Effects of Dietary Protein Level on Appetitive Peptide Gene Expression of Rabbits

Abstract

This experiment was conducted to investigate the effects of dietary protein level on appetitive peptide gene expression of rabbits. Thirty 30-day-old weaning Ira rabbits with the similar weight were randomly divided into 3 groups (10 repelicates per group and 1 rabbit per repelicate), which were fed the experimental diets with protein levels were 12.8%, 16.0% and 19.2%, respectively. The trial lasted for 7 days for adaptation, and 35 days for trial. After trial, arcuate nucleus (ARC), duodenum, jejunum and ileum were sampled to measure the relative expression levels of proopiomelanocortin (POMC), cocaine-amphetamine (CART), neuropeptide Y (NPY), agouti gene-related protein (AgRP), cholecystokinin (CCK) genes in ARC and CKK gene in intestine. The results showed as follows: the relative expression level of CCK gene in duodenum was significantly higher than that in jejunum, ileum and ARC (P0.05). Compared with 12.8% and 16.0% protein diet groups, the relative expression levels of POMC and CART genes in ARC of 19.2% protein diet group were significantly increased (P0.05). The results suggest that the rabbits with low protein level (12.8% to 16.0%) diets have higher appetite that is relative to the brain-gut neuropeptides. Besides, relative expression levels of CCK gene in duodenum, CART and POMC genes in ARC of high protein level (19.2%) diet are significantly higher than those of low protein level diets, which indicates that the appetite network formed by CCK gens in duodenum, CART and POMC genes in ARC are involved in the regulation process of dietary protein on appetite of rabbits. But CCK gene in ileum and ARC, NPY and AgRP genes in ARC are not sensitive to the dietary protein level variation.
