生长猪小麦氨基酸消化率测定及预测模型建立

摘要

本试验旨在测定生长猪对小麦的氨基酸表观回肠消化率(AIDAA)和氨基酸标准回肠消化率(SIDAA),并基于小麦中有效化学成分构建SIDAA的预测模型。选取22头体况相近、健康的生长猪[“杜×长×大”,(30.5±2.2) kg],每头猪在回肠末端安装1个“T”型瘘管。按照2个“11×3”的不完全拉丁方设计,分别饲喂10个以小麦为唯一氮源的饲粮和1个无氮饲粮,所有试验饲粮中均添加0.30%的二氧化钛作为指示剂。本试验共包含3个连续的周期,每期有5 d的预试期和2 d的回肠食糜收集期,采用无氮饲粮法测定回肠末端内源氨基酸损失。结果显示: 饲喂基础下,10个不同来源小麦的粗蛋白质(CP)、赖氨酸(Lys)、蛋氨酸(Met)、色氨酸(Trp)和苏氨酸(Thr)的标准回肠消化率(SIDCP、SIDLys、SIDMet、SIDTrp、SIDThr)的平均值分别为68.53%、71.68%、77.77%、86.46%和74.20%。小麦SIDLys与中性洗涤纤维(NDF)呈极显著负相关(P<0.01),SIDMet和SIDThr均与干物质(DM)呈显著正相关(P<0.05),SIDTrp与酸性洗涤纤维(ADF)呈极显著负相关(P<0.01)。SIDLys、SIDMet、SIDTrp、SIDThr最佳预测方程分别为SIDLys=113.111-2.804×NDF[剩余标准差(RSD)=1.84,决定系数(R2)=0.873,P<0.01]、SIDMet=-1 054.812+12.959×DM(RSD=5.90,R2=0.524,P<0.05)、SIDTrp=118.828-10.603×ADF(RSD=2.48,R2=0.799,P<0.01)、SIDThr=-681.144+8.643×DM(RSD=4.86,R2=0.419,P<0.05)。综上可知,不同来源小麦的化学成分含量和氨基酸消化率变异较大,DM是SIDMet和SIDThr的最佳预测因子,NDF是SIDLys的最佳预测因子,ADF是SIDTrp的最佳预测因子。

Abstract

In this experiment, the apparent and standardized ileal digestibility of amino acids (AIDAA and SIDAA) of wheat fed to growing pigs were determined, and prediction models of SIDAA was established based on the effective chemical components of wheat. Twenty-two healthy growing pigs with similar body condition [Drouc×Landrace×Yorkshire, (30.5±2.2) kg] were selected, and a T-cannula was installed in the distal ileum of each pig. According to two “11×3” incomplete Latin square design, the pigs were fed 10 diets with wheat as the sole nitrogen source and one diet without nitrogen, respectively, and all diets were supplemented with 0.30% titanium dioxide as an indicator. The experiment had 3 consecutive periods, each period had 5 d of adaptation period and 2 d of ileal digesta collection period. The loss of endogenous amino acids at the end of ileum was measured by nitrogen-free diet. The results showed as follows: on as-fed basis, the average values of standardized ileal digestibility of crude protein (CP), lysine (Lys), methionine (Met), tryptophan (Trp) and threonine (Thr) (SIDCP, SIDLys, SIDMet, SIDTrp and SIDThr) in wheat from 10 different sources were 68.53%, 71.68%, 77.77%, 86.46% and 74.20%, respectively. SIDLys was extremely significantly negatively correlated with neutral detergent fiber (NDF) (P<0.01), both SIDMet and SIDThr were significantly positively correlated with dry matter (DM) (P<0.05), and SIDTrp was extremely significantly negatively correlated with acid detergent fiber (ADF) (P<0.05). The optimal prediction equations for SIDLys, SIDMet, SIDTrp and SIDThr were: SIDLys=113.111-2.804×NDF [residual standard deviation (RSD)=1.84, coefficient of determination (R2)=0.873, P<0.01], SIDMet=-1 054.812+12.959×DM (RSD=5.90, R2=0.524, P<0.05), SIDTrp=118.828-10.603×ADF (RSD=2.48, R2=0.799, P<0.01), SIDThr=-681.144+8.643×DM (RSD=4.86, R2=0.419, P<0.05). In conclusion, the chemical chemical component contents and amino acid digestibility of wheat from different sources varied greatly. DM is the best predictor of SIDMet and SIDThr, NDF is the best predictor of SIDLys, and ADF is the best predictor of SIDTrp.

随着我国畜禽饲料行业的不断发展和壮大,玉米-豆粕型饲料配方长期占据主导地位。然而,近年来受多种因素影响,特别是玉米价格长期高位运行,导致饲料成本不断增加,给养殖业带来了较大压力。为此,寻找合理的替代饲料原料,特别是非玉米型能量饲料原料,已成为当前畜禽饲料行业的紧迫课题。小麦作为我国重要的粮食作物之一,具有高营养价值、适应性强、高产性、耐储藏性和多功能性等特点,在全球范围内得到广泛种植。随着我国粮食需求的增加,小麦产业迅速发展,年产量和进口量也大幅增长。刘悦等[1]报道,我国小麦种植面积约占全球的11%,产量约占全球的18%,全球排名第4,仅次于欧盟。小麦的有效能值低于玉米,且含有一定量的抗营养因子[2],然而,小麦的粗蛋白质(crude protein,CP)含量较玉米高,必需氨基酸(essential amino acid,EAA)含量也更为丰富[3]。研究表明,小麦部分或完全替代玉米的饲喂效果与玉米相当[4]。这些优点使得小麦成为一种理想的非玉米型能量饲料原料,在猪饲粮中若能实现合理配比,可以降低对玉米的依赖程度,减轻国际贸易争端带来的冲击,从而稳定养殖业的发展。
然而,小麦化学成分含量和氨基酸(amino acids,AA)消化率数值在各国饲料原料数据库中差异较大,且受生长环境、气候条件、品种和加工工艺的影响,小麦的营养价值存在高变异性[5⇓-7]。目前,国内外对不同来源小麦的氨基酸表观回肠消化率(apparent ileal digestibility of amino acids,AIDAA)与氨基酸标准回肠消化率(standardized ileal digestibility of amino acids,SIDAA)的研究甚少,仅依靠现有文献和数据库中有关AA消化率的静态值无法满足精准配方的要求。因此,测定小麦AA消化率等重要营养参数,对进一步完善国内的饲料原料数据库具有重要意义。本试验旨在测定生长猪对10个不同来源小麦的AIDAA和SIDAA,并基于小麦中的有效化学成分构建生长猪对小麦SIDAA的预测方程。

1 材料与方法

1.1 伦理审查

本试验方案均获得了中国科学院亚热带农业生态研究所动物护理和使用委员会的审查和批准(IACUC#201302)。

1.2 样品采集

选择来自安徽、江苏、河南、湖南和湖北的10个小麦(样品编号为XM1~10,详细信息见文献[8]),小麦原料在饲料厂粉碎后用于配制试验饲粮,采用四分法收集小麦原料与试验饲粮,存于-20 ℃冷库以待后续的化学分析。

1.3 试验设计

选取健康、体重为(30.5±2.2) kg的“杜×长×大”阉猪共22头,经手术在回肠末端分别安装1个“T”型瘘管,采取2个“11×3”的重复不完全拉丁方设计,分别饲喂1个无氮饲粮和10个小麦试验饲粮(RXM1~10,表1)。在10个小麦饲粮中,分别以10个不同来源的小麦作为饲粮中唯一的蛋白质来源,加入玉米淀粉配制成一种半纯合饲粮,而无氮饲粮则以玉米淀粉为主。在所有试验饲粮中均添加0.30%的二氧化钛(TiO2)作为外源性指示剂,用于计算生长猪对小麦的粗蛋白质表观回肠消化率(apparent ileal digestibility of crude protein,AIDCP)和粗蛋白质标准回肠消化率(standardized ileal digestibility of crude protein,SIDCP)以及AIDAA和SIDAA。试验共包含3个周期,每期7 d(5 d的预试期+2 d的回肠食糜收集期),试验结束后每个处理有6个重复。

预混料为每千克饲粮提供 Premix provided the following per kilogram of diets:VA 4 200 IU,VD3 400 IU,VE 36 IU,VK3 1.2 mg,VB12 23 μg,VB2 5.63 mg,VB5 20.5 mg,VB3 28 mg,氯化胆碱 choline chloride 1.00 g,叶酸 folic acid 0.8 mg,VB1 3.4 mg,VB6 2.7 mg,VH 0.18 mg, Mn (as manganese sulfate) 40.0 mg,Fe (as ferrous sulfate) 70.0 mg,Zn (as zinc sulfate) 70.0 mg,Cu (as copper sulfate) 70 mg,I (as potassium iodide) 0.3 mg,Se (as sodium selenite) 0.3 mg。

1.4 饲养管理

试验猪单独饲养在代谢笼(1.4 m×0.7 m×0.5 m)内,代谢室内的温度被控制在(23±1) ℃。每个周期开始前,对试验猪称重,按照猪平均体重的4%确定每日投喂量,并平均分成2份,分别于08:00和15:00饲喂给猪,猪全程可自由饮水。在食糜收集期按照猪代谢试验操作规程进行样品收集、动物护理和清洁卫生等工作。

1.5 食糜收集与处理

在每个试验周期的第6~7天,采集猪回肠末端的食糜,采样时间从当天08:00开始,持续到17:00结束。使用预先编号的自封袋套在猪瘘管上,当食糜收集量达到1/3时更换袋子,收集的食糜立即转移至-20 ℃冷库冻存。食糜样品放入冷冻干燥机(SCIENTZ-50F/A,宁波新芝冻干设备股份有限公司)中冻干72 h,冻干后的食糜经粉碎并过40目筛后存入-20 ℃冰箱中待测。

1.6 指标测定

1.6.1 样品的化学分析

参照GB/T 18246—2019[9]中的普通酸水解法测定小麦原料、饲粮和食糜中除含硫AA和色氨酸(tryptophan,Trp)外的14种AA的含量;参照GB/T 18246—2019[9]中的氧化酸水解法测定样品中半胱氨酸(cysteine,Cys)和蛋氨酸(methionine,Met)等含硫AA的含量。样本中除Trp外的16种AA先使用邻苯二甲醛(o-phthalaldehyde,OPA)和9-芴基甲基氯甲酸酯(9-fluorenylmethyl chloroformate,FMOC)进行柱前衍生化后,再经高效液相色谱仪(1100 HPLC,美国安捷伦科技公司)进行定性与定量分析。参照GB/T 15400—2018[10]中的分光光度法测定样品中Trp的含量;参照GB/T 21912—2008[11]测定小麦饲粮、食糜中TiO2的含量。分别参照GB/T 6435—2006、GB/T 6432—2018、GB/T 6433—2006、GB/T 6438—2007、GB/T 6436—2018、GB/T 6437—2018、GB/T 20806—2006、NY/T 1459—2007和GB/T 6434—2006测定小麦原料与饲粮中的干物质(dry matter,DM)、CP、粗脂肪(ether extract,EE)、粗灰分(crude ash,Ash)、钙(calcium,Ca)、总磷(total phosphorus,TP)、中性洗涤纤维(neutral detergent fiber,NDF)、酸性洗涤纤维(acid detergent fiber,ADF)、粗纤维(crude fiber,CF)含量;使用Megazyme生产的淀粉总量检测试剂盒(威克洛公司,爱尔兰)测定小麦原料与饲粮中的总淀粉(total starch,TS)含量;利用5E-AC8018等温式全自动量热仪(长沙凯德测控仪器有限公司)测定小麦原料与饲粮中总能(gross energy,GE);使用植物木聚糖(Xylans)酶联免疫吸附测定(ELISA)检测试剂盒(凡科维生物科技有限公司,中国)测定小麦原料中Xylans的含量。

1.6.2 生长猪对小麦AIDAA与SIDAA、AIDCP与SIDCP的测定

采用指示剂法测定生长猪对小麦的AIDAA与SIDAA,计算公式[12-13]如下:
AIDAA(%)=[1-(AAi×Td)/(AAd×Ti)]×100。
式中:AAi和Ti分别是回肠食糜中AA和TiO2的含量;AAd和Td分别是小麦饲粮中AA和TiO2的含量。
使用相同的公式计算AIDCP 。
回肠末端内源性AA损失(IAAend,%)=[AAi×(Td/Ti)]。
式中:AAi和Ti分别是饲喂无氮饲粮的猪的回肠食糜中AA和TiO2的含量;Td是无氮饲粮中TiO2的含量
使用相同的公式计算回肠末端内源性CP损失。
SIDAA(%)=AIDAA+(IAAend/AAd)×100。
使用相同的公式计算SIDCP。

1.7 数据统计与分析

通过Excel 2010初步统计整理测得的数据,使用SPSS 20.0统计分析软件中的ANOVA 过程进行单因素方差分析,单因素方差分析显著时,用Duncan氏法对组间数据进行多重比较,以P<0.05作为差异显著性的判断标准;采用SPSS 20.0软件中的Correlate 过程对10种小麦原料的CP、DM、Ash、EE、Ca、TP、CF、NDF、ADF、TS、赖氨酸(lysine,Lys)、Met、苏氨酸(threonine,Thr)、Trp含量与Lys、Met、Thr、Trp标准回肠消化率(SIDLys、SIDMet、SIDLys、SIDTrp)之间的相关性进行分析,用Regression过程进行逐步回归分析,建立基于小麦原料有效化学成分的SIDLys、SIDMet、SIDLys、SIDTrp最佳预测方程。

2 结果与分析

2.1 小麦的化学成分及AA含量

饲喂基础下,10个小麦的DM平均含量为87.40%,GE为15.48 MJ/kg,CP为13.34%,EE为1.94 %,Ash为1.83%,CF为1.56%,NDF为14.78%,ADF为3.05%,Ca为0.05%,TP为0.31%,Xylans为6.44%,TS为55.19%。小麦原料化学成分含量数据详见刘小杰等[8]的报道。
由表2可知,饲喂基础下,10个小麦EAA中Arg平均含量为0.55%(0.48%~0.65%),His为0.35%(0.30%~0.45%),Ile为0.43%(0.35%~0.53%),Leu为0.85%(0.71%~1.03%),Lys为0.45%(0.38%~0.59%),Met为0.33%(0.23%~0.45%),Phe为0.60%(0.48%~0.71%),Thr为0.34%(0.29%~0.41%),Trp为0.10%(0.08%~0.11%),Val为0.52%(0.45%~0.63%);10个小麦必需氨基酸(non-essential amino acid,NEAA)中His、Ile、Lys、Met、Phe、Trp含量的变异系数(CV)均超过10%。

2.2 小麦饲粮的化学成分与AA含量

由表3可知,饲喂基础下,10个小麦饲粮中DM平均含量为89.66%(89.50%~89.90%),CP为4.91%(4.04%~6.28%),Arg为0.25%(0.19%~0.32%),His为0.16%(0.13%~0.23%),Ile为0.18%(0.15%~0.24%),Leu为0.37%(0.31%~0.46%),Lys为0.19%(0.15%~0.28%),Met为0.06%(0.03%~0.10%),Phe为0.26%(0.21%~0.33%),Thr为0.15%(0.12%~0.21%),Trp为0.06%(0.05%~0.07%),Val为0.22%(0.18%~0.28%);10个小麦饲粮的EAA中Arg、His、Ile、Leu、Lys、Met、Phe、Thr、Trp、Val含量的CV均大于10%。

由表4可知,除甘氨酸(glycine,Gly)外,生长猪对10个不同来源小麦的AIDCP和大多数AIDAA的差异显著(P<0.05)或极显著(P<0.01);10个小麦AIDCP的平均值为30.93%(21.31%~52.27%);10个小麦的EAA中AIDLys的平均值为33.22%(25.70%~43.62%),AIDIle为60.74%(51.85%~65.68%),AIDLeu为69.57%(65.28%~73.00%),AIDMet为58.98%(40.07%~77.21%),AIDTrp为66.37%(61.92%~77.44%),AIDThr为29.57%(16.83%~47.83%),AIDPhe为74.23%(70.16%~78.06%),AIDHis为50.96%(38.01%~63.14%),AIDArg为24.19%(18.95%~34.88%),AIDVal为54.04%(47.72%~60.90%);10个小麦的EAA中AIDLys、AIDMet、AIDThr、AIDHis、AIDArg及AIDCP的CV均大于10%。

2.4 生长猪对小麦的SIDCP与SIDAA

由表5可知,除Leu、Glu、Ser、Gly、Ala、Pro外,生长猪对10个不同来源小麦的SIDCP和大多数SIDAA的差异显著(P<0.05)或极显著(P<0.01);10个小麦SIDCP的平均值为68.53%(60.32%~84.50%);10个小麦EAA中SIDLys的平均值为71.68%(64.41%~78.47%),SIDIle为80.38%(71.52%~87.09%),SIDLeu为85.35%(81.61%~87.72%),SIDMet为77.77%(69.68%~88.85%),SIDTrp为86.46%(77.98%~93.05%),SIDThr为74.20%(64.86%~82.85%),SIDPhe为83.77%(78.76%~88.10%),SIDHis为73.09%(60.50%~83.84%),SIDArg为68.54%(58.25%~79.34%),SIDVal为80.24%(74.82%~83.54%);10个小麦的EAA中SIDMet的CV大于10%。

2.5 SIDCP、SIDLys、SIDMet、SIDTrp、SIDThr与小麦中化学成分的相关性分析

由表6可知,DM与SIDLys、SIDMet、SIDThr呈显著正相关(P<0.05);NDF与SIDLys呈极显著负相关(P<0.01);ADF与SIDMet呈显著负相关(P<0.05),与SIDTrp呈极显著负相关(P<0.01);Ca与CP呈显著性正相关(P<0.05);Xylans与SIDTrp呈显著正相关(P<0.05);Lys与CP呈显著正相关(P<0.05);Thr与CP、TP呈显著正相关(P<0.05),与Lys呈极显著正相关(P<0.01);Trp与CP呈显著正相关(P<0.05)。

2.6 生长猪对小麦SIDCP、SIDLys、SIDMet、SIDTrp、SIDThr的预测方程

由表7可知,DM为SIDMet与SIDThr的最佳预测因子,NDF和ADF分别为SIDLys与SIDTrp的最佳预测因子。SIDLys的最佳预测方程为SIDLys=113.111-2.804×NDF[剩余标准差(RSD)=1.84,决定系数(R2)=0.873,P<0.01],SIDMet的最佳预测方程为SIDMet=-1 054.812+12.959×DM(RSD=5.90,R2=0.524,P<0.05),SIDTrp的最佳预测方程为SIDTrp=118.828-10.603×ADF(RSD=2.48,R2=0.799,P<0.01),SIDThr的最佳预测方程为SIDThr=-681.144+8.643×DM(RSD=4.86,R2=0.419,P<0.05)。

3 讨论

3.1 小麦的CP和AA含量

在本试验中,测得10个小麦的CP平均含量为13.34%,与《中国猪营养需要》(2020)[14]的报道值相近,略低于NRC(2012)[15]和法国INRA(2004)[16]中硬质小麦的CP含量(NRC,CP含量=14.46%;INRA,CP含量=14.50%),高于NRC(2012)[15]和法国INRA(2004)[16]中软质小麦的CP含量(NRC,CP含量=10.92%;INRA,CP含量=10.50%),这说明不同的数据库中小麦CP含量差异较大。这可能是由于各类小麦在生长环境上的差异所致。例如,在降雨量充沛、气温较低的季节,小麦CP含量较低,在温暖的天气条件下,小麦CP含量高[17]。另外,不同品种也会造成小麦CP含量的差异[18]。
本试验测得10个小麦的AA平均含量与多个数据库的公布值或文献报道值相当[19⇓⇓-22],其中Lys和Met含量略高于NRC(2012)[15]中硬红小麦(Lys含量=0.39%,Met含量=0.22%)、法国INRA(2004)[16]中硬质小麦(Lys含量=0.38%,Met含量=0.23%)和《中国猪营养需要》(2020)[14]中小麦(Lys含量=0.37%,Met含量=0.20%)的公布值,但Lys含量略低于Zhao等[19]的报道值(Lys含量=0.48%);Thr和Trp含量略低于NRC(2012)[15]中硬红小麦(Thr含量=0.40%,Trp含量=0.17%)、法国INRA(2004)[16]中硬质小麦(Thr含量=0.43%,Trp含量=0.16%)和《中国猪营养需要》(2020)[14]中小麦(Thr含量=0.37%,Trp含量=0.15%)公布值,与NRC(2012)[15]中软红小麦(Thr含量=0.35%,Trp含量=0.14%)、法国INRA(2004)[16]中软质小麦(Thr含量=0.32%,Trp含量=0.13%)公布值相近,表明不同数据库及研究报道中小麦AA含量差异较大。实际上,小麦受到生长环境、品种等诸多方面影响,其AA含量具有较高的变异性。在本试验中,10个小麦His、Ile、Lys、Met、Phe、Trp、Asp、Cys、Ser、Tyr含量的CV均超过10%,说明试验所选用样本具有很好的代表性,可以良好地反映小麦的AA含量变异情况。已有多项研究表明,小麦的生长区域、季节、品种以及储藏和加工方式的多样性会导致其AA含量出现一定的波动[23⇓⇓-26]。
需要强调的一点是,Cys与Tyr含量的测定值偏低可能与我们使用的AA衍生剂有关,本试验中样品经前处理等一系列操作后,在上高效液相色谱仪检测时,所使用的AA衍生剂为OPA与FMOC,此法的缺点在于不能对Tyr与Cys进行准确定量,因为这些AA的衍生物的比例不稳定,衍生剂及水解产物都有荧光,且Tyr和Cys的衍生物的荧光容易淬灭,这些因素都会干扰Tyr、Cys等AA测定结果的准确性[27],因此,我们才观察到小麦原料中Cys和Tyr的含量低于《中国猪营养需要》(2020)[14]的公布值。

3.2 生长猪对小麦的AA消化率

SIDAA和AIDAA是评定AA消化率的2项重要营养参数,由于SIDAA能更准确地表示AA的消化率,因此在当前猪饲粮配制中,SIDAA被广泛采用,作为首选的AA营养体系[13],本试验测得的生长猪对10个小麦的SIDLys、SIDMet和SIDThr平均值低于NRC(2012)[15]的公布值(硬质小麦,SIDLys=82%,SIDMet=88%,SIDThr=84%),也低于《中国猪营养需要》(2020)[14]的公布值(小麦,SIDLys=88%,SIDMet=92%,SIDThr=95%),略高于Sauer等[28]的报道值(SIDLys=63%,SIDMet=76%,SIDThr=55%),而SIDTrp低于《中国猪营养需要》(2020)[14]的公布值(SIDTrp=93%),但与NRC(2012)[15]的公布值(硬质小麦,SIDTrp=88%)及McGhee等[29]的报道值(SIDTrp=89.6%)近似。综合来看,这可能是由于小麦储藏年份、品种、环境温度及季节的不同所造成的差异,董文轩等[30]研究表明,2015、2016年入库储存的小麦SIDLys显著高于2014年入库储存的小麦,2015、2016年入库储存的小麦SIDTrp显著高于2012年入库储存的小麦;此外,猪对不同类型小麦的SIDCP和SIDAA也存在差异[31]。基于此,通过选择更多来源和品种的小麦进行营养价值评估,测定生长猪对不同来源小麦中AIDAA和SIDAA,并基于小麦中的有效化学成分构建生长猪对小麦中SIDAA的预测方程,这将有利于实现对小麦SIDAA的动态预测。
在本试验中,观察到小麦来源对SIDCP和几乎所有的SIDAA均有显著或极显著的影响。例如,生长猪对XM5的SIDCP相较于大多数小麦样品更高,这可能是由于RXM5的CP含量较高(CP含量=6.28%)所致。生长猪对XM7的SIDLys较XM5和XM10更好,这可能是由于XM7含有更低水平的NDF(NDF含量=12.25%<16.91%和16.06%),高水平的纤维组分可通过刺激肠道黏蛋白、改变黏液成分、延长消化物保留时间和增加内源性氮损失来降低SIDCP与SI D A A [32⇓⇓⇓-36]。同理,本试验也观察到了生长猪对NDF含量较高的XM10有较低的SIDMet和SIDTrp。最后,一个有趣的现象是,生长猪对XM2的SIDIle、SIDThr、SIDPhe、SIDHis、SIDVal、SIDAsp和SIDTyr为各样品间最低值,这可能意味着XM2的蛋白质营养价值相较于其他小麦差。然而,总体而言,XM2的纤维含量并不是最高的,它现有的化学成分在含量上的表现相对中等。因此,本试验并未完全掌握影响不同来源小麦中SIDCP和SIDAA的关键化学成分,这也揭示出充分研究小麦样本的化学成分,特别是细分其淀粉组分(如快速消化淀粉、慢速消化淀粉、抗性淀粉)及纤维组分(如可溶性膳食纤维、不溶性膳食纤维、总膳食纤维、纤维素、木质素、非淀粉多糖)的必要性。
另一个值得注意的现象是,本试验中Gly与脯氨酸(proline,Pro)的AID与SID大多呈现负值,这意味着Gly和Pro的回肠消化率可能并不准确。这主要是由于回肠食糜中含有过高浓度的Gly和Pro所致。长期采食低蛋质白饲粮及无氮饲粮将影响畜禽体内正常的AA代谢,导致回肠中内源性AA的回收率增加,特别是回肠中Gly与Pro的损失增加,而这可能与谷氨酰胺代谢为Gly和Pro有关[37⇓⇓-40]。Moughan等[41]的研究证实,回肠末端的内源性DM质以蛋白质为主,其AA组成又以精氨酸(arginine,Arg)、天冬氨酸(aspartate,Asp)、谷氨酸(glutamate,Glu)、Pro、Gly和丙氨酸(alanine,Ala)为主,尤其是Pro的浓度很高,这与我们的试验结果一致。回肠食糜中大量Gly和Pro的存在是生长猪处于异常生理状态的表现之一,也是与蛋白质耗竭状态相关的一种假象。因此,在使用无氮饲粮法测定谷物中的SIDCP和SIDAA时,常加入少量合成纤维及酪蛋白等高可消化蛋白质来改善畜禽的异常生理状况[35]。
同时,由于本试验中配制了以玉米淀粉和部分小麦相结合的10种“半纯合饲粮”,且小麦的比例仅为40%,虽然可以避免因饲粮纤维水平过高而导致的瘘管堵塞、适口性差等问题,但饲粮中AA的含量仍然较低,这可能导致猪在试验期内生长率不佳[42]。目前已有试验表明,可以在适应期内而非食糜收集期额外补充一定量的AA晶体混合物,以保证猪在试验期内有更加正常的生理状况,并且这样的操作也不会影响猪对被测试成分的AID与SID[43-44]。然而,在本试验中我们却并未这样做,从而导致了部分SIDAA在数值的不稳定性,这是本研究在试验设计上的局限性之一。

3.3 生长猪对小麦SIDAA预测方程的建立

本试验获得了2个SIDLys和2个SIDTrp的预测方程,以及1个SIDMet和1个SIDThr的预测方程,NDF和ADF是SIDLys和SIDTrp预测方程的最佳预测因子,且在它们的预测方程中起负贡献,这与前人的研究结果[45⇓-47]一致。Feng等[45]的研究表明,生长猪对高粱的SIDLys可以通过NDF和Trp进行预测(SIDLys=158.984-811.047×Trp-1.342×NDF,R2=0.889,RSD=3.24,P<0.01),其中NDF与SIDLys呈负相关。Li等[46]的研究表明,生长猪对花生粕的SIDLys可以通过NDF和Lys进行预测(SIDLys=48.10×Lys-0.42×NDF-0.61,R2=0.88,RSD=2.07,P<0.01),NDF与SIDLys呈负相关。Yun等[47]的研究发现,在肉鸡对小麦的SIDTrp的预测方程中,以CP、NDF、CF和ADF为预测因子可以实现对SIDTrp的准确预测(SIDTrp=58.282+0.594×CP+2.934×NDF+4.307×CF-6.408×ADF,R2=0.866,P<0.01),其中ADF与SIDTrp呈负相关。Lenis等[48]的研究结果显示,在饲粮中添加NDF显著降低了SIDLys,进一步证明了NDF与SIDLys呈显著性负相关,表明NDF可作为SIDLys的预测因子,上述结论均与本试验结果具有高度的一致性。
总而言之,构建SIDAA预测方程有助于减少对AA消化率的测量,从而节约成本,并为畜禽饲料原料的高效利用提供便利[49]。由于CP和AA的供应过多或不足均会对猪的生产性能和环境产生负面影响[13],因此,准确评估饲粮中的CP和AA,以及构建SIDCP和SIDAA的预测方程具有重要意义。

4 结论

饲喂基础下,10个不同来源小麦的化学成分含量变异较大,AIDCP、SIDCP以及大多数AIDAA和SIDAA受小麦来源的影响亦显著或极显著。NDF和ADF分别是SIDLys、SIDTrp的最佳预测因子,DM是SIDMet和SIDThr的共同最佳预测因子;SIDLys的最佳预测方程为SIDLys=113.111-2.804×NDF,SIDMet的最佳预测方程为SIDMet=-1 054.812+12.959×DM,SIDTrp的最佳预测方程为SIDTrp=118.828-10.603×ADF,SIDThr的最佳预测方程为SIDThr=-681.144+8.643×DM。
