Effects of Replacing Dietary Fish Oil with Soybean Oil on Fatty Acid Metabolism in Hepatocytes of Atlantic Salmon (Salmo salar L.)

Abstract

In order to investigate the effects of replacing dietary fish oil (FO) with soybean oil on fatty acid metabolism in hepatocytes of Atlantic salmon (Salmo salar L.), a total of 50 post-smolt Atlantic salmon with an average body weight of (165±15) g were fed diets containing either FO or soybean oil (SO) as lipid sources for 5 months, and then hepatocytes were isolated and cultured. Some of the cultured hepatocytes were incubated with [1-14C] oleic acid (OA) and then the radioactivity of the cell, cellular lipid component and fatty acid β-oxidation products were analysed. Meanwhile, total RNA of the other hepatocytes were harvested and the relative mRNA expression levels of fatty acid binding protein 3 (FABP3), fatty acid binding protein 10 (FABP10), delta 5 desaturase (D5D), delta 6 desaturase (D6D), and carnitine palmityl transferase Ⅰ (CPT-Ⅰ) were determined by real-time quantitative polymerase chain reaction (RT-qPCR). The results showed as follows: 1) the radioactive activity percentage (RAP) of free fatty acid (FFA) in hepatocytes of FO and SO groups was (62.7±13)% and (55.9±9.1)%, respectively, and was not significantly different (P>0.05); the RAP of phospholipids (PLs) in hepatocytes of FO and SO groups was (20.9±8.7)% and (17.9±5.4)%, respectively, and there was still no significant difference between them (P>0.05); the RAP of triacylglycerol (TAG) in hepatocytes of FO group was (2.0±0.9)%, which was significantly higher than (0.8±0.4)% of SO group (P<0.05); the RAP of β-oxidation products in hepatocytes of FO and SO groups was (1.0±0.3)% and (1.0±0.2)%, respectively, and there was no significant difference between them (P>0.05). 2) The relative expression levels of FABP3, D5D, D6D genes of SO group were significantly higher than those of FO group (P<0.05), while the relative expression levels of FABP10 and CPT-Ⅰ genes were not significantly different between FO and SO groups (P>0.05). The results indicate that replacing dietary FO with SO has no effects on fatty acid metabolism pathways, such as the synthesis of FFA and PLs, and β-oxidation of fatty acid in hepatocytes, but can significantly decrease the synthesis of TAG and promote the fatty acid desaturation in hepatocytes. Therefore, the fatty acid metabolism in hepatocytes is affected in a certain by replacing dietary FO with SO.
