Effects of Apparent Digestible Threonine on Growth Performance, Serum Biochemical Indexes and Free Amino Acids Contents of Meat Rabbits Challenged by Lipopolysaccharide

Abstract

This study was conducted to investigate the effects of different levels of apparent digestible threonine (AFDT) on growth performance, serum biochemical indexes and free amino acid contents of meat rabbits challenged with lipopolysaccharide (LPS). A 2×3 double factors design was used in this experiment. A total of 180 healthy New Zealand white rabbits with the same genetic background and initial average weight of (808.11±12.17) g weaned at 35 days old were selected. Two challenge treatment groups were intraperitoneal injection of 100 μg/kg BW LPS and the same amount of sterilized saline (on days 17 and 20 of the test), and three AFDT treatments groups were supplemented with 100%, 125% and 150% of the recommended amount of AFDT (the actual measured values were 0.43%, 0.54% and 0.64%). There were 6 treatments, each treatment consisted of 30 replicates with one rabbit per replicate. Blood samples were collected at day 22 of the test, and the experiment lasted for 35 days. The results showed as follows: 1) dietary AFDT significantly increased the average daily gain (ADG) from days 17 to 22, days 23 to 35 and days 1 to 35, and average daily feed intake (ADFI) from days 23 to 35 and days 1 to 35, while the feed conversion rate (FCR) was decreased from days 17 to 22 (P<0.05); the morbidity (P=0.08) and health risk index (P=0.08) tended to decreased; there was no significant effect on other serum biochemical indexes and serum free amino acid contents except for high density lipoprotein (HDL), alanine (Ala) and globulin (GLB) (P>0.05). 2) LPS challenge significantly reduced the growth performance of meat rabbits (P<0.05), and the decline of 0.43% AFDT group was greater than that of 0.54% AFDT and 0.64% AFDT groups; there was a tendency to increase the mortality rate (P=0.05) and health risk index (P=0.09) of meat rabbits from days 17 to 22; LPS challenge significantly increased the activities of alanine aminotransferase (ALT) and alkaline phosphatase (ALP), and the content of albumin (ALB) in the serum of meat rabbits (P<0.05), and significantly increased the total cholesterol (TC), low density lipoprotein (LDL), free threonine, phenylalanine (Phe) and arginine (Arg) contents in the serum (P<0.05). Adding AFDT to the diets improved health status and growth performance of meat rabbits. Under the condition of LPS challenge, the effect of dietary AFDT supplementation on growth performance of meat rabbits is limited, but it can improve numerically, and 0.64% AFDT has better improvement effects.

由于兔肉的营养价值高,市场需求量大,使得家兔养殖业迅速发展,而在家兔集约化养殖过程中,其发病率大大增加[1]。家兔断奶前后经常发生肠道疾病,同时,外界各种不利因素引起的刺激也会使家兔机体处于紧张状态,引起机体内部代谢功能紊乱,导致家兔出现腹泻、采食量降低、死亡率升高等症状,给家兔养殖业造成较大的经济损失[2-3]。苏氨酸(Thr)作为畜禽的第二或第三限制性氨基酸,在机体中发挥着多种生物学功能,Thr可平衡氨基酸,提高营养物质的利用效率[4-5],通过参与免疫球蛋白的合成来发挥免疫调节作用[6],进而改善动物的生产性能[7]。Thr还具有改善肠道形态,调节黏蛋白合成,维持肠道屏障等功能[8-9],在预防动物肠道疾病和维护肠道健康方面有重要作用,在养殖中具有广阔的应用前景。Thr作为家兔的必需氨基酸,在家兔生长和生产中发挥着重要作用。饲粮中Thr水平过高或者过低都会抑制家兔的生长[10]。脂多糖(lipopolysaccharides,LPS)会引起机体免疫应激,诱导机体产生炎症反应,破坏肠道的屏障功能,干扰营养物质的转运和吸收,最终导致生长性能下降[11]。在多种动物的研究中表明,饲粮添加Thr可缓解应激和攻毒对机体的影响,从而改善生长性能[6,8,12]。但Thr能否缓解LPS应激对肉兔生长性能和血清生化指标的影响还未见报道。因此,为了在家兔生产过程中更合理地应用Thr,本试验选用新西兰白兔为研究对象,考察在正常饲养和LPS应激条件下,饲粮添加表观可消化苏氨酸(AFDT)对肉兔生长性能、健康状况、血清生化指标和游离氨基酸含量的影响,为饲粮中添加适宜水平的Thr来缓解肉兔免疫应激、提高生长性能和调节机体代谢提供参考。

1 材料与方法

1.1 试验材料

Thr为饲料级,纯度≥98.5%;LPS(大肠杆菌血清型O127∶B8)使用前用无菌生理盐水配制成浓度为100 μg/mL的LPS稀释液。

1.2 试验设计

试验采用2×3双因素设计(表1),选用35日龄断奶的健康新西兰白兔180只,体重为(808.11±12.17) g。按照体重相近原则,随机分为6组,每组30个重复,每个重复1只兔。其中3个组于试验第17和20天空腹称重后,腹腔注射100 μg/kg LPS稀释液(即LPS应激处理),其余3个组于同一时间注射等量的无菌生理盐水(即正常饲养)。正常饲养组和LPS应激组各自饲喂AFDT水平分别为0.43%、0.54%和0.64%的饲粮。试验期共35 d,试验第22天进行心脏采血取样。

1.3 试验饲粮

饲粮参照De Blas等[13]推荐的生长肉兔营养需要标准进行配制。3组饲粮AFDT水平分别为0.43%、0.54%和0.65%,分别对应《Nutrition of the Rabbit》(第3版)推荐量的100%、125%和150%,其AFDT的实测值分别为0.43%、0.54%和0.64%。通过调节饲粮中玉米和丙氨酸含量,达到试验饲粮的等能等氮处理。试验饲粮组成及营养水平见表2。饲粮中消化能及可消化氨基酸水平由消化试验测定所得,采用酸不溶灰分作为内源指示剂进行营养物质表观消化率测定[14]。

1.4 饲养管理

试验在四川农业大学动物营养研究所教学科研基地兔舍进行。在试验前对兔舍和所用笼具进行彻底的冲洗和熏蒸消毒。每天09:00和18:00进行饲喂,自由采食和饮水。每天08:00称算前1天的余料量和损料量,每天19:00饲喂后称算给料量。每天清粪2次,保持兔舍清洁卫生。兔舍温度控制在(20±1) ℃,相对湿度保持在55%~60%,自然通风和光照。

1.5 样品的采集

试验的第22天对试验兔空腹称重后,从各组中随机选取6只接近平均体重的试验兔,心脏采血约10 mL,注入不含抗凝剂的采血管中,室温静置30 min后3 000 r/min离心15 min,分离血清,-20 ℃保存备用。

1.6 指标的测定及方法

1.6.1 生长性能的测定

试验期间每天以重复为单位,准确称重并记录试验兔的给料量、余料量和损料量。在试验的第1、17、22天和第36天进行空腹称重,以重复为单位计算各组试验兔不同阶段的平均日增重(ADG)、平均日采食量(ADFI)和饲料转化率(FCR)。参照Gidenne[15]的方法计算患病或死亡试验兔的采食量。
ADG=(试验兔末体重-试验兔初体重)/试验天数;
ADFI=试验兔总采食量/试验天数;
FCR=总采食量/(试验兔末体重-试验兔初体重)。

1.6.2 健康状况的测定

每天以重复为单位,仔细观察试验兔的健康状况(包括腹泻、便秘和精神状态等临床症状),对试验兔的发病数量和死亡数量做记录。参照Gidenne等[16]的方法计算发病率、死亡率和健康风险指数(其中,同一重复试验兔,发病次数和死亡次数只统计1次)。相关计算公式如下:
发病率(%)=100×试验期发病动物数量/试验动物总数量;
死亡率(%)=100×试验期死亡动物数量/试验动物总数量;
健康风险指数(%)=100×(试验期发病动物数量+试验期死亡动物数量)/试验动物数量。

1.6.3 血清生化指标和游离氨基酸含量的测定

采用全自动生化分析仪3100,测定试验兔血清中总蛋白(TP)、白蛋白(ALB)、球蛋白(GLB)、葡萄糖(GLU)、尿素氮(UN)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)含量及碱性磷酸酶(ALP)、谷丙转氨酶(ALT)、谷草转氨酶(AST)活性;利用氨基酸分析仪LA8080测定血清游离氨基酸含量。

1.7 数据统计分析

试验数据采用Excel 2013初步处理,使用统计软件SAS 9.2对试验第1~17天试验兔的生长性能数据进行单因素方差分析及Duncan氏法多重比较,并对其发病率、死亡率和健康风险指数采用χ2检验分析。使用统计软件SAS 9.2的GLM对数据(除试验第1~17天试验兔的生长性能和健康状况数据外)进行双因素方差分析。以饲粮AFDT水平和LPS应激为主效应,饲粮AFDT水平×LPS应激为交互效应。在交互效应显著时,用Duncan氏法进行多重比较,结果用平均值及均值标准误(SEM)表示。以P<0.05表示差异显著,P<0.01表示差异极显著,0.05≤P<0.10表示有显著趋势。

2 结果

2.1 AFDT对LPS应激肉兔生长性能的影响

由表3可知,添加不同水平的AFDT对试验第1~17天肉兔的ADG和FCR无显著影响(P>0.05)。与0.43% AFDT和0.54% AFDT组相比,0.64% AFDT组有增加试验第1~17天肉兔ADFI的趋势(P=0.06)。

由表4可知,LPS应激显著降低了试验第17~22天和第1~35天肉兔ADG和ADFI(P<0.05),显著增加试验第17~22天和第1~35天的FCR(P<0.05)。与0.43% AFDT组相比,0.64% AFDT组试验第17~22天、第23~35天和第1~35天肉兔的ADG以及第23~35天和第1~35天的ADFI显著增加(P<0.05),试验第17~22天的FCR显著降低(P<0.05)。饲粮AFDT水平与LPS应激对肉兔ADG、ADFI和FCR均无显著交互作用(P>0.05)。但无论LPS应激与否,0.54% AFDT和0.64% AFDT组肉兔的生长性能效果均优于0.43% AFDT组。

2.2 AFDT对LPS应激肉兔健康状况的影响

由表5可知,与0.43% AFDT和0.54% AFDT组相比,0.64% AFDT组有降低试验第1~17天肉兔发病率(P=0.08)和健康风险指数(P=0.08)的趋势。

由表6可知,LPS应激有增加试验第17~22天肉兔死亡率(P=0.05)和健康风险指数(P=0.09)的趋势;饲粮添加AFDT对肉兔的健康状况无显著影响(P>0.05)。饲粮AFDT水平与LPS应激交互作用对肉兔的发病率、死亡率和健康风险指数无显著影响(P>0.05)。

2.3 AFDT对LPS应激肉兔血清生化指标的影响

由表7可知,LPS应激显著降低了肉兔血清ALT和ALP活性及ALB含量(P<0.05),显著增加了血清TC和LDL含量(P<0.05),有增加血清GLB(P=0.09)和GLU(P=0.09)含量的趋势。与0.43% AFDT组和0.64% AFDT组相比,0.54% AFDT组血清HDL含量显著降低了(P<0.05)。与0.54% AFDT组相比,0.64% AFDT组血清GLB含量显著增加(P<0.05),有增加血清TP(P=0.05)含量的趋势(P=0.05)。饲粮AFDT水平与LPS应激对肉兔的血清生化指标无显著交互作用(P>0.05)。

2.4 AFDT对LPS应激肉兔血清游离氨基酸含量的影响

由表8和表9可知,LPS应激可显著增加肉兔血清中游离Thr、苯丙氨酸(Phe)和精氨酸(Arg)的含量(P<0.05),有增加血清中游离缬氨酸(Val)(P=0.06)和半胱氨酸(Cys)(P=0.07)含量的趋势。与0.43% AFDT组和0.64% AFDT组相比,0.54% AFDT组有增加血清游离丙氨酸(Ala)含量的趋势(P=0.05)。与0.43% AFDT组和0.54% AFDT组相比,0.64% AFDT组有增加血清游离Cys含量的趋势(P=0.07)。饲粮AFDT水平与LPS应激对肉兔的血清游离氨基酸含量无显著交互作用(P>0.05)。

3 讨论

3.1 AFDT对LPS应激肉兔生长性能和健康状况的影响

LPS应激模型是动物营养学中常见的诱导动物免疫应激模型。研究表明,LPS应激会使机体在生理和行为上出现典型的厌食、嗜睡、腹泻、发热和精神倦怠等特征,降低采食量,诱导机体产生炎症反应,损害肠道结构与功能,影响营养物质的转运和吸收,从而导致动物的生长性能下降,发病率和死亡率增加[17-18]。因此,本试验参照前人研究结果,以断奶肉兔为研究对象,按体重腹腔注射100 μg/kg LPS建立免疫应激模型[19],通过观察发现,注射LPS后肉兔出现体温升高、嗜睡、厌食并伴有腹部震颤等症状;通过分析发现,肉兔生长性能显著降低,死亡率和健康风险指数较生理盐水组均有升高的趋势,由此可见,本试验LPS应激模型建立成功。前人研究结果表明,在饲粮中添加适宜水平的Thr可提高家兔的生长性能[20⇓-22]。同时,在其他动物上的研究指出,饲粮额外添加Thr在一定程度上可促进动物机体的生长发育[23-24]。饲喂杂交兔较低水平的Thr饲粮在一定程度上增加了其死亡率[25]。这与本试验结果相似,在试验前期,0.64% AFDT组肉兔的ADFI增加了,有降低发病率和健康风险指数的趋势,在一定程度上维护了肉兔机体的健康状况。且本试验中0.64% AFDT组提高了LPS应激期间、应激后期和整个试验周期的肉兔生长性能。Chen等[12]在基础饲粮中添加0.30%的Thr可显著缓解LPS应激对肉鸡的生长性能的影响。Bi等[26]研究表明,在免疫应激条件下,饲粮Thr水平的变化对断奶至21日龄北京肉鸭的生长性能无显著的缓解作用,但饲粮Thr水平从0.49%提高到0.56%时可以提高其体增重。
Wellington等[27]在饲粮中额外添加0.13% Thr可以缓解鼠伤寒沙门氏菌对仔猪生长性能降低的影响。在本试验中,饲粮AFDT水平对LPS应激后肉兔的生长性能没有显著的改善作用,但与未LPS应激的0.43% AFDT组相比,0.43% AFDT、0.54% AFDT和0.64% AFDT的LPS应激组在试验第17~22天ADG分别降低了39.62%、33.53%和23.99%,试验第23~35天ADG分别降低了5.75%,增加了0.39%和12.02%。这表明饲粮添加AFDT对LPS应激导致肉兔的生长性能下降在数值上有明显的缓解作用,其中以0.64% AFDT组的缓解和恢复效果最佳。其可能原因是在应激条件下,机体免疫应答增强,体内的氨基酸重新分配,需要摄取更多的Thr来合成免疫球蛋白,同时Thr参与肠道健康的调节,促进营养物质的消化吸收,缓解了LPS应激引起的肉兔生长性能下降。

3.2 AFDT对LPS应激肉兔血清生化指标的影响

血清生化指标可在一定程度上反映动物机体的代谢状况和组织器官的病理变化。TP和ALB含量越高,反映体内蛋白质代谢越旺盛,有利于蛋白质和氨基酸的吸收,从而提高动物的生长性能;GLB是反映机体免疫能力的重要指标,其含量升高是机体免疫力提高的表现[28]。血清中的ALB、GLB、AST和ALT等是应激的重要指标,在应激状态下会导致其在血清中的含量或活性发生改变[3]。此外,ALP参与机体的消化过程,其血清活性的降低与肠道炎症有关[29]。本试验中,LPS应激显著降低了肉兔血清ALT、ALP活性和ALB含量,增加血清LDL和TC含量。这可能是LPS在一定程度上损害了肉兔肠道健康,影响脂质代谢,激发了机体的免疫机能,诱导肉兔体内应激蛋白的合成[28-29]。应激状态下细胞膜受损,会使血清中ALT和AST的活性升高[30]。而在本试验中,血清ALT活性降低,可能原因是由于机体的代偿反应造成。Min等[31]发现,42日龄肉鸡血清中的TP和GLB含量随饲粮Thr水平的增加而升高,并在Thr水平为NRC(1994)推荐量的125%时达到最高值。本试验中,与0.54% AFDT组相比,0.64% AFDT组肉兔血清TP和GLB含量显著增加,表明饲粮更高水平的AFDT可能有利于肉兔对蛋白质和氨基酸代谢,在一定程度上促进抗体合成,发挥机体免疫功能。Thr参与脂质的代谢和沉积,饲粮添加Thr对机体脂质代谢有正向调节作用[32]。张艳蕾[20]研究表明,饲粮Thr水平变化对断奶至2月龄肉兔血清TC、TG和LDL含量无显著影响。崔斌[21]研究指出,饲粮Thr水平变化对生长獭兔血清TG和LDL含量无显著影响,而高水平的Thr显著降低了妊娠母兔的血清TG含量。本试验中,饲粮添加AFDT并不能有效改善肉兔脂质代谢,其可能原因是动物的种属、所处阶段以及生理状态不同所造成。

3.3 饲粮添加AFDT对肉兔血清游离氨基酸含量的影响

血清游离氨基酸可作为评判饲粮蛋白质和氨基酸是否平衡的重要指标。应激和采食行为均可使机体的蛋白质降解,从而导致血清中的游离氨基酸含量增加,而体内的蛋白质和氨基酸代谢处于动态平衡中,最终趋向于降低血清氨基酸含量[33-34]。本试验LPS应激导致家兔血清中游离Thr、Phe和Arg含量显著增多,Cys含量有增多的趋势。可能是LPS应激导致肉兔血清中游离氨基酸含量发生改变,影响机体氨基酸代谢。Abe等[35]通过设置饲粮中蛋氨酸(Met)和Thr不同的比例饲喂荷兰幼兔和荷兰成年兔发现,其比例的变化显著影响血浆Met和Thr含量,同时血浆中其他氨基酸含量也受到了影响。研究表明,给小鼠饲喂高剂量的Thr,血清中游离Thr含量可增高5~47倍[34]。Azzam等[4]在高温高湿条件下,配制Thr水平分别为0.47%、0.57%、0.67%、0.77%和0.87%的5种饲粮饲喂40周龄蛋鸡8周后发现,蛋鸡血清中游离的Thr含量随饲粮Thr水平的增加呈二次曲线变化,对血清中其他游离氨基酸的含量并没有显著影响。Mao等[36]研究表明,在伪狂犬攻毒条件下,仔猪血清中游离的Thr含量随饲粮Thr水平的增加而增加,同时可缓解伪狂犬病毒刺激引起的仔猪血清Val和亮氨酸(Leu)含量的增加。本试验中,饲粮添加AFDT对血清游离Thr含量影响不显著,但在数值上均有明显提高,在一定程度上表明本试验AFDT添加成功,血清中游离的Thr含量的增加可为机体提供合成免疫球蛋白的原料,在一定程度上增加了肉兔的免疫功能。目前结果表明,饲粮添加AFDT并没有显著影响血清其他游离氨基酸含量,可能是肉兔体内氨基酸利用率较高,体内氨基酸平衡且代谢良好[37]。

4 结论

在本试验中,LPS应激导致肉兔机体代谢紊乱,血清生化指标和血清游离氨基酸含量发生变化,生长性能下降。饲粮AFDT水平为0.64%时可促进生长肉兔体内氨基酸平衡、改善健康状况、提高机体的免疫功能和生长性能。饲粮AFDT水平的增加并不能显著缓解LPS应激对肉兔生长性能造成的影响,但在数值上均有所改善,且以0.64% AFDT的缓解和恢复效果最佳。
