Effects of Riboflavin on Rumen Fermentation Parameters, Enzyme Activities and Microflora Numbers of Sheep in Vitro

Abstract

This experiment was conducted to study the effects of riboflavin on rumen fermentation parameters, enzyme activities and microflora numbers of sheep in vitro. A Rumen fermentation in vitro method was adopted, and supplemented with 0 (control), 15, 30, 45, 60 and 75 mg/kg RF in the fermentation substrate with a concentrate-to-forage ratio of 50:50, respectively, after 48 h in vitro fermentation, the gas production, methane yield, fermentation parameters, enzyme activities and microflora numbers in fermentation broth were measured. The results showed as follows:1) compared with the control group, the 48 h cumulative gas production, slow fermentation part gas production and potential gas production of 30 mg/kg RF group were significantly increased (P<0.05). 2) Compared with the control group, the pH of 15, 30, 45, 60 and 75 mg/kg RF groups was significantly decreased (P<0.05), the concentrations of total volatile fatty acid, propionate, butyrate, valerate and isovalerate of 30 mg/kg RF group were significantly increased (P<0.05), the acetate concentration of 75 mg/kg RF group was significantly decreased (P<0.05), the acetate/propionate of 60 and 75 mg/kg RF groups was significantly decreased (P<0.05), and the ammoniacal nitrogen concentration of 45, 60 and 75 mg/kg RF group was significantly increased (P<0.05). 3) Compared with the control group, the xylanase activity of 75 mg/kg RF group was significantly decreased (P<0.05), the α-amylase activity of 15, 30, 45, 60 and 75 mg/kg RF groups was significantly increased (P<0.05), and the protease activity of 45 mg/kg RF group was significantly increased (P<0.05). 4) Compared with the control group, the numbers of total bacteria, total fungi, total protozoa, Ruminococcus albus, Ruminococcus flavefaciens, Filamentous succinogenes, Ruminobacter amylophilus, Prevotella ruminicola and Butyrivibrio fibrisolvens of 30 mg/kg RF group were significantly increased (P<0.05). In conclusion, under the experimental condition, adding 30 mg/kg RF can increase the fermentation broth volatile fatty acid concentrations, microflora numbers and α-amylase activity.
