体外批次培养法研究莫能菌素对亚油酸氢化及反式11-油酸合成的影响

摘要

采用体外批次培养技术,在以亚油酸(C18∶2)为脂肪酸源的培养液中添加不同水平的莫能菌素,研究其对C18∶2氢化及反式11油酸(t11-C18∶1)合成的影响。试验采用单因子试验设计,以微晶纤维素为培养底物,莫能菌素以其占底物的质量浓度添加,对照组、M1组、M2组、M3组和M4组莫能菌素添加水平分别为0、10、20、30和40 mg/kg。于培养后2、4、8、16和24 h 5个时间点采集样品,每个时间点4个重复,分别测定各时间点样品中硬脂酸(C18∶0)、t11-C18∶1和顺式9,顺式12亚油酸(c9, c12-C18∶2)含量。结果表明:添加莫能菌素能显著降低培养液中C18∶0含量(P<0.05),显著提高t11-C18∶1和c9, c12-C18∶2含量(P<0.05);且莫能菌素浓度高于10 mg/kg时培养液中t11-C18∶1含量随着莫能菌素浓度的增加逐渐升高。结果提示,体外培养液中添加莫能菌素能提高t11-C18∶1含量,且莫能菌素浓度超过10 mg/kg时t11-C18∶1合成随剂量增加而加强。

Abstract

The study was conducted to evaluate the effects of different supplemental levels of monensin on the hydrogenation of linoleic acid (C18∶2) and the synthesis of trans11-oleinic acid (t11-C18∶1) in culture medium, which contained C18∶2 as the source of fatty acids, by a in vitro batch culture method. Single-factor experimental design was used in the experiment. Avicel was used as the substrate. Monensin was added by its mass concentration to the substrate and the levels were 0 (control group), 10 (group M1), 20 (group M2), 30 (group M3), and 40 mg/kg (group M4), respectively. Samples were collected after 2, 4, 8, 16 and 24 h of incubation, four replicates at each time point. Contents of stearic acid (C18∶0), t11-C18∶1 and cis9, cis12-linoleic acid (c9, c12-C18∶2) were determined.The results showed as follows: the content of C18∶0 in culture medium was reduced significantly (P<0.05), meanwhile, t11-C18∶1 and c9, c12-C18∶2 contents were significantly increased (P<0.05) by the supplemention of monensin in vitro; content of t11-C18∶1 was increased as the supplemental level of monensin increased when it was higher than 10 mg/kg. The results indicate that the supplemention of monensin in culture medium can increase the t11-C18∶1 content, and the synthsis of t11-C18∶1 was enhanced as the increasing of monensin level after the level is higher than 10 mg/kg.[Chinese Journal of Animal Nutrition,2011,23(2):343-348]
