A Detection Method Based on Ion Exclusion Chromatography for Simultaneous Determination of Lactic Acid and Seven Short-Chain Fatty Acids

摘要

本研究介绍了一种基于离子排斥色谱原理同时测定乳酸和7种短链脂肪酸的检测方法,并以肠道内容物和粪便为基质,对该方法进行方法学评价。结果表明: 本试验方法测定的8种有机酸含量线性范围为1~500 μg/mL时,其线性相关系数均>0.999,方法检出限为0.02~0.15 g/kg;在添加不同含量(1、4和10 g/kg)的8种有机酸标准品条件下,方法回收率为80.28%~116.99%,相对标准偏差均<10%。本方法前处理简单,可同时检测与动物肠道相关的多种关键有机酸含量,为短链脂肪酸和乳酸的检测提供了一种新的方法参考。

Abstract

In this study, a detection method for simultaneous determination of lactic acid and seven short-chain fatty acids based on the principle of ion exclusion chromatography was introduced, and the method was evaluated using intestinal contents and feces as substrates. The results showed that when the linear range of the contents of eight organic acids was from 1 to 500 μg/mL, the linear correlation coefficients were all greater than 0.999, and the detection limit was from 0.02 to 0.15 g/kg. The recovery rates ranged from 80.28% to 116.99%, and the relative standard deviations were all smaller than 10%, when eight organic acid standard substances with different contents (1, 4 and 10 g/kg) were added. This preprocessing of this method is simple and can simultaneously detect the contents of many key organic acids related to animal gut, providing a new method for the detection of short-chain fatty acids and lactic acid.

短链脂肪酸(short chain fatty acids,SCFAs)又称挥发性脂肪酸(volatile fatty acids,VFAs),是由肠道微生物发酵不易消化的膳食纤维等碳水化合物产生的代谢产物,指碳链长度为1~6个碳原子的有机饱和羧酸,包括甲酸、乙酸、丙酸、丁酸、戊酸、己酸及其同分异构体,是肠道和宿主相互作用的重要介质,已被证实参与肠道健康、代谢调控以及神经调控[1⇓⇓-4]等生理活动,近年来成为医药、食品和畜牧等领域的关注热点之一。乳酸是动物体内糖代谢的中间产物,是体内糖异生的主要前体和能量底物,也是重要的信号分子,已被证实在糖代谢和炎症调控过程中发挥重要作用[5-6],在乳品行业和发酵领域有广泛的应用,在类别上虽不属SCFAs之列,但在肠道健康研究方面也被认为是重要代谢活动的表征指标。
SCFAs和乳酸是表征动物代谢调控和肠道健康等生理状态的重要指标,其简单、高效的检测技术作为评估物质含量的底层技术不可或缺。关于SCFAs和乳酸检测方法的研究比较多[7⇓⇓-10],截至2024年3月6日,仅在中国知网(CNKI)数据库以“短链脂肪酸”或“挥发酸”、“检测”为关键词,可检索到文献89篇,其中近10年文献占比85%以上,这可能与近年来肠道微生物和动物营养健康引起广泛关注有关;以“乳酸”、“检测”为关键词,可检索到文献373篇,其中近十年文献占比70%以上,这一方面是由于乳酸及其衍生物是医学血液检测中重要的检测指标之一,其检测方法一直在不断更新迭代;另一方面是由于近年来关于乳酸的作用不仅定义在传统意义上疾病状态的表征指标,其在肠道健康方面的“代谢因子”作用被越来越多的研究证实[11-12]。现有的方法学工作分别研究SCFAs或乳酸的报道较多,将两类有机酸融合到一种检测方法里的工作未见报道。此外,现有报道的SCFAs分析方法前处理方法多采用衍生化技术或针对某类样品特定复杂的前处理技术,实验时间及经济成本较高。在当前肠道微生物受到越来越多关注的大背景下,建立适用范围广、前处理简单、灵敏度适宜以及可以同时检测SCFAs和乳酸的检测方法有利于高效支撑多个方向的科学研究。本研究建立了一种基于离子排斥色谱同时测定乳酸、甲酸、乙酸、丙酸、异丁酸、丁酸、戊酸和异戊酸(以下简称“8种有机酸”)的检测方法,该方法可定量检测饲料和动物样本中的8种有机酸含量,近年来在动物营养领域中多个方向的科研工作中均有利用[13⇓⇓⇓-17],为相关科学研究工作提供了有效的技术支撑。

1 材料与方法

1.1 试验材料

1.1.1 试验原料

粪便样品来源于中国农业科学院饲料研究所宠物营养团队南口试验基地猫舍试验用猫;肠道内容物来源于中国农业科学院饲料研究所反刍动物养殖基地试验用牛。

1.1.2 试验试剂

标准品:乳酸、甲酸、乙酸、丙酸、丁酸、异丁酸、戊酸和异戊酸(Sigma,纯度≥99%)。化学试剂:0.50 mol/L ZnSO4·7H2O溶液(国药集团化学试剂有限公司),0.25 mol/L K4Fe(CN)6·3H2O溶液(西陇化工股份有限公司),0.50 mmol/L H2SO4(国药集团化学试剂有限公司),100 mmol/L LiCl溶液(广州市金华大化学试剂有限公司),均为分析纯。

1.2 试验仪器与设备

940-Professional IC离子色谱仪(瑞士Metrohm公司),Milli-Q去离子水发生器(美国Millipore公司),IEC CL-31R 离心机(美国Thermo Electron Corporation公司),VX-Ⅲ振荡器[安简(北京)科技有限公司]。

1.3 试验方法

1.3.1 试验设计

从方法学角度评价8种有机酸在本方法下的样品检出限、定量限和线性范围;以代表性样本动物粪便和肠道内容物为基质,评价8种有机酸在两类代表性动物样本中的添加回收率、精密度和相对标准偏差指标。

1.3.2 标准溶液配制

精确称取8种有机酸于容量瓶中用超纯水稀释定容,标准曲线含量范围为1~500μg/mL,过0.22 μm微孔过滤膜,采用外标法建立标准曲线。

1.3.3 前处理方法

准确称取均质的样品0.25 g于带刻度的离心管中,加入约3 mL水浸润样品,振荡提取5 min,先加入0.50 mol/L ZnSO4·7H2O溶液200 μL摇匀,通过沉淀除去提取液中蛋白多糖等大分子水溶物;再加入0.25mol/L K4Fe(CN)6·3H2O溶液100 μL摇匀,用于沉淀体系中引入的锌离子(Zn2+)等杂质;加水定容至5 mL,于9 400×g离心10 min,去沉淀、取上清液通过0.22 μm微孔过滤膜,取1 mL滤液于10 mL小试管中,加入4 mL水,摇匀,待上机。

1.3.4 色谱条件

色谱柱为Metrosep Organic Acids-250/7.8(基于聚合物复合材料的离子交换柱);抑制器为MSM抑制器;流动相为0.50 mmol/L H2SO4溶液,等度洗脱,流速为0.60 mL/min;再生液为100 mmol/L LiCl溶液;柱箱温度为30 ℃;上机体积为20 μL。

1.4 数据统计分析

采用仪器自带MagIC Net3.1软件进行色谱峰积分以及数据分析。

2 结果与分析

2.1 8种有机酸检出限、定量限、标准曲线相关性及线性范围

由图1和表1可知,8种有机酸含量的线性范围为1~500 μg/mL时,线性相关系数>0.999;以各色谱峰的3倍信噪比计算检出限,3倍检出限为定量限,本试验中8种有机酸的方法检出限为0.02~0.15 g/kg,方法定量限为0.05~0.45 g/kg。

2.2 回收率和精密度

粪便和肠道内容物中自然存在游离乳酸和SCFAs。在已知8种有机酸含量的样品中添加不同含量的乳酸和SCFAs,每个添加含量设5个重复(图2和图3),计算相对标准偏差和添加回收率,试验结果见表2和表3。肠道内容物和粪便中添加不同含量8种有机酸,回收率为80.28%~116.99%,相对标准偏差均<10%,可以满足常规动物样品检测需求。肠道内容物和粪便中8种有机酸色谱图分别见图4和图5。

3 讨论

本试验建立了一种基于离子排斥色谱原理同时测定乳酸和7种短链脂肪酸的检测方法,方法检出限为0.02~0.15 g/kg,仪器检出限为3.27~7.25 μmol/L,回收率为80.28%~116.99%。本试验方法与部分其他方法[18⇓⇓-21]指标对比见表4。

由于SCFAs具挥发性,气相色谱法和气相色谱-质谱法是众多分析方法中[7,22]相对较为常用的检测手段。本试验方法与常用的气相色谱法和气相色谱-质谱法等比较,有以下几个特点:1)前处理简单。本方法前处理用水提取,简单除杂大分子后上机,无需经过衍生化过程,前处理方面时间成本、试剂成本较低。2)柱分离阶段不同。本方法色谱柱分离采用离子排斥色谱原理,根据Donnon膜排斥效应,电离组分受排斥不被保留,而弱酸则一定保留,本方法检测到的是低电离度的弱酸分子,而不是衍生化后的脂肪酸甲酯,相对柱耗较低。3)本方法检测的有机酸种类较为丰富,在其他可见报道的文献基础上,可检测的目标范围增加了乳酸和甲酸这2种有机酸,可供关注这2种有机酸的研究使用。4)本方法在肠道内容物和粪便2种基质中的回收率在可接受的范围内,说明在从前处理到柱分离及检测的整体测试流程中,目标有机酸逸失率及假阳性率均在可接受的范围内,整个方法可为其他有机酸检测提供参考。同时,本方法灵敏度没有上述气相色谱法和气相色谱-质谱法的灵敏度高,说明适用于原样有机酸含量较高的样品。研究者可据样品不同情况、目标物质含量范围及试验条件选择合适的检测方法。

4 结论

本研究建立了一种基于离子排斥色谱原理同时测定8种有机酸的检测方法。与常见的气相色谱法和气相色谱-质谱法相比,本方法1次上样可同时检测8种有机酸,前处理较为简单,检出限、定量限和回收率等在一定范围内可满足检测需求,适用于饲料、血液和瘤胃液等类型样品中乳酸和SCFAs的测定,是一种成本较低、前处理简单以及检测效率较高的检测方法,为关注肠道健康与机体关系的医学、食品和畜牧等领域的研究提供了一种技术选择和参考。
