空怀绵羊粪便微生物和血液代谢组特征分析

摘要

本试验旨在研究空怀绵羊的粪便微生物和血液代谢组特征,探索断奶后自然交配群体中母羊妊娠失败的可能原因。绵羊处于相同饲养管理条件下,通过B超检测妊娠与否,将绵羊分为妊娠组和空怀组(n=6),分别采集粪便和血液样本,经过Illumina MiSeq测序和液相色谱串联质谱(LC-MS/MS)分析微生物多样性和血清差异代谢物。结果表明:1)与妊娠组相比,空怀组Shannon指数显著降低(P<0.05);主坐标分析(PCoA)和相似性分析(ANOSIM)结果表明,在门和属水平,妊娠组和空怀组粪便微生物组成存在显著差异(P<0.05);与妊娠组相比,空怀组粪便放线菌门(Actinobacteriota)相对丰度显著下调(P<0.05),粪便脱硫弧菌属(Desulfovibrio)、Turicibacter相对丰度显著上调(P<0.05),粪便瘤胃梭菌属(Ruminiclostridium)、棒状杆菌属(Corynebacterium)相对丰度显著下调(P<0.05)。2)妊娠组与空怀组共发现78种血清差异代谢物[投影变量重要性(VIP)值>1、P<0.05],其中空怀组血清LysoPC (O-18:0)含量显著高于妊娠组(P<0.05),血清脱氧肌苷、berteroin和C16-鞘氨醇含量显著低于妊娠组(P<0.05);差异代谢物显著富集于11条信号通路(P<0.05)。3)普氏分析显示妊娠组与空怀组的微生物组学相对丰度和代谢组学表达量趋势显著一致(P<0.05),微生物与代谢物相关性分析发现脱硫弧菌属相对丰度与3a,7a,12b-三羟基-5b-胆烷酸含量呈显著正相关(P<0.05)。综上所述,与妊娠绵羊相比,空怀绵羊粪便微生物和血清代谢物组成呈现明显差异,肠道菌群与血清代谢物的相互作用能够影响母羊的妊娠生理状态。

Abstract

This study aimed to investigate the fecal microbiota and blood metabolome characteristics of non-pregnant ewes, to explore the possible reasons for pregnancy failure in natural mating flocks of ewes after weaning. The ewes were fed and managed under the same condition and divided into pregnant group and non-pregnant group by B ultrasound detection. The fecal and blood samples were collected, and the fecal microbial diversity and differential metabolites were analyzed by Illumina MiSeq sequencing and liquid chromatography-tandem mass spectrometry (LC-MS/MS), respectively. The results showed as follows:1) compared with the pregnant group, the Shannon index of non-pregnant group was significantly decreased (P<0.05); the principal co-ordinates analysis (PCoA) and analysis of similarities (ANOSIM) showed that at the phylum or genus levels, the fecal microbial composition had significant difference between pregnant group and non-pregnant group (P<0.05); compared with the pregnant group, the fecal Actinobacteria relative abundance of non-pregnant group was significantly decreased (P<0.05), the relative abundances of Desulfovibrio and Turicibacter in feces were significantly increased (P<0.05), and the relative abundances of Ruminiclostridium and Corynebacterium in feces were significantly decreased (P<0.05). 2) A total of 78 serum differential metabolites were found in pregnant group and non-pregnant group[variable importance in projection (VIP)>1, P<0.05], the serum LysoPC(O-18:0) content of non-pregnant group was significantly higher than that of pregnant group (P<0.05), and the contents of Deoxyinosine, berteroin and C16-sphingosine in serum were significantly lower than that of pregnant group (P<0.05). Differential metabolites were significantly enriched in 11 signal pathways (P<0.05). 3) Procrustes analysis showed that the trend of microbiological relative abundance was consistent with metabolome expression in pregnant group and non-pregnant group (P<0.05), and the correlation analysis between microorganisms and metabolites showed that Desulfovibrio relative abundance was positively correlated with 3a,7a,12b-trihydroxy-5b-cholanoic acid content (P<0.05). To sum up, compared with pregnant ewes, the composition of fecal microbial and serum metabolites of non-pregnant ewes have significant difference, the interaction of intestinal flora and serum metabolites can affect the physiological state of pregnancy ewes.
