Regulation of N-Carbamyl Glutamate, Proline and Putrescine on Sodium Dependent Neutral Amino Acid Transporter-2 Expression and Nutrition Sensing Signaling in Jejunum and Ileum of Piglets

Abstract

The purpose of this study was to investigate regulation of the sodium dependent neutral amino acid transporter-2 (SNAT2) on the nutrition sensing signaling of intestinal mucosa of piglets via polyamines. Thirty-two neonatal piglets (Duroc×Landrace×Large Yorkshire) from 4 litters (8 piglets per litter) were assigned to 4 groups, which received oral administration of equal volumes of saline (control group), putrescine (5 mg/kg BW), proline (25 mg/kg BW) and N-carbamylglutamate (NCG, 8 mg/kg BW) twice daily from days 1 to 14. At day 3 post-weaning, all piglets were slaughtered and their blood, jejunum and ileum were collected. Results showed that compared with control group, oral administration with putrescine, proline and NCG significantly promoted the average daily gain of piglets (P<0.05); supplementation with putrescine and NCG significantly increased the SNAT2 protein expression level in the jejunal and ileal mucosa (P<0.05); proline and NCG treatments significantly increased the mRNA relative expression level of amino acid transport signaling-related genes[proton coupled amino acid transporter 1 (PAT1), calcium sensing receptor (CasR), phospholipase β2 (PLCβ2) and taste receptor 1 family member 3 (T1R3)] in the jejunal mucosa of piglets (P<0.05); meanwhile, NCG treatment significantly increased the mRNA expression level of mammalian target of rapamycin (mTOR) signaling pathway-related genes[mitogen-activated protein kinase 3 (MAP4K3) and mTOR] (P<0.05), as well as the content of cholecystokinin (CCK) in serum (P<0.05) and the mRNA relative expression level of the receptor of CCK in the jejunal mucosa (P<0.05). Therefore, NCG can regulate the release of signal molecules and gene expression of amino acid sensing pathway by up-regulating the expression of genes related to amino acid transporters in intestinal tract of piglets.
