Effects of Corn Gluten Meal Replacing Soybean Meal on Growth Performance, Slaughter Performance, Meat Quality, Antioxidant Capacity and Intestinal Score of Yellow Feet Broilers

Abstract

This experiment was conducted to investigate the effects of corn gluten meal replacing soybean meal at different levels on growth performance, slaughter performance, meat quality, antioxidant capacity and intestinal score of yellow feet broilers. A total of 1 280 nineteen-days-old yellow feet broilers were selected and randomly divided into 4 groups with 10 replicates per group and 40 broilers per replicate. Broilers in group A (control group) were fed a basal diet, for groups B, C and D, and additional 1.3%, 2.6% and 3.8% corn gluten meal was added to the basal diet replacing soybean meal, respectively. The experiment lasted for 38 days, which divided into 2 stages: 19 to 36 days of age and 37 to 56 days of age. The results showed that there were no significant differences in growth performance at 19 to 36 days of age, 37 to 56 days of age and 19 to 56 days and slaughter performance at 56 days of age among all groups (P>0.05). The pH24 h of breast muscle in group A was significantly higher than that in group B (P<0.05), and had no significant difference with groups C and D (P>0.05). There was no significant difference in serum and breast muscle superoxide dismutase (SOD) activity among all groups (P>0.05), but with the increase of corn gluten meal supplemental level, the serum and breast muscle malondialdehyde (MDA) content decreased, and it in group D was significantly lower than that in group A (P<0.05). There were no significant differences in intestinal scores among groups at 42 and 56 days of age (P>0.05). In conclusion, under the conditions of this experiment, the supplementation of corn gluten meal with different levels to replace soybean meal has no adverse effects on growth performance, slaughter performance and intestinal scores of yellow feet broilers, but can significantly reduce the serum MDA content and improve the antioxidant capacity of yellow feet broilers.

玉米蛋白粉又称玉米麸质粉,是玉米经过湿磨工艺生产玉米糖浆、玉米淀粉或酿酒过程中的副产品,是一种高能量高蛋白质的蛋白质饲料原料,含有至少60%以上的粗蛋白质[1]。由于其粗蛋白质含量高,在饲料行业常用于替代豆粕或动物蛋白质饲料。虽然玉米蛋白粉的粗蛋白质含量高,但近50%为醇溶蛋白,在单胃动物体内不易于被消化吸收[2],且其氨基酸组成不平衡(蛋氨酸和谷氨酸含量丰富,但色氨酸和赖氨酸含量较少),再加上抗营养因子含量较高,导致玉米蛋白粉饲用效果差,限制了其在饲粮中的添加量[3]。我国农业农村部于2021年颁布了《猪鸡饲料玉米豆粕减量替代技术方案》,建议肉鸡饲粮中玉米蛋白粉的最高添加量为10%[4]。目前玉米蛋白粉在黄脚麻鸡生产中的相关研究较少,因此,本试验使用玉米蛋白粉替代部分豆粕,研究其对黄脚麻鸡生长性能、屠宰性能、抗氧化能力、肉品质和肠道评分的影响,为玉米蛋白粉在黄脚麻鸡生产中的合理使用提供理论依据。

1 材料与方法

1.1 试验材料

试验使用玉米蛋白粉购自山东寿光某生物科技有限公司,其检测结果见表1。

1.2 试验设计与试验饲粮

选取体重[(482.5±0.8) g]接近、健康的19日龄黄脚麻公鸡1 280只,随机分为4组,每组8个重复,每个重复40只。A组为对照组,饲喂基础饲粮;B、C、D组为试验组,分别使用1.3%、2.6%和3.8%的玉米蛋白粉等蛋白质替代基础饲粮中的豆粕。试验饲粮参照育种公司推荐标准及NRC(1994)标准配制,其组成及营养水平见表2。试验期为38 d,分19~36日龄和37~56日龄2个阶段进行,每个阶段各组饲粮的营养水平一致。

1.3 饲养管理

饲养试验采用地面平养,每栏为1个重复。自由采食和饮水,每日观察鸡群健康情况与精神状况,按正常免疫程序进行免疫。鸡舍内温度、相对湿度、光照和卫生学指标符合肉鸡饲养的卫生要求(GB 14925—2010)。

1.4 测定指标与方法

1.4.1 生长性能

分别在肉鸡36和56日龄时,对整栏试验鸡进行空腹称重并统计存活数和采食量,以计算平均日增重(ADG)、平均日采食量(ADFI)、料重比(F/G)和存活率(SR)。

1.4.2 屠宰性能

在56日龄时,于每个重复随机选取1只肉鸡称重后屠宰,称量全净膛重、胸肌重、腿肌重、翅膀重、腹脂重和皮重,参照NY/T 823—2020方法计算胸肌率、腿肌率、翅膀率、腹脂率和皮重率。
全净膛率(%)=[全净膛重(g)/活重(g)]×100;
胸肌率(%)=[胸肌重(g)/全净膛重(g)]×100;
腿肌率(%)=[腿肌重(g)/全净膛重(g)]×100;
翅膀率(%)=[翅膀重(g)/全净膛重(g)]×100;
腹脂率(%)={腹脂重(g)/[全净膛重(g)+腹脂重(g)]}×100;
皮重率(%)=[皮重(g)/全净膛重(g)]×100。

1.4.3 抗氧化指标

于56日龄称重计算生长性能数据后,从每个重复中随机选取1只肉鸡采血,将采集的血液室温静置30 min后3 000 r/min离心15 min制备血清,-20 ℃保存待测。将屠宰后采集的胸肌与冰生理盐水按照1∶9的比例混匀,用组织研磨仪进行匀浆。组织匀浆液中蛋白质含量的测定采用南京建成生物工程研究所生产的总蛋白含量(BCA法)测定试剂盒测定,严格按照试剂盒说明书操作。
血清和胸肌中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量使用南京建成生物工程有限公司生产的可见显色法试剂盒测定,操作步骤按试剂盒说明进行。

1.4.4 肉品质

取一侧胸肌进行肉品质的检测,具体检测指标及方法如下。
pH:分别在屠宰后45 min和24 h,用pH计(Testo-205,德国)固体探头直接插入胸肌样品0.5~1.0 cm深处,每块胸肉重复测定3次,取平均值。pH计使用前要用pH=7和pH=4的校正液进行校正,每次测定前都要清洗、沥干,以免影响测定结果。
肉色:在屠宰后24 h,用便携式色度仪(爱色丽SP62测色仪,美国),根据CIELAB表色系统,测定胸肌亮度(L*)、红度(a*)、黄度(b*)值,每块胸肉重复测定6次,取平均值。

1.4.5 肠道评分

于42和56日龄时,每个重复随机选取1只肉鸡屠宰,取出肠道,对胀气、水分增多、肠壁变薄、弹性减弱、黏液增多、黏膜脱落、后肠段含料量增多、出血性肠炎和气泡增多等指标进行评分,分值为0~4分,分值越高则代表该病变越严重。

1.5 数据处理与分析

试验数据采用SPSS 16.0统计软件中一般线性模型(GLM)以方差分析(ANOVA)方法进行分析(肠道评分指标使用卡方检验分析),所有指标以重复为试验单位。若方差分析显著,则采用Duncan氏法进行多重比较。结果用平均值±标准误表示,P<0.05为差异显著。

2 结果与分析

2.1 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡生长性能的影响

由表3可知,各组黄脚麻鸡初始体重(19日龄体重)均无显著差异(P>0.05),36和56日龄体重亦无显著差异(P>0.05)。在试验前期(19~36日龄)、后期(37~56日龄)和全期(19~56日龄),各组黄脚麻鸡的ADG、F/G、ADFI和SR均无显著差异(P>0.05)。

2.2 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡屠宰性能的影响

由表4可知,各组黄脚麻鸡的全净膛率、胸肌率、腿重率、翅膀率、腹脂率和皮重率均无显著差异(P>0.05)。

2.3 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡肉品质的影响

由表5可知,各组黄脚麻鸡胸肌的肉色L*、a*、b*值和pH45 min均无显著差异(P>0.05),但随着玉米蛋白粉添加水平的升高,a*值有降低的趋势(P=0.064);此外,A组黄脚麻鸡胸肌的pH24 h显著高于B组(P<0.05),与C、D组则无显著差异(P>0.05)。

2.4 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡抗氧化能力的影响

由表6可知,各组黄脚麻鸡血清与胸肌中SOD活性均无显著差异(P>0.05);血清与胸肌中MDA含量均随着玉米蛋白粉添加水平的升高而降低,其中D组血清中MDA含量显著低于A、B组(P<0.05),胸肌中MDA含量显著低于A组(P<0.05)。

2.5 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡肠道评分的影响

由表7可知,黄脚麻鸡42和56日龄的各项肠道评分指标(胀气、水分增多、肠壁变薄、弹性减弱、黏液增多、黏膜脱落、后肠段含料量增多、出血性肠炎、气泡增多)各组之间均无显著差异(P>0.05)。

《猪鸡饲料玉米豆粕减量替代技术方案》中明确提出肉鸡饲粮中玉米蛋白粉的最高添加量为10%,并搭配菜籽饼、棉籽饼和花生粕等其他饼粕使用,豆粕用量可降低为0[4]。但是由于玉米蛋白粉中醇溶蛋白含量较高且氨基酸组成不平衡等原因,若未经发酵、酶解等预处理,其在单胃动物体内不易于被消化吸收,饲用添加效果不理想[5]。在一项用玉米蛋白粉替代肉鸡饲粮中豆粕的研究发现,使用20%玉米蛋白粉替代豆粕会在一定程度上降低肉鸡的出栏体重与ADG,但对整个试验周期的F/G无显著影响[6],这与本研究结果部分相似。本试验结果表明,饲喂不同水平玉米蛋白粉替代豆粕的饲粮对黄脚麻鸡试验前期、后期以及全期的生长性能指标均无显著影响。但是也有研究发现,与使用豆粕和菜籽粕相比,饲粮中使用玉米蛋白粉降低了9~33日龄罗斯308肉仔鸡的ADFI、ADG和饲料转化率[7]。Abhijeet等[8]研究发现,饲粮中添加10%玉米蛋白粉可显著降低1~42日龄肉鸡的ADFI和F/G并提高出栏体重。这与本试验结果不同,可能是所用玉米蛋白粉的添加水平和质量差异造成了生长性能结果的差异。由此可见,一定水平的玉米蛋白粉能够有效替代豆粕,不会对肉鸡的生长性能产生不良影响。本研究结果表明,使用3.8%的玉米蛋白粉等蛋白质替代豆粕不会对黄脚麻鸡的生长性能造成负面影响。

3.2 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡屠宰性能和肉品质的影响

屠宰性能和肉品质可反映出家禽生长状况与产肉质量,密切影响着养殖效益[9]。童辉等[10]研究发现,饲粮中添加不同水平的玉米蛋白粉酶解产物玉米蛋白多肽,对30和60日龄豁眼鹅的屠宰率没有显著影响。这与本研究结果相似,本研究发现不同水平玉米蛋白粉替代豆粕对黄脚麻鸡的屠宰性能没有不良影响。本研究还发现,低剂量玉米蛋白粉的添加降低了胸肌pH24 h,但随着玉米蛋白粉添加水平的提高,这种现象消失。细胞受到自由基攻击之后会加速糖原酵解并产生大量乳酸,最后导致肌肉pH的降低,而随着玉米蛋白粉添加水平的提高,延缓了肉中pH的下降。
玉米蛋白粉中叶黄素含量是玉米的15~20倍,对家禽的皮肤和蛋黄着色具有一定作用[11]。本试验发现,玉米蛋白粉的添加没有导致黄脚麻鸡胸肌的肉色出现明显变化,原因有可能是本试验基础饲粮中未使用玉米,因而导致配方中色素含量很低,同时玉米蛋白粉添加量不足,使得叶黄素沉积量未达到显著效果,有研究表明类胡萝卜素类色素在动物组织上的沉积量与摄入量呈非线性正相关[12],较低的添加量可能达不到应有的效果。
肌肉a*值在一定程度上反映了肉中肌红蛋白的数量,肌红蛋白与氧结合形成鲜红色的氧合肌红蛋白,随着时间延长氧气缺少,肌红蛋白与水结合,呈暗红色[13]。值得一提的是,本试验发现,随着玉米蛋白粉添加水平的提高,黄脚麻鸡胸肌的a*值有较为明显的降低趋势。其原因可能是玉米蛋白粉的使用促进了肌红蛋白与水的结合或抑制了与氧的结合,使胸肌的a*值在一定程度上降低。另外,在热应激[14]、球虫感染[15]、抓鸡等应激条件下会导致肌肉失水率升高,水分渗出产生白肌肉,a*值下降。目前几乎没有关于玉米蛋白粉对黄脚麻鸡屠宰性能和肉品质影响的研究,其降低黄脚麻鸡胸肌a*值的具体原因还需进一步研究。

3.3 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡抗氧化能力的影响

机体氧化还原系统的平衡反映了动物的健康程度,而肉鸡在现代高密度的规模化养殖中,容易受到温度、相对湿度、疾病等因素的影响导致机体产生氧化应激,应激状态下的肉鸡机体会产生大量自由基,继而引发肠道损伤、消化系统紊乱并降低免疫力[16]。抗氧化酶SOD的活性和脂质过氧化产物MDA的含量均可以反映机体抗氧化应激的能力[17]。玉米蛋白粉中的叶黄素具有一定的抗氧化和免疫调节功能[18]。研究表明,饲粮添加不同梯度的叶黄素可以线性地提高肉鸡肝脏和腿肌中SOD的活性,并降低MDA的含量[19]。另外,蛋白质中肽链氨基酸的组成、结构和定位与其抗氧化活性密切相关,虽然玉米蛋白粉中醇溶蛋白含量较高,但其特殊的理化性质与氨基酸组成,使其具有良好的抗氧化活性[20-21]。Jiang等[22]从玉米蛋白粉酶解反应产物中鉴定出多种具有较强自由基吸附能力的抗氧化肽。另外,Jin等[23]也发现,Alcalase+Flavourzyase催化水解玉米蛋白粉后也得到了具有较好还原能力和对1,1-二苯基-2-三硝基苯肼(DPPH)自由基和超氧阴离子自由基清除能力的抗氧化肽。同样地,蔡佳[24]对玉米蛋白粉的黑曲霉发酵产物进行了抗氧化能力测定,结果表明其对DPPH自由基及羟基自由基均具有一定的清除能力。以上研究均表明玉米蛋白粉及其酶解产物具有一定的抗氧化活性,本试验也印证了这一点。饲粮中添加1.3%、2.6%和3.8%玉米蛋白粉均可降低56日龄黄脚麻鸡血清和胸肌中MDA含量,并以添加水平为3.8%时降低效果最好,虽然玉米蛋白粉对血清中SOD活性的提高效果未达到显著水平,但3个添加玉米蛋白粉的试验组血清SOD活性均高于对照组。还有研究表明,饲粮中添加玉米蛋白粉能够显著降低56日龄黄脚麻鸡血清中谷胱甘肽过氧化物酶(GSH-Px)与肝脏中总超氧化物歧化酶(T-SOD)活性,降低肝脏中MDA含量,并且肉鸡血清对超氧阴离子自由基和羟基自由基的清除能力显著提高[25]。

3.4 不同水平玉米蛋白粉替代豆粕对黄脚麻鸡肠道评分的影响

肠道是营养物质消化吸收的最重要的场所,与动物健康状况密不可分,肠道结构的健康是保证动物健康生产的必要前提条件[26]。本试验对肠道的胀气、黏膜脱落、坏死性肠炎等肠道损伤指标进行了评分,肠道评分可以在一定程度上反映肠道的健康程度[27]。Giannenas等[6]研究表明,使用20%的玉米蛋白粉替代豆粕饲喂罗斯308肉鸡,对其十二指肠、空肠和回肠的绒毛高度、隐窝深度和绒隐比均无显著影响。Olukosi等[28]在基础饲粮中额外添加50%的玉米蛋白粉构建高蛋白质饲粮模型,饲喂14~21日龄肉鸡,试验结束时,高蛋白质饲粮组肉鸡空肠绒毛高度、绒毛宽度、隐窝深度和绒隐比与对照组均无显著差异。Wang等[29]在基础饲粮中添加10%的发酵玉米蛋白粉,结果发现发酵玉米蛋白粉不仅对肠道形态没有负面影响,反而提高了十二指肠、空肠和回肠的绒隐比。肠道评分与肠道形态一样均可以揭示肠道健康的一些信息。在本试验中,不同水平玉米蛋白粉替代豆粕后各肠道评分指标均没有发生显著变化,在黄脚麻鸡胃肠道中也没有发现明显的临床或亚临床炎症反应的病变,所有的试验鸡都很健康,试验过程中没有出现因肠道疾病而死亡或腹泻的现象。以上研究结果表明,使用一定水平的玉米蛋白粉替代豆粕不会对黄脚麻鸡肠道形态结构产生不良影响。

4 结论

在本试验条件下,饲粮中使用3.8%的玉米蛋白粉等蛋白质替代饲粮中的豆粕不会对黄脚麻鸡的生长性能、屠宰性能和肠道评分造成负面影响,但会在一定程度上降低胸肌的a*值;此外,玉米蛋白粉的使用会提高黄脚麻鸡机体的抗氧化能力。
