中速型黄羽肉仔鸡对不同产地小麦的氨基酸标准回肠消化率评定及其预测方程构建

摘要

试验旨在分析不同产地小麦的化学成分含量,评定中速型黄羽肉仔鸡对小麦氨基酸表观回肠消化率(AID)和标准回肠消化率(SID),并根据其化学成分含量构建预测方程。选取276只63日龄健康中速型黄羽肉仔鸡公鸡,采用单因素完全随机试验设计,设置1个无氮饲粮组和10个小麦饲粮组,每组6个重复,无氮饲粮组每个重复6只鸡,小麦饲粮组每个重复4只鸡。试验期为5 d。在67日龄时收集回肠食糜,测定原料、饲粮和回肠食糜中氨基酸含量,以及饲粮和回肠食糜中二氧化钛(TiO2)含量,并计算氨基酸AID和SID。结果表明:1)10种小麦的总能(GE)及干物质(DM)、粗蛋白质(CP)、粗脂肪(EE)、粗灰分(Ash)、粗纤维(CF)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)、植酸(PA)和无氮浸出物(NFE)含量平均值分别为14.32 MJ/kg、88.42%、12.66%、1.49%、1.46%、2.03%、11.16%、2.78%、0.58%、70.78%;小麦中18种氨基酸含量的平均值在0.12%~4.20%。2)中速型黄羽肉仔鸡对不同产地小麦18种氨基酸AID和SID的平均值分别在58.6%~89.7%和75.8%~94.1%,其中谷氨酸AID和SID均为最高。不同产地小麦对黄羽肉仔鸡18种氨基酸AID和17种氨基酸SID均具有显著影响(P<0.05),而对丝氨酸SID无显著影响(P>0.05)。3)本研究共构建了中速型黄羽肉仔鸡对小麦18种氨基酸SID的预测方程,除了亮氨酸、异亮氨酸和组氨酸SID的预测值外,其余15种氨基酸的SID预测值均在验证小麦样品相应的氨基酸SID实测值±标准差范围内,且多数方程的决定系数(R2)大于0.8,表明具有较好的预测效应。综上所述,不同产地小麦的化学成分含量和氨基酸AID与SID存在差异,所建立的预测方程可为今后快速预测中速型黄羽肉仔鸡对小麦的多数氨基酸SID提供重要参考。

Abstract

This study was conducted to analyze the contents of chemical components in wheat from different sources, and evaluate the apparent ileal digestibilities (AID) and standardized ileal digestibilities (SID) of amino acids in wheat for medium-growing yellow-feathered broilers, as well as establish their predicting equations based on their contents of chemical components. Two hundred and seventy-six healthy 63-day-old medium-growing yellow-feathered broilers were used in a one-way completely randomized design, included 1 nitrogen-free diet group and 10 wheat diet groups, with six replicates per group. There were 6 chickens per replicate in the nitrogen-free diet group, and there were 4 chickens per replicate in the wheat diet groups. The experimental period was 5 days. At 67 days of age, the ileal digesta was collected. The amino acid contents in ingredients, diets and ileal digesta, as well as the titanium dioxide (TiO2) content in diets and ileal digesta were determined, and then both AID and SID of amino acids were calculated. The results showed as follows: 1) the means of gross energy (GE), dry matter (DM), crude protein (CP), ether extract (EE), crude ash (Ash), crude fiber (CF), neutral detergent fiber (NDF), acid detergent fiber (ADF), phytic acid (PA) and nitrogen-free extract (NFE) of 10 wheats were 14.32 MJ/kg, 88.42%, 12.66%, 1.49%, 1.46%, 2.03%, 11.16%, 2.78%, 0.58% and 70.78%, respectively. Additionally, the mean contents of 18 amino acids in wheat ranged from 0.12% to 4.20%. 2) The means of AID and SID of 18 amino acids of wheat from different sources in medium-growing yellow-feathered broilers ranged from 58.6% to 89.7% and 75.8% to 94.1%, respectively, glutamic acid exhibited the highest AID and SID. wheat from different sources significantly affected the AID of 18 amino acids and the SID of 17 amino acids (P<0.05), but not affected the SID of serine (P>0.05). 3) In this study, 18 predicting equations of amino acids SID of wheat for medium-growing yellow-feathered broilers were established. Excepted the predicted values of leucine, isoleucine and histidine SID, the predicted values of SID of other 15 amino acids in the checked wheat sample fell within the ranges of the measured value±standard deviation of corresponding amino acids SID, and most equations had coefficients of determination (R2) greater than 0.8, indicating their good predicting responses. It is concluded that the contents of chemical components and amino acids AID and SID of wheat from different sources exist difference, and the established predicting equations can serve as a valuable reference for quickly predicting the SID of amino acids of wheat for medium-growing yellow-feathered broilers in the future.

精准评定黄羽肉仔鸡对其常用饲料原料中的氨基酸标准回肠消化率(standard ileal digestibility,SID),对于黄羽肉仔鸡制定均衡的饲料配方、精准满足其对氨基酸的需求、提高其对饲料中营养物质的利用效率、减少饲粮中营养物质的浪费、提高生产效益、减少氮排放、解决饲料短缺问题均具有重要意义[1-3]。因此,准确评定中速型黄羽肉仔鸡对不同产地小麦氨基酸SID,对于提高其养殖经济和生态效益意义重大。国家农业农村部畜牧兽医局2020年9月发布的《饲料原料营养价值评定及参数建立项目技术规程》中所应用的回肠末端食糜收集法是当前测定家禽氨基酸消化率经典的方法[4-5],一些学者已利用该方法评定了黄羽肉仔鸡对其常用饲料原料中的氨基酸SID,取得了很好的效果[6-7]。此外,已有研究者提出并建立了基于饲料原料化学特性的氨基酸SID预测方程[8-10]。因此,本研究假设,通过对不同产地小麦饲料原料的化学成分分析,精准评定中速型黄羽肉仔鸡对小麦氨基酸SID和表观回肠消化率(apparent ileal digestibility,AID),并通过其化学成分含量可构建其氨基酸SID预测方程。本研究旨在对中速型黄羽肉仔鸡的小麦氨基酸SID进行准确评价,建立基于其化学成分含量的动态预测模型,以验证上述假设,同时为进一步验证及推广应用上述动态预测模型奠定坚实基础。

1 材料与方法

1.1 试验设计和饲粮

采用单因素完全随机试验设计,使用63日龄中速型黄羽肉仔鸡公鸡进行本试验,试验期为5 d。分别设置无氮饲粮组和由各不同产地小麦配制的10个小麦饲粮组,共11个组。10个不同品种的小麦样品分别由本实验室前期从总共采集的15个不同品种的小麦样品的系列相关分析中筛选得出,小麦样品信息见表1。

购买60日龄的健康温氏天露麻公鸡300只。将鸡只空腹称重,确保体重达到中速型黄羽肉仔鸡60日龄体重的要求,然后选取276只体重相近(重复内鸡只个体体重或重复间鸡只平均体重的变异不超过全群鸡只总体平均体重的5%)的黄羽肉仔鸡公鸡,随机分为11个组,每组6个重复(小麦饲粮组每个重复4只鸡,无氮饲粮组每个重复6只鸡)。鉴于无氮饲粮组肉鸡采食量偏低,可能导致回肠末端食糜收集量不足,因此本研究通过扩大该组试验动物数量(由4只增至6只),以确保样品量满足试验分析要求。试验期环境温度、相对湿度及光照时间均满足黄羽肉仔鸡饲养管理的相应要求。试验鸡饲养在长90 cm、宽70 cm和高45 cm的不锈钢鸡笼中。
根据黄羽肉仔鸡的自由采食量计算每种饲粮的需要量。63日龄前黄羽肉仔鸡饲喂其商品颗粒料。小麦饲粮中,小麦作为唯一蛋白质和氨基酸来源。用小麦和微量成分预混剂等配制成各小麦饲粮,用玉米淀粉、葡萄糖和微量成分预混剂等配制成无氮饲粮,无氮饲粮和小麦饲粮组成及营养水平见表2。各饲粮的必需微量元素和维生素含量均满足我国黄羽肉仔鸡相应阶段的营养需要量(NY/T 3645—2020)。以二氧化钛(TiO2)作为外源指示剂,在小麦饲粮和无氮饲粮中的添加量均为0.4%。

1.2 样品采集与制备

试验鸡于60日龄自由采食商品颗粒料适应3 d后,于63日龄开始自由采食相应组饲粮5 d,于67日龄时,试验鸡自由采食状态下使用二氧化碳窒息死亡后屠宰。无氮饲粮组每个重复屠宰6只鸡,小麦饲粮组每个重复屠宰4只鸡,鸡屠宰后分离回肠,对折后取回肠后段1/2(弃去最后2 cm),通过注射器以蒸馏水冲洗食糜到收集管中,重复冲洗2次,注意避免挤压肠黏膜。收集后的食糜立即放置于-20 ℃储存,冷冻后的食糜放入冷冻干燥机中冻干,置室温下回潮24 h。各重复内所有鸡只的冻干食糜经充分混合后,粉碎备分析。

1.3 指标测定

饲料原料中干物质(DM)、粗脂肪(EE)、粗灰分(Ash)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)和粗纤维(CF)含量分别按照相应国标或行业标准方法(GB/T 6435—2014、GB/T 6433—2006、GB/T 6438—2007、GB/T 20806—2022、NY/T 1459—2022和GB/T 6434—2022)进行测定,植酸(PA)含量采用苏州科铭生物技术有限公司生产的试剂盒测定,总能(GE)采用全自动量热仪测定。参照GB/T 6432—2018测定小麦和饲粮中粗蛋白质(CP)含量,参照GB/T 18246—2019和GB/T 15400—2018测定小麦、饲粮和食糜中18种氨基酸含量,参照GB 5009.246—2016测定饲粮和食糜中TiO2含量,并计算无氮浸出物(NFE)含量。
中速型黄羽肉仔鸡对小麦中回肠氨基酸消化率的计算公式如下:
氨基酸AID(%)=100-[(食糜中氨基酸含量×饲粮中TiO2含量)/(饲粮中氨基酸含量×食糜中TiO2含量)]×100;
无氮饲粮中内源性氨基酸含量(mg/kg)=(食糜中氨基酸含量×饲粮中TiO2
含量)/食糜中TiO2含量;
氨基酸SID(%)=氨基酸AID+(无氮饲粮中内源性氨基酸含量/饲粮中氨基酸含量)×100。

1.4 数据统计分析

采用SAS 9.4软件中的一般线性模型(GLM)对10种不同小麦中氨基酸的AID和SID数据进行单因素方差分析,方差分析显著者,以LSD法比较各平均值间的差异显著性。按照RANDGEN子程序,以随机取样法抽取1种小麦样品来验证预测方程,接着采用SAS 9.4版软件中的皮尔逊相关性分析法进行其余9种小麦样品的化学成分含量与氨基酸SID之间的相关性分析。最后,运用逐步回归法进行拟合回归分析,建立相应氨基酸的最佳预测方程。以每个重复为试验单元,以P<0.05作为差异显著性检验水平。

2 结果

2.1 不同产地小麦的化学成分含量

由表3可知,10种不同产地小麦的GE、DM、CP、EE、Ash、CF、NDF、ADF、PA和NFE含量范围分别在13.76~14.79 MJ/kg、86.58%~90.31%、10.66%~14.81%、0.99%~2.92%、1.25%~1.88%、1.23%~2.55%、8.20%~13.84%、1.06%~5.02%、0.47%~0.74%和67.15%~73.38%。其中,GE、DM和NFE的变异系数较小,分别为2.29%、1.35%和2.83%;EE、CF、ADF的变异系数较大,分别为35.98%、21.00%、41.66%。氨基酸含量平均值范围在0.12%~4.20%。小麦必需氨基酸的变异系数为6.89%~42.40%,其中,蛋氨酸的变异系数最大,色氨酸的变异系数最小;非必需氨基酸的变异系数在8.03%~41.36%,其中,半胱氨酸的变异系数最大,丙氨酸的变异系数最小。

2.2 不同产地小麦对中速型黄羽肉仔鸡回肠氨基酸消化率的影响

由表4可知,不同产地小麦对肉仔鸡18种氨基酸AID均有显著影响(P<0.05)。氨基酸AID的变异系数在2.11%~12.27%,其中,谷氨酸AID变异系数最小,为2.11%;天冬氨酸AID变异系数最大,为12.27%。18种氨基酸AID平均值在58.6%~89.7%。必需氨基酸中,蛋氨酸AID平均值最高,为81.9%;苏氨酸平均值最低,为59.4%。非必需氨基酸中,谷氨酸AID平均值最高,为89.7%;天冬氨酸AID平均值最低,为58.6%。

由表5可知,除丝氨酸外,不同产地小麦对肉仔鸡其他17种氨基酸SID均有显著影响(P<0.05)。氨基酸SID的变异系数在1.67%~9.22%,整体低于氨基酸AID的变异系数,表现出更好的稳定性。其中,谷氨酸SID变异系数最小,为1.67%;色氨酸SID变异系数最大,为9.22%。18种氨基酸SID平均值在75.8%~94.1%,普遍高于氨基酸AID平均值。必需氨基酸中,缬氨酸SID平均值最高,为90.6%;色氨酸SID平均值最低,为75.8%。非必需氨基酸中,谷氨酸SID平均值最高,为94.1%;天冬氨酸SID平均值最低,为81.0%。

2.3 中速型黄羽肉仔鸡对小麦的氨基酸SID与小麦化学成分含量的相关性分析

由表6可知,组氨酸SID与CP含量呈显著正相关(P<0.05),精氨酸SID与CF含量呈显著正相关(P<0.05)。此外,除缬氨酸、蛋氨酸、异亮氨酸、苏氨酸、色氨酸、半胱氨酸和脯氨酸外,其余氨基酸SID均与EE含量呈显著负相关(P<0.05)。

2.4 中速型黄羽肉仔鸡对小麦氨基酸SID预测方程的构建

利用逐步回归法,将小麦的化学成分作为自变量,预测中速型黄羽肉仔鸡对小麦的氨基酸SID,预测方程结果见表7。所建立的预测方程均有效(P<0.05),且多数方程的决定系数(R2)大于0.8,以天冬氨酸、甘氨酸和半胱氨酸SID的预测方程拟合效果最好,其中的主要预测因子包括CP、EE和GE。

2.5 中速型黄羽肉仔鸡对小麦氨基酸SID预测方程的预测准确性检验

用小麦样品10检验中速型黄羽肉仔鸡对小麦氨基酸SID预测方程的准确性。由表8可知,在所有预测方程中,除了亮氨酸、异亮氨酸和组氨酸SID的预测值外,其余15种氨基酸SID的预测值均在该小麦样品相应的氨基酸SID实测值±标准差范围内,表明这些预测方程均具有较好的准确性和适用性。

3 讨论

本试验研究发现,不同产地小麦的化学成分平均含量分别为:GE 14.32 MJ/kg、DM 88.42%、CP 12.66%、EE 1.49%、Ash 1.46%、CF 2.03%、NDF 11.16%、ADF 2.78%、PA 0.58%及NFE 70.78%。18种氨基酸含量的平均值在不同小麦原料中的变化范围在0.12%~4.20%。本研究测定了18种氨基酸AID,其平均值在58.6%~89.7%;氨基酸SID的平均值则在75.8%~94.1%,明显高于氨基酸AID的平均值。基于不同产地小麦的化学成分含量,共构建了18个用于预测中速型黄羽肉仔鸡对小麦氨基酸SID的方程,其中大多数方程的R2大于0.8,显示出较好的预测能力。特别是天冬氨酸、甘氨酸和半胱氨酸SID的预测方程拟合度最高。本研究结果验证了提出的科学假说,为进一步快速预测肉鸡对小麦的氨基酸SID奠定了坚实基础。
本研究中,通过对10种不同产地小麦的常规化学成分含量进行分析发现,除GE、DM和NFE外,其余化学成分含量变异系数均大于10%,表明本试验所选的10种不同产地的小麦样品具有较好的代表性。本试验测定的10种小麦样品CP含量平均值为12.66%,这与Fang等[11]测定的小麦样品CP含量(12.74%)相近。本试验中小麦GE平均值为14.32 MJ/kg,而冯佳婷[12]测得小麦的GE为16.66 MJ/kg,这与本试验结果有一定差异,可能是由于不同研究中所使用的小麦的种植环境不同导致的。本试验中小麦的NDF和ADF含量平均值分别为11.16%和2.78%,与陈思[13]测定的小麦NDF(11.20%)和ADF含量(2.97%)相近。本试验中小麦的EE含量平均值为1.49%,与Ning等[14]报道的小麦EE含量(2.0%)差别略大。本试验中小麦的DM含量平均值为88.42%,与《中国饲料与营养价值表(2024年第35版)》[15]中的DM含量(88%)相近。本试验不同产地小麦的CP和Ash含量平均值分别为12.66%和1.46%,略低于张静波等[16]测得的13.4%和1.9%。此外,本试验测得的CF含量平均值为2.03%,略高于Liu等[17]测得的1.9%。出现上述差异的原因可能与小麦的产地、生长环境、品种以及储存条件等因素有关[18]。本试验测定不同产地小麦样品的18种氨基酸中,变异系数普遍较大,与文献中报道的结果[19-20]一致,部分氨基酸结果与前人研究有所差异,可能受小麦的来源、基因型和环境等因素的影响[21]。
本研究中,中速型黄羽肉仔鸡对不同产地小麦的氨基酸AID平均值介于天冬氨酸(58.6%)至谷氨酸(89.7%)。Barua等[22]在对7~14日龄白羽肉鸡小麦氨基酸AID的研究中发现,在所有氨基酸中,谷氨酸AID最高,而天冬氨酸AID最低,这与本研究结果一致。余建等[23]在仔猪小麦氨基酸AID的研究中表明,赖氨酸和苏氨酸AID较低,这与本试验结果相符。本试验黄羽肉仔鸡对小麦赖氨酸和苏氨酸AID分别为63.3%和59.4%。此外,不同产地小麦对黄羽肉仔鸡18种氨基酸AID有显著影响,这与周雅倩[24]在白羽肉鸡上测得的9种不同产地的小麦麸氨基酸AID的规律相似。本试验中测得苏氨酸、丝氨酸和脯氨酸AID与Osho等[25]在21日龄白羽肉鸡小麦氨基酸AID上的研究结果非常接近。然而,与部分氨基酸AID存在较大差异,这可能与肉鸡品种、日龄、测定方法等因素有关[19]。
中速型黄羽肉仔鸡对小麦的氨基酸SID的变异系数显示,色氨酸最大,谷氨酸最小,这与汪子怡等[26]在对28日龄白羽肉鸡小麦氨基酸SID上的研究结果一致。除丝氨酸外,不同产地小麦对黄羽肉仔鸡其他17种氨基酸SID均有显著影响。本试验测得的赖氨酸、蛋氨酸、异亮氨酸和谷氨酸SID结果与Zhao等[27]在生长猪上测得的小麦氨基酸SID结果接近。本试验中,丝氨酸、脯氨酸和丙氨酸SID结果与陈思[13]对11~13日龄白羽肉鸡小麦氨基酸SID的报道值接近。18种氨基酸SID平均值在色氨酸(75.8%)至谷氨酸(94.1%)之间,而Lu等[28]在26日龄白羽肉鸡上报道的小麦氨基酸SID中,苏氨酸最低为81.6%,谷氨酸最高为95.7%。这些差异可能由于肉鸡品种、日龄、生长环境及小麦产地等因素所致[29-30]。
目前,基于饲料原料营养成分的回归预测方程研究多集中于代谢能,而对氨基酸利用率的研究相对较少,特别是在家禽小麦氨基酸SID预测方程上的研究更少[31]。本研究发现,黄羽肉仔鸡对小麦组氨酸SID与小麦CP含量呈显著正相关,这一结果与Yun等[32]在13日龄白羽肉鸡上小麦氨基酸SID的研究相一致。此外,汪子怡等[26]在28日龄白羽肉鸡小麦氨基酸SID研究中发现,精氨酸SID与小麦CF含量呈正相关,这与本试验结果一致。本研究中,多数小麦氨基酸SID和小麦EE含量之间存在显著负相关关系,这与潘泳霖等[33]在育雏期蛋鸡玉米氨基酸SID的研究以及Feng等[34]在生长育肥猪高粱氨基酸SID研究中的报道结果相符。本试验中,色氨酸、谷氨酸和丙氨酸SID的预测方程中预测因子的数量均为1~2个,R2均在0.8以下,其余方程中的预测因子均在3个以上,并且R2基本大于0.8,这表明方差的预测因子越多,预测方程的预测效果越好[35-36]。现有研究证实,样品的预测值可以验证预测方程的准确性[8]。本研究中,通过小麦样品10对现有预测方程进行验证,除了亮氨酸、异亮氨酸和组氨酸SID的预测值外,其余15种氨基酸SID的预测值均在该小麦样品相应的氨基酸SID实测值±标准差范围内,证明了本研究建立的多数预测方程的准确度较高。最终,本试验共构建了18个氨基酸SID的预测方程,且多数方程的R2大于0.8,显示出较好的预测效果。因此,这些方程可作为预测中速型黄羽肉仔鸡对小麦氨基酸SID的重要参考依据。

4 结论

① 不同产地小麦化学成分含量变异较大,其中GE、DM和NFE含量的变异系数小于10%,而CP、EE、Ash、CF、NDF、ADF和PA含量的变异系数均大于10%。18种氨基酸含量平均值在0.12%~4.20%,变异系数在6.89%~42.40%,其中蛋氨酸的变异系数最大,色氨酸的变异系数最小。
② 中速型黄羽肉仔鸡对不同产地小麦的氨基酸AID平均值在58.6%~89.7%,其中天冬氨酸AID平均值最低为58.6%,谷氨酸AID平均值最高为89.7%;氨基酸SID平均值在75.8%~94.1%,其中色氨酸SID平均值最低为75.8%,谷氨酸SID平均值最高为94.1%。
③ 通过逐步回归法建立了中速型黄羽肉仔鸡对小麦18种氨基酸SID的预测方程,多数方程的R2大于0.8,且具有较好的预测效果。
