Analysis of Fecal Microbiota and Blood Metabolome Characteristics of Non-Pregnant Ewes

Abstract

This study aimed to investigate the fecal microbiota and blood metabolome characteristics of non-pregnant ewes, to explore the possible reasons for pregnancy failure in natural mating flocks of ewes after weaning. The ewes were fed and managed under the same condition and divided into pregnant group and non-pregnant group by B ultrasound detection. The fecal and blood samples were collected, and the fecal microbial diversity and differential metabolites were analyzed by Illumina MiSeq sequencing and liquid chromatography-tandem mass spectrometry (LC-MS/MS), respectively. The results showed as follows:1) compared with the pregnant group, the Shannon index of non-pregnant group was significantly decreased (P<0.05); the principal co-ordinates analysis (PCoA) and analysis of similarities (ANOSIM) showed that at the phylum or genus levels, the fecal microbial composition had significant difference between pregnant group and non-pregnant group (P<0.05); compared with the pregnant group, the fecal Actinobacteria relative abundance of non-pregnant group was significantly decreased (P<0.05), the relative abundances of Desulfovibrio and Turicibacter in feces were significantly increased (P<0.05), and the relative abundances of Ruminiclostridium and Corynebacterium in feces were significantly decreased (P<0.05). 2) A total of 78 serum differential metabolites were found in pregnant group and non-pregnant group[variable importance in projection (VIP)>1, P<0.05], the serum LysoPC(O-18:0) content of non-pregnant group was significantly higher than that of pregnant group (P<0.05), and the contents of Deoxyinosine, berteroin and C16-sphingosine in serum were significantly lower than that of pregnant group (P<0.05). Differential metabolites were significantly enriched in 11 signal pathways (P<0.05). 3) Procrustes analysis showed that the trend of microbiological relative abundance was consistent with metabolome expression in pregnant group and non-pregnant group (P<0.05), and the correlation analysis between microorganisms and metabolites showed that Desulfovibrio relative abundance was positively correlated with 3a,7a,12b-trihydroxy-5b-cholanoic acid content (P<0.05). To sum up, compared with pregnant ewes, the composition of fecal microbial and serum metabolites of non-pregnant ewes have significant difference, the interaction of intestinal flora and serum metabolites can affect the physiological state of pregnancy ewes.
