Effects of Feeding Colostrum on Intestinal Permeability and Mucosa-Attached Microbial Composition of Neonatal Goats

Abstract

The objective of this study was to investigate the effects of feeding colostrum on intestinal permeability and mucosa-attached microbial composition of neonatal goats. Sixteen healthy neonatal Ganxi black goats, which had similar birth weight [(1.99±0.08) kg] and did not receive colostrum from their dams, were selected and randomly divided into 2 groups with 8 goats of each group. Goats in milk replacer group and colostrum group received 5% of body weight of milk replacer and bovine colostrum containing 15.43 mg/mL of immunoglobulin G during the first 2 h after birth, respectively. To investigate the changes of intestinal permeability in neonatal goats between 2 groups, six neonatal goats were selected, and 0.5 g mannitol and 1.0 g lactulose were added into milk replacer or colostrum to feed goats. All experimental goats were humanly slaughtered at 12 h after birth; meanwhile, jejunum tissue was collected to determine intestinal mucosa-attached microbial diversity. The results showed as follows: based on 16S sequencing result, the Shannon index of jejunal mucosal-attached microbiota in colostrum group was significantly higher than that in milk replacer group (P<0.05), however, the Simpson index in colostrum group was significantly lower than that in milk replacer group (P<0.05). On the phylum level, the relative abundance of Proteobacteria in jejunal mucosal-attached microbiota in colostrum group was significantly lower than that in milk replacer group (P<0.05), whereas, the relative abundances of Firmicutes and Epsilonbacteraeota were significantly higher than those in milk replacer group (P<0.05). On the genus level, the relative abundance of Escherichia-Shigella in jejunal mucosal-attached microbiota in colostrum group was significantly lower than that in milk replacer group (P<0.05), however, the relative abundances of Lactobacillus and Bifidobacterium were significantly higher than those in milk replacer group (P<0.05). In conclusion, feeding colostrum has no effect on intestinal permeability, but can improve the diversity of intestinal microbiota, promotes the colonization of beneficial bacteria such as Firmicutes, Lactobacillus and Bifidobacterium, inhibits the colonization of harmful bacteria such as Proteobacteria and Escherichia-Shigella in neonatal goats, which is conducive to the establishment of healthy intestinal microbial community in neonatal goats.
