Effects of Nano-Ceria on Activity and Gene Expression of Pancreatic Lipase in Hens

Abstract

A test method of pancreatic lipase by real-time fluorescence quantitative PCR was established to investigate the effects of nano-ceria on activity and gene expression of pancreatic lipase in hens. A total of 384 healthy commercial hens at 26 weeks of age were selected and randomly allotted to 4 groups with 8 replicates each group and 12 hens per replicate. The basal diet was a corn-soybean meal diet. The experimental diets were the basal diet supplemented with 0 (group Ⅰ), 30 (group Ⅱ), 60 (group Ⅲ) and 90 mg/kg (group Ⅳ) nano-ceria, respectively. The experiment lasted for 7 weeks. The results showed as follows: 1) this study had established the test method to examine the mRNA expression level of pancreatic lipase using real-time fluorescence quantitative PCR, and this test method had some advantages such as good linear relationship, specificity, sensitivity and reproducibility; 2) the pancreatic lipase activity in group Ⅲ was significantly higher than that in groups Ⅰ and Ⅳ (P<0.05 or P<0.01), and the pancreatic lipase mRNA expression level in group Ⅲ was significantly higher than that in other groups(P<0.05). The results indicate that supplementation of 60 mg/kg nano-ceria in the diet can significantly improve the pancreatic lipase activity and up-regulate the pancreatic lipase mRNA expression level.
