Effects of Phytol on Metabolic Enzyme Activity of Skeletal Muscle and Muscle Fiber Type of Mice

Abstract

This study was aimed to investigate the effects of phytol on metabolic enzyme activity of skeletal muscle and muscle fiber type of mice. Forty 3-week-old Kunming male mice were randomly divided into 4 groups with 10 in each group, raised in separate cages, and fed diets containing 0 (control group), 0.05%, 0.50% phytol and 0.48% stearic acid (positive control group). The average body weight, relative weight of the gastrocnemius, activities of succinodehydrogenase (SDH), hexokinase (HK), and lactic dehydrogenase (LDH) and triglyceride (TG) content of mice were detected, and ATPase staining was conducted to observe muscle fiber type of mice. The experiment lasted 28 d. The results showed as follows:1) compared with the control group, adding 0.50% phytol significantly decreased the average body weight of mice in the first 2 weeks (P<0.05), and significantly increased the relative weight of the gastrocnemius (P<0.05). 2) Compared with the control group, adding 0.50% phytol significantly reduced TG content in the gastrocnemius of mice (P<0.05), and significantly increased HK activity in the gastrocnemius (P<0.05); adding 0.05% phytol significantly increased SDH activity in the gastrocnemius of mice (P<0.05). 3) Compared with the control group, 0.50% phytol significantly increased the proportion of Ⅰ muscle fiber type in the gastrocnemius of mice (P<0.05). Above all, these results indicate that phytol could increase the relative weight of the gastrocnemius, the activities of SDH and HK, and the proportion of Ⅰ muscle fiber type in the gastrocnemius of mice.
