Abstract

An 8-week feeding experiment was conducted to investigate the effects of dietary carbohydrate to lipid ratio on growth performance, hepatopancreas metabolic enzyme activities and expression of fatty acid transport-and Toll-like receptor pathway-related genes of oriental river prawn (Macrobrachium nipponense). The carbohydrate to lipid ratio in 5 isoenergetic (crude protein content was 39%) experimental diets was 1.12 (CL1), 2.00 (CL2), 3.56 (CL3), 7.10 (CL4) and 24.12 (CL5), respectively. Oriental river prawns with the average body weight of (0.104±0.003) g were divided into 5 groups with 5 replicates in each group and 50 prawns in each replicate. The results showed that no significant differences were observed in weight gain rate when carbohydrate to lipid ratio increased from 1.12 to 7.10 (P>0.05), however, when the carbohydrate to lipid ratio continually increased to 24.12, the weight gain rate of prawns was significantly increased (P<0.05), and no significant difference was observed in survival rate among all groups (P>0.05). The hepatopancreas glycogen content in CL5 group was significantly higher than that in CL4 group (P<0.05). The highest activities of hexokinase (HK) and pyruvate kinase (PK) in hepatopancreas were observed in CL4 group, the HK activity of prawns in CL4 group was significantly higher than that in CL1 group (P<0.05), and the PK activity of prawns in CL4 group was significantly higher than that in other groups (P<0.05). No significant differences were observed in hepatopancreas malondialdehyde (MDA) content, total antioxidant capacity (T-AOC) and catalase (CAT) activity among all groups (P>0.05), but the hepatopancreas superoxide dismutase (SOD) activity in CL1 and CL2 groups was significantly higher than that in CL4 and CL5 group (P<0.05). In CL1 group, the expression levels of scavenger recepter class B type Ⅰ (SR-BⅠ) and fatty acid transport protein 4 (FATP4) genes in hepatopancreas were significantly higher than those in other groups (P<0.05). The expression level of hepatopancreas fatty acid-binging protein 10 (FABP10) gene in the higher carbohydrate to lipid ratio group (CL4 group) was significantly higher than that in the lower carbohydrate to lipid ratio groups (CL1, CL2 and CL3 groups) (P<0.05). The expression levels of hepatopancreas Toll-like receptor 3 (TLR3), myeloid differentiation primary response protein 88 (MyD88), interleukin 1 receptor associated kinase 4 (IRAK4), tumor necrosis factor receptor associated factor 6 (TRAF6), mitogen-activated protein kinase kinase kinase 7 (MAP3K7) and mitogen-activated protein kinase 14 (MAPK14) genes in CL3 group were significantly higher than those in other groups (P<0.05). Therefore, oriental river prawn has a great adaptability for carbohydrate to lipid ratio, but too high dietary carbohydrate to lipid ratio (24.12) will depress the growth and increase the glycogen storage in hepatopancreas. Dietary carbohydrate to lipid ratio can modulate the expression of fatty acid transport-and Toll-like receptor pathway-related genes in hepatopancreas of oriental river prawn.
