed meal. It can decrease the contents of phytin acid and glucosinolate in rapeseed meal effectively. The aim of the study is to establish an analytical method for the determination of glucosinolate in rapeseed meal with myrosinase. In this study, the myrosinase is extracted from selected Aspergillus niger H23 with glucosinolate-decomposing function via ultrasonication. It is the first time that microbial enzymes are used in the quantitative determination of glucosinolate. The process is simple and standard. The stability analysis shows that the method is much more stable than that of the endogenous myrosinase method. The study provides a new concept of glucosinolate analysis and a new way for the application of microbial myrosinase.[Chinese Journal of Animal Nutrition, 2011, 23(6):1006 -1010]
