Effects of Combined Exposure to Deoxynivalenol and Zearalenone on Homeostasis of Chicken Splenic Lymphocytes Cultured in Vitro

Abstract

The aim of this experiment was conducted to study the effects of combined exposure to deoxynivalenol (DON) and zearalenone (ZEA) on the homeostasis of chicken splenic lymphocytes cultured in vitro. The chicken splenic lymphocytes were cultured in vitro with different doses of DON and ZEA in cultured fluid, and the combined exposure doses were DON 0.012 50 μg/mL and ZEA 0.006 25 μg/mL, DON 0.050 μg/mL and ZEA 0.025 μg/mL, DON 0.2 μg/mL and ZEA 0.1 μg/mL, DON 0.8 μg/mL and ZEA 0.4 μg/mL, individually. After cultured 48 hours, the activities of cellular membrane ATPases (Ca2+-ATPase and Na+/K+-ATPase), and the intracellular Ca2+ level, pH and calmodulin (CaM) mRNA expression level in chicken splenic lymphocytes were determined. And the blank contrast group was set separately. The results showed as follows: the intracellular Ca2+ level and CaM mRNA expression level in the treated groups were increased with the increase of toxin concentration, which were significantly or extremely significantly higher than those in the blank control group (P<0.05 or P<0.01). The intracellular pH and the activities of cellular membrane Ca2+-ATPase and Na+/K+-ATPase in the treated groups were decreased with the increase of toxin concentration, which were significantly or extremely significantly lower than those in the control group (P<0.05 or P<0.01). In conclusion, combined exposure to DON and ZEA can lead to a series of intracellular homeostasis, such as intracellular acidification and imbalance of ion homeostasis, which is dose dependent.
