The pharmacokinetic characteristics of canthaxanthin in Carophyll Red in chickens were investigated in this study. After oral administration of Carophyll Red at a dose of 9.6 mg per kg Hy-Line laying hens (12 hens, aged 19 weeks) body weight, venous blood was collected for 10 times in different time periods within 72 h, and serum canthaxanthin concentration was determined by high performance liquid chromatography. Simultaneously, all of the serum drug concentration-time data were dealt with the 3P97 pharmacokinetic software. The results showed that after oral administration of Carophyll Red, serum drug concentration-time data of canthaxanthin in chickens were best described by a one-compartment model with first-order absorption. The theoretical equation was C=0.471(e-0.036-e-0.190), and the main pharmacokinetic parameters as follows: absorption half-life t1/2(Ka)=(3.643±0.205) h, elimination half-life t1/2(Ke)=(19.263±1.312) h, time of the maximum concentration Tmax=(10.795±1.007) h, maximum concentration Cmax=(0.259±0.048) μg/mL, area under serum concentration-time curve AUC=(10.607±1.029) μg/(mL·h), total body clearance CLB=(0.905±0.076) L/(kg·h), and apparent volume of distribution Vd=(2.515±0.133) L/kg. These results indicate that the concentration change of canthaxanthin in serum can reflect the metabolic regularity of Carophyll Red in chickens. Canthaxanthin is absorbed rapidly, and reaches the peak concentration fast, distributes in tissues and organs widely and eliminates slowly in chickens.
