Effects of Hyodeoxycholic Acid on Growth Performance, Energy Metabolism and Fat Digestion and Absorption of Mice

Abstract

This experiment was conducted to investigate the effects of dietary hyodeoxycholic acid (HDCA) on growth, energy metabolism and fat digestion and absorption of mice. Twenty 3-week-old C57BL male mice were divided into control group and HDCA group, and were fed a basal diet and the basal diet supplemented with 1.5% HDCA, respectively. The experiment lasted for 10 weeks. The results showed that compared with the control group:1) HDCA had no significant effects on feed intake and final body weight of mice (P>0.05), but significantly decreased body weight gain of mice (P<0.05). In addition, HDCA significantly reduced the subcutaneous adipose tissue content of mice (P<0.05). 2) HDCA increased thermogenesis of brown adipose tissue (BAT) at 25 and 4℃ (P<0.05), and increased the basal metabolism in both day and night, suggesting that HDCA increased energy metabolism of mice. 3) HDCA extremely significantly reduced the apparent digestibility of crude fat of mice (P<0.01), thereby significantly increasing the crude fat content and fecal energy of mice (P<0.01). 4) HDCA tended to decrease the mRNA expression levels of pancreatic lipase-related genes bile salt-stimulating lipase (BSSL), pancreatic phospholipase A2 (PLA2), pancreatic triglyceride lipase (PTL), pancreatic lipase-related protein 1 (PLRP1) and pancreatic lipase-related protein 2 (PLRP2) of mice (0.05 ≤ P<0.10). In conclusion, dietary supplementation of 1.5% HDCA can improve the energy metabolism of mice, and reduce the digestion and absorption of fat of mice, thereby reducing the subcutaneous adipose tissue content and body weight gain of mice.
