为研究赖氨酸对绵羊生长的影响及其机理,选用1岁左右的绵羊15只,平均分为A、B和C组。3组分别在基础日粮中添加0、4和10 g赖氨酸盐酸盐。饲喂28 d后全部屠宰,取肝脏和背最长肌样,提取组织总RNA,反转录后扩增胰岛素样生长因子-Ⅰ(IGF-Ⅰ)和GAPDH基因,用荧光定量PCR法分析不同处理肝脏和背最长肌IGF-Ⅰ基因的表达量。结果表明:C组IGF-Ⅰ基因的相对表达量极显著高于B组(P<0.01),B组IGF-Ⅰ基因的相对表达量极显著高于A组(P<0.01),说明添加一定量的赖氨酸可以调节绵羊组织的生长发育
The objectives of the present study were to investigate the effects and mechanisms of lysine on sheep performance. Fifteen sheep of 1 year old around were divided into 3 groups. Sheep in groups A, B and C were fed basal diets supplemented with 0, 4 and 10 g lysine, respectively. All sheep were slaughtered to collect the samples of liver and longissimus dorsi muscle on day 28. Total RNA was extracted from the tissues, and then IGF-Ⅰ and GAPDH genes were amplified by RT-PCR after reverse transcription. Finally, the expression of IGF-Ⅰ mRNA in liver and longissimus dorsi muscle was analyzed by real time PCR. Results showed relative expression of IGF-Ⅰ gene in group C was significantly higher than that in group B (P<0.01) and relative expression of IGF-Ⅰ gene in group B was significantly higher than that in group A (P<0.01). It was concluded that adding a certain quantity of lysine could regulate growth and development of tissue in sheep.[Chinese Journal of Animal Nutrition, 2010,22(2):487-491]
