Effects of γ-Aminobutyric Acid on Pancreatic Tissue Structure, Antioxidant Capacity, Digestive Enzyme Activities and Cell Apoptosis of Heat-Stressed Chicks

Abstract

The purpose of this experiment was to investigate the effects of γ-aminobutyric acid (GABA) on pancreatic tissue structure, antioxidant capacity, digestive enzyme activities and cell apoptosis of heat-stressed chicks. Two hundred and forty 1-day-old healthy male Wenchang chicken chicks were randomly divided into 4 groups with 6 replicates per group and 10 chicks per replicate, namely control group (CK group), heat stress group (HS group), control+GABA group (CK+G group) and heat stress+GABA group (HS+G group), and regular feeding for 6 weeks. Starting from 2 weeks of age, each chick in the CK group and HS group was given 0.2 mL of normal saline per day, and each chick in the CK+G group and HS+G group was given 0.2 mL of GABA aqueous solution per day (GABA perfusion was 50 mg/kg BW); chicks in the HS group and HS+G group were given heat stress [(40.0±0.5) ℃, relative humidity (70±5)%] every day 12:00 to 14:00. The results showed that GABA could reduce the damage of heat stress to the pancreas of the chicks, and keep the chick’s pancreatic tissue structure intact. At 5 and 6 weeks of age, the activity of superoxide dismutase (SOD) in pancreatic tissue in the CK+G group and HS+G group was higher than that in the CK group and HS group (P>0.05); at 6 weeks of age, the content of malondialdehyde (MDA) in pancreatic tissue in the CK+G group and HS+G group was significantly lower than that in the HS group (P<0.05). At 3 weeks of age, the lipase activity in pancreatic tissue in the HS+G group was significantly higher than that in the CK group (P<0.05); at 4 weeks of age, the trypsin activity in pancreatic tissue in the CK+G group and HS+G group increased sharply, and it in the CK+G group was significantly higher than that in the HS group (P<0.05). At 2, 3 and 4 weeks of age, the relative expression level of amylase mRNA in pancreatic tissue in the CK+G group was significantly higher than that in the other 3 groups (P<0.05); at 3 weeks of age, the relative expression level of lipase in pancreatic tissue in the HS+G group was significantly higher than that in the CK group (P<0.05), and the relative expression level of trypsin mRNA was significantly higher than that of the HS group (P<0.05). At 5 and 6 weeks of age, the distribution density of apoptotic cells in the HS group was significantly higher than that of the other 3 groups (P<0.05); at 4 and 6 weeks of age, the distribution density of apoptotic cells in the HS+G group decreased, and the difference was not significant compared with the CK group (P>0.05). From the above results, it can be seen that the addition of GABA can help alleviate the damage of the pancreas of heat-stressed chicks, increase the activities of digestive enzymes in the pancreatic tissue of the chicks, enhance the antioxidant capacity, and reduce cell apoptosis.
