Screening and Degradation Characteristics of Strains Degrading Zearalenone

Abstract

The purpose of this study was to screen the microorganism strains that could safely and effectively degrade zearalenone (ZEN). ZEN was used as the sole carbon to isolate and screen the strains in moldy feed, animal feces and other samples, the strains with higher degradation rates were selected out for identification. The ZEN degradation mechanism of the strains was preliminarily explored. The effects of different inoculation amount, temperature, pH and culture time on the degradation rate were studied, and the safety of the strain was evaluated by mouse experiments. The results showed that two strains that can effectively degraded ZEN were selected from the samples. They were identified as Bacillus amyloliquefaciens and Bacillus subtilis, respectively. After cultured 48 h, 71.3% of the ZEN was degraded by the NA-J21 strain, and 61.5% of ZEN was degraded by the MLS-H32 strain. Both strains degrade ZEN mainly through extracellular enzymes secreted by cells, and the cell walls of the strains had a certain degree of adsorption capacity for ZEN. The optimal conditions for NA-J21 strain to degrade ZEN were that the inoculation amount was 2.0%, the initial pH was 7.0, at a temperature of 37℃ and culture time was 12 h; and for MLS-H32, the optimal conditions for ZEN degradation were that the inoculation amount was 2.5%, the initial pH was 7.0, 32℃, and culture time was 14 h. NA-J21 and MLS-H32 strains could alleviate the organ damage caused by ZEN poisoning in mice, and were safe on some level. The Bacillus amyloliquefaciens and Bacillus subtilis screened in this study can effectively degrade ZEN and both are good microbial for the degradation of ZEN.
