Effects of Proline on Collagen Deposition and Related Gene Expression in Swim Bladder of Chu's Croaker (Nibea coibor)

Abstract

Swim bladder was rich in collagen, and proline (Pro) was a substrate required for collagen synthesis. This experiment was designed to study the effects of different supplemental levels of Pro in diets on growth, collagen deposition and related gene expression of Nibea coibor. A total of 450 Nibea coibor with an initial body weight of (8.64±0.14) g were randomly divided into 6 groups with 3 replicates (cages) in each group and 25 fish in each replicate, and each group was fed isonitrogenous and isolipid diets supplemented with 0, 5, 10, 15, 20 and 25 g/kg Pro, respectively. The experiment lasted for 8 weeks. After the experiment, the growth performance, collagen and amino acid contents in each tissue and relative expression levels of genes related to collagen metabolism of Nibea coibor were determined. The results showed that different supplemental levels of Pro in diets had no significant effects on growth performance of Nibea coibor (P>0.05), but significantly promoted the collagen deposition in swim bladder (P<0.05), and significantly affected the contents of some amino acids in swim bladder (P<0.05), especially the amino acids related to collagen metabolism. The optimal supplemental level of Pro in the diet was estimated to be 14.88 g/kg based on the collagen content in swim bladder. In addition, the relative expression levels of collagen type Ⅰ alpha 1 chain (COL1A1), collagen type Ⅰ alpha 2 chain (COL1A2) and prolyl 4-hydroxylase subunit alpha-1 (P4HA1) genes in swim bladder were increased with the increase of dietary Pro supplemental level (P<0.05). In conclusion, dietary Pro can promote collagen deposition in the swim bladder of Nibea coibor, which is potentially regulated by collagen type Ⅰ alpha chain genes. and the optimal supplemental level is 14.88 g/kg.
