Effects of Dandelion Polysaccharide on Antioxidant Enzyme Activity and Related Gene Expression in Mice

Abstract:To investigate the effects of dandelion polysaccharide on antioxidant enzyme activity and related gene expression in mice, 48 specific pathogen free (SPF) ICR 5-week-old male mice were randomly divided into control group, high concentration dandelion polysaccharide group [600 mg/(kg·d)], medium concentration dandelion polysaccharide group [300 mg/(kg·d)] and low concentration dandelion polysaccharide group [150 mg/(kg·d)]. Dandelion polysaccharide group was given different concentrations of dandelion polysaccharide once a day by regular gavage, while the control group was given the same amount of normal saline for 4 weeks. The activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities, total antioxidant capacity (T-AOC) and malonaldehyde (MDA) content were determined in the serum and liver tissues of mice, and the relative mRNA expression levels of copper and zinc superoxide dismutase (Cu-ZnSOD), manganese superoxide dismutase (Mn-SOD), glutathione peroxidase-1 (GPX-1) and glutathione peroxidase-4 (GPX-4) in liver of mice were determined by qRT-PCR. The results showed that after gavaged with dandelion polysaccharide, the activities of GSH-Px, SOD and T-AOC in mice serum and liver increased significantly (P<0.05), the content of MDA reduced significantly (P<0.05), and the mRNA relative expression levels of Cu-ZnSOD, Mn-SOD, GPX-1 and GPX-4 were up-regulated significantly (P<0.05). Dandelion flavonoids can significantly improve the antioxidant capacity of mice and have anti-aging effects.
