Effects of Dietary Protein and Methionine on Fur Development Related Gene Expressions in Skin of Minks

Abstract

This experiment was conducted to investigate the effects of dietary protein and methionine on fur development related gene expressions in skin of minks. Fifty-six male minks aged 125 days were divided into 4 groups (groups A, B, C and D) with 14 replicates per group. Minks in groups A, B and C were fed the diets with 32%, 24% and 16% protein levels, respectively, and those in group D were fed 24% protein level diet with 0.8% DL-methionine supplementation. Four minks with similar body weight from each group were slaughtered to collect the samples of belly skin on day 85. Total RNAs were extracted from the skin tissues, and then insulin-like growth factor-Ⅰ (IGF-Ⅰ), insulin-like growth factor-Ⅰ receptor (IGF-ⅠR), epidermal growth factor (EGF) and β-actin genes were amplified after reverse transcription. Finally, the expressions of IGF-Ⅰ, IGF-ⅠR and EGF genes in mink's skin were analyzed by real-time fluorescence quantitative PCR. The results showed as follows: IGF-Ⅰ, IGF-ⅠR and EGF gene expressions were significantly affected by different dietary protein levels and methionine supplementation in 24% protein diet (P<0.01), those in group A were significantly higher than those in group C (P<0.01), and those in group D were significantly higher than those in groups B and C (P<0.01). The expressions of IGF-Ⅰ and IGF-ⅠR genes in group D were not significantly different from those in group A (P>0.05), but the expression of EGF gene was significantly lower than that in group A (P<0.01). It is concluded that high dietary protein level and low protein diet adding methionine can improve growth and development of fur in minks.
