Study on Serum Substance Metabolomics in Acutely Heat-Stressed Broilers Using Gas Chromatography-Mass Spectrometry Technique

Abstract

This experiment was conducted to investigate the changes of serum substance metabolomics in broilers during acute heat stress using gas chromatography-mass spectrometry (GC-MS) technique, and the identified differential metabolites and metabolic pathways were used to explore the metabolic mechanisms of nutrient metabolism pathway changes of acutely heat-stressed broilers. Twenty 35-day-old Arbor Acres (AA) broilers were randomly divided into two groups with 5 replicates per group and 6 broilers per replicate. The control group was fed at normal feeding standard temperature, and the heat stress group was fed at (32±1)℃, both for 12 hours. Blood samples were collected from inferior wing vein, and the separated serum was derivative, serum metabolic profiles of two groups were obtained by GC-MS technique. Analytical methods of principal component and supervised partial least squares discriminant were used to analyze the resultant data, the differential metabolites and metabolic pathways enrichment analysis were conducted. The results showed that 144 metabolites were determined in serum of broilers by GC-MS technique, thirty differential metabolites were screened out[variable important in projection (VIP)>1, P<0.05]. The contents of fourteen metabolites such as fumaric acid, oxybutyric acid and alanine were up-regulated[fold change (FC)>1], the contents of 16 metabolites such as oxalic acid, malic acid and dihydroxyacetone were down-regulated (FC<1). Through the metabolic pathway enrichment analysis, it was found that tricarboxylic acid cycle, galactose metabolism, propionate metabolism, fatty acid biosynthesis and other pathways were significant changed (P<0.05 or P<0.01). In conclusion, serum metabolites and substance metabolic pathway have significant change during acutely heat stress in broilers, and the identified 30 metabolites and 7 metabolic pathways may be used as metabolic biomarkers for the pathogenesis of acute heat stress.
