Abstract

The purpose of the study was to explore the effects of conjugated linoleic acid (CLA)and α-linolenic acid (ALA)on mRNA and protein expression of nuclear transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2) and glutathione S-transferase A1 (GSTA1) in HepG2 cells. The experiment groups were used 0.2, 0.5 and 1.0 mmol/L CLA and ALA treated HepG2 cells for 24 h, the mRNA expression levels were quantified by quantitative reverse transcription PCR analysis, and the protein expression levels were detected by western blotting method. The results showed as follows: compared with the control group, the mRNA and protein expressions of Nrf2 and GSTA1 were significantly incresed in cells treated with CLA with the concentration of 0.2 and 0.5 mmol/L (P<0.01), and with the CLA concentration increased, the mRNA and protein expressions of Nrf2 and GSTA1 were decreased. However, when HepG2 cells were treated with ALA, the mRNA and protein expressions of Nrf2 and GSTA1 were significantly increased with the ALA concentration increased of compared to control group (P<0.01). In conclusion, CLA with a relatively lower concentration elevates the mRNA and protein expressions of Nrf2 and GSTA1, on the contrary, the mRNA and protein expressions of Nrf2 and GSTA1 are increase gradually with the CLA concentration increased.
