Effects of Cobalt Source and Cobalt Content on Growth Performance, Hematological Indexes and Cobalt Accumulation in Tissues of Juvenile Cobia (Rachycentron canadum)

Abstract

This experiment was carried out to investigate the effects of dietary cobalt (Co) content on growth performance, hematological indexes and Co accumulation in tissues of juvenile cobia (Rachycentron canadum) under two Co sources[cobalt chloride (Co-Cl) and cobalt methionine (Co-Met)], and to compare the bioavailability of Co-Cl and Co-Met. A basal diet was formulated with vitamin-free casein and fish meal as main protein sources, and 11 experimental diets (the control diet was shared) were prepared by adding 0 (control), 2, 4, 8, 16 and 32 mg/kg Co from Co-Cl and Co-Met, respectively. A total of 990 juvenile cobia with an initial body weight of (22.18±0.35) g were randomly divided into 11 groups with 3 cages (replicates). Each cage had 30 fish, and the fish in 3 cages were fed a kind of experimental diet to saturation for 10 weeks. The results showed as follows:1) under two Co sources, the specific growth rate (SGR) and weight gain ratio (WGR) was increased firstly and then decreased with dietary Co content increasing. Dietary Co content extremely significantly affected the WGR, SGR and feed conversion ratio (FCR) (P<0.01), and the interaction of dietary Co source and Co content significantly affected SGR, WGR and survival rate (SR) (P<0.05). 2) Dietary Co source extremely significantly affected erythrocyte count (RBC) (P<0.01), dietary Co content extremely significantly affected RBC, hemoglobin concentration (HGB) and hematocrit (HCT) (P<0.01), and the interaction of dietary Co source and Co content extremely significantly affected HCT (P<0.01). 3) Dietary Co source extremely significantly affected Co content in vertebra (P<0.01), dietary Co content extremely significantly affected Co content in vertebra and whole body (P<0.01), and the interaction of dietary Co source and Co content significantly affected Co content in vertebra and whole body (P<0.05). In conclusion, suitable dietary Co content can improve the growth performance and hematopoietic indexes of juvenile cobia, and enhance the Co accumulation in tissues. Using Co-Cl and Co-Met as Co sources, the highest SGR of juvenile cobia can be obtained at the Co content of 17.75 and 19.40 mg/kg, respectively. Based on SGR, RBC and Co content in vertebra, the bioavailability of Co-Met is 1.47, 1.49 and 1.12 times that of Co-Cl.
