Interventional Effect of Eucommia ulmoides Flavones on Growth Inhibition, Oxidative Stress Injury and Inflammatory Injury of Weaned Piglets Induced by Deoxynivalenol

摘要

本试验旨在探究杜仲黄酮(EUF)对呕吐毒素(DON)诱导的断奶仔猪生长抑制、氧化应激损伤与炎性损伤的干预效应。选择24头初始体重为(6.50±0.28) kg的健康三元杂交(杜×长×大)断奶仔猪,随机分为3组,分别为对照组(Control组,基础饲粮)、DON组(饲粮中含1.5 mg/kg DON)、DON+EUF组(饲粮中含1.5 mg/kg DON和100 mg/kg EUF),每组8头,每头为1个重复。预试期7 d,正试期21 d。结果表明:1)与Control组相比,DON组的终末体重、平均日增重(ADG)和平均日采食量(ADFI)显著降低(P<0.05),F/G显著升高;DON+EUF组的ADG、ADFI和F/G与Control组无显著差异(P>0.05),但其ADG显著高于DON组(P<0.05),F/G显著低于DON组(P<0.05)。与Control组相比,DON组的肝脏和脾脏相对指数显著升高(P<0.05)。2)与Control组相比,DON组的血清碱性磷酸酶(ALP)、谷丙转氨酶(ALT)和谷草转氨酶(AST)活性显著提高(P<0.05);DON+EUF组血清ALP活性显著低于DON组(P<0.05)。3)与Control组相比,DON组血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和总抗氧化能力(T-AOC)显著降低(P<0.05),血清丙二醛(MDA)和活性氧(ROS)含量显著提高(P<0.05);与DON组相比,DON+EUF组血清SOD和GSH-Px活性显著提高(P<0.05),血清MDA和ROS含量显著降低(P<0.05)。与Control组相比,DON组血清白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量显著提高(P<0.05),DON+EUF组血清IL-6、IL-1β和TNF-α含量则较DON组显著降低(P<0.05)。综上所述,饲粮中添加EUF对于缓解DON诱导的断奶仔猪生长抑制与肝脏、脾脏病变,改善机体抗氧化能力与抗炎能力具有一定效果。

Abstract

This experiment was conducted to explore the intervention effect of Eucommia ulmoides flavones (EUF) on growth inhibition, oxidative stress injury and inflammatory injury of weaned piglets induced by deoxynivalenol (DON). Twenty-four healthy three crossbred (Duroc×Landrace×Yorkshire) weaned piglets with an initial body weight of (6.50±0.28) kg were randomly divided into 3 groups, they were control group (basal diet), DON group (diet contained 1.5 mg/kg DON) and DON+EUF group (diet contained 1.5 mg/kg DON and 100 mg/kg EUF) with 8 piglets in each group, each of which was a repeat. The formal experiment lasted for 21 days after 7 days adaptation. The results showed as follows:1) compared with the control group, the final body weight, average daily gain (ADG) and average daily feed intake (ADFI) of piglets in the DON group were significantly decreased (P<0.05), and the feed/gain ratio (F/G) was significantly increased (P<0.05); there were no significant differences in ADG, ADFI and F/G between the DON+EUF group and control group (P>0.05). The ADG of piglets in the DON+EUF group was significantly higher than that in the DON group (P<0.05), but the F/G was significantly lower than that in the DON group (P<0.05). Compared with the control group, the relative indexes of liver and spleen in the DON group was significantly increased (P<0.05). 2) Compared with the control group, the activities of serum alkaline phosphatase (ALP), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in the DON group were significantly increased (P<0.05). The activity of serum ALP in the DON+EUF group as significantly lower than that in the DON group (P<0.05). 3) Compared with control group, the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC) in the DON group were significantly decreased (P<0.05), and the contents of malondialdehyde (MDA) and reactive oxygen species (ROS) were significantly increased (P<0.05). Compared with the DON group, the activities of serum SOD and GSH-Px in the DON+EUF group were significantly increased (P<0.05), and the contents of MDA and ROS were significantly decreased (P<0.05). Compared with the control group, the contents of serum interleukin-6 (IL-6), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in the DON group were significantly increased. The contents of IL-6, IL-1β and TNF-α in serum in the DON+EUF group were significantly reduced compared with the DON group (P<0.05). In conclusion, dietary supplementation with EUF has a certain effect on alleviating growth inhibition, liver and spleen lesions, and improving antioxidant capacity and anti-inflammatory capacity of weaned piglets induced by DON.
