This experiment was conducted to study the effects of dietary selenium on selenium content and the mRNA relative expression level of selenium protease gene in the testis of Hy-line breeder cocks. Two hundred and twenty-four 48-week-old Hy-line breeder cocks were selected and randomly divided into 7 groups with 4 replicates per group and 8 Hy-line breeder cocks per replicate. Three levels of senium from different sources (sodium selenite, SS and selenium yeast, SY) were added into the basal diet at 0.2, 0.5 and 0.8 mg/kg (by selenium content), respectively, but no extra selenium was added to the diet in the control group. The experiment lasted for 35 days. The results showed as following: 1) the addition of either SS or SY significantly increased selenium content in testis of breeder cocks (P<0.01), and SY had a more effect than SS (P<0.01). With an increase of the supplemental selenium level, the selenium content in testis of breeder cocks had a significant increase (P<0.01). 2) The supplementation of 0.5 mg/kg SS, 0.5 and 0.8 mg/kg SY significantly increased glutathione peroxidase (GPx) activity in plasma of breeder cocks (P<0.05). 3) The differences of mRNA relative expression level of iodothyronine deiodinase 1 (ID1) gene among all groups were not significant (P>0.05). The mRNA relative expression level of GPx4 gene in supplemental groups was significantly higher than that in the control group (P<0.01), and 0.8 mg/kg SY group was significantly higher than 0.2 mg/kg SY group (P<0.01). The mRNA relative expression level of thioredoxin reductases 2 (TrxR2) in supplemental groups was significantly higher than that in the control group (P<0.01), and 0.8 mg/kg group was significantly higher than 0.2 mg/kg group (P<0.05 or P<0.01). These results indicate that supplemental SY can significantly increase the antioxidant capacity in Hy-line breeder cocks, and 0.8 mg/kg SY has a better effect.[Chinese Journal of Animal Nutrition, 2011, 23(6):1011 -1018]
