Effects of Dietary Methionine Level on Growth Performance, Muscle Quality and Muscle Synthesis Pathway Related Gene Expression of Songpu Mirror Carp

Abstract

This experiment was conducted to study the effects of dietary methionine (Met) level on growth performance, muscle quality and muscle synthesis pathway related gene expression of Songpu mirror carp. A total of 540 juvenile Songpu mirror carp with the initial body weight of (24.42±0.19) g were randomly divided into 6 groups with 3 replicates per group and 30 fish per replicate. Fish in 6 groups were fed the isonitrogenous and isoenergetic diets with the Met levels of 0.56% (control), 0.66%, 0.75%, 0.83%, 0.92% and 1.01%, respectively. The experiment lasted for 8 weeks. The results showed as follows:1) the weight gain rate, specific growth rate, protein efficiency ratio and condition factor of 0.83%, 0.92% and 1.01% groups were significantly higher than those of the control group (P<0.05), and the feed conversion ratio was significantly lower than that of the control group (P<0.05). 2) The muscle moisture content of 0.83%, 0.92% and 1.01% groups was significantly lower than that of the control group (P<0.05), the muscle crude protein content of 0.83%, 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05), and the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) in muscle of 0.92% and 1.01% groups were significantly lower than those of the control group (P<0.05). 3) The gene expression of insulin-like growth factor (IGF-Ⅰ) in fast-twitch muscle and slow-twitch muscle of 0.83% and 0.92% groups was significantly higher than that of the control group (P<0.05), the gene expression of mammalian target of rapamycin (mTOR) in fast-twitch muscle and slow-twitch muscle of 0.83%, 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05), the gene expression of serine/threonine protein kinase 11 (STK11) in slow-twitch muscle of 0.66%, 0.75%, 0.83%, 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05), the gene expression of phosphoinositide-3-kinase regulatory subunit 3 (PIK3R3) in fast-twitch muscle of 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05), the gene expression of myosin heavy chain (MHC) in fast-twitch muscle and slow-twitch muscle of 0.66%, 0.75%, 0.83%, 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05), and the gene expression of nuclear factor E2-related factor 2 (Nrf2) in fast-twitch muscle and slow-twitch muscle of 0.83%, 0.92% and 1.01% groups was significantly higher than that of the control group (P<0.05). In conclusion, the addition of Met in diet can improve the growth and muscle quality of carp, and promote expression of muscle synthesis pathway related gene. When dietary cystine level is 0.64%, based on specific growth rate and protein efficiency ratio, using the broken-line model analysis indicate that the Met requirement of juvenile Songpu mirror carp is 0.90%.
