Effects of Betaine on Growth Performance, Liver Damage and Antioxidant Function of Intrauterine Growth Retarded Piglets

Abstract

This study was conducted to investigate the effects of betaine on growth performance, liver damage and antioxidant function of intrauterine growth retarded (IUGR) piglets. One male normal birth weight (NBW) and two same-sex IUGR piglets (total 18 piglets) were chosen from each of six litters of newborn piglets that met the selection criteria for NBW and IUGR. All piglets were allowed to suck the dam naturally up to 7 days of age. After that, the NBW and IUGR piglets in each litter were assigned to the NBW control group, IUGR control group and IUGR experimental group with 6 replicates each and 1 piglet in each replicate, respectively. The NBW control group and IUGR control group received the basal formula milk, while the IUGR experimental group fed the basal formula milk supplemented with 1.0 g/kg betaine (as dry matter of milk powder basis). The trial lasted for 15 days. The results showed as follows:1) compared with the NBW control group, IUGR significantly decreased the body weights at 7 and 21 days of age, the average daily gain (ADG) and average daily feed intake (ADFI) of piglets from 7 to 21 days of age (P<0.05). 2) Compared with the NBW control group, the IUGR control group exhibited significantly higher activities of plasma alanine aminotransferase (ALT) and aspartate aminotransferase (AST) (P<0.05), whereas the IUGR experimental group showed a significantly decreased ALT activity in the plasma of piglets in comparison with the IUGR control group (P<0.05). 3) Compared with the NBW control group, the hepatocellular reactive oxygen species (ROS) level and the contents of malondialdehyde (MDA), protein carbonyl (PC) and oxidized glutathione (GSSG) in liver of piglets in the IUGR control group were significantly increased (P<0.05); in contrast, the activities of γ-glutamyl cysteine ligase (γ-GCL) and glutathione reductase (GR) and the contents of total glutathione (T-GSH), reduced glutathione (GSH), and S-adenosylmethionine (SAM) in liver of piglets were significantly decreased (P<0.05). 4) Compared with the IUGR control group, the hepatocellular ROS level and PC content in liver of piglets in the IUGR experimental group were significantly decreased (P<0.05), while the γ-GCL activity and contents of T-GSH, GSH and SAM were significantly increased (P<0.05). It is concluded that supplementation with betaine at a dose of 1.0 g/kg can improve the antioxidant function and alleviate the oxidative stress and tissue damage in liver of IUGR piglets.
