Effects of Compound Carbon Source Diets with Different Carbon to Nitrogen Ratios on Growth Performance and Body Nutrient Contents of Black Soldier Fly (Hermetia illucens) Larvae

Abstract

This experiment was conducted to investigate the effects of two types of dietary carbon sources with different mixing ratios and different dietary carbon to nitrogen (C/N) ratios on growth performance and body nutrient contents of black soldier fly (Hermetia illucens) larvae. Forty-five thousand 3-day-old black soldier fly larvae with the same genetic background, the same batch, normal development, and the same body size and body weight (about 11.33 mg/head) were randomly divided into 25 treatments with 3 replicates per treatment and 600 larvae per replicate. Meanwhile, using peanut meal as nitrogen raw material, a total of 25 diets for black soldier fly larvae with five different ratios of carbon sources [100∶0 (P100), 75∶25 (P75M25), 50∶50 (P50M50), 25∶75 (P25M75) and 0∶100 (M100)], which were composed of easily bioavailable carbon source glucose and difficultly bioavailable carbon source wood chips, and with C/N ratios of 10, 15, 20, 25 and 30 were prepared, and the initial water content was adjusted to 72%. The experiment was stopped when 10 larvae pupated during the experiment. The results showed as follows: 1) compared with the other treatments, when the C/N ratio was 20 or above, the relative moisture content (RMC) of substrate in P100 treatment was more than 139%, and the larval survival rate was significantly decreased (P<0.05). Under the same carbon source conditions, with the increase of C/N ratio, the larval yield, body weight gain rate and a hundred worm weight of black soldier fly larvae showed a gradual decline. 2) With the increase of dietary wood chip ratio, the body contents of crude protein, crude fiber and crude ash of black soldier fly larvae were significantly increased under each C/N ratio (P<0.05), on the contrary, the body ether extract content of black soldier fly larvae was significantly decreased under each C/N ratio (P<0.05). The body ether extract content in P75M25 and P50M50 treatments was higher when the C/N ratio was 25 (47.88% and 46.57%), which was significantly higher than that in the other treatments (P<0.05). 3) Through orthogonal partial least square (OPLS) modeling and variable importance in projection (VIP) analysis, the VIP values of the RMC and ratio of available carbon and nitrogen were all greater than 0.8, and the two types of component indices contributed most significantly to the growth performance and the body nutrient contents of body of black soldier fly larvae. At the same time, the total carbon to total nitrogen (TC/TN) ratio and the ratio of carbon components to total nitrogen (TN) had no significant influence on larval indices. In conclusion, when compound carbon sources are added into the diet for black soldier fly larvae, the proportion of difficultly bioavailable carbon sources should not exceed 25%. When easily bioavailable carbon sources are selected as external carbon sources, the C/N ratio should be controlled below 20 and RMC should be lower than 139%. RMC and TC/TN ratio have some limitations in the dietary ingredients of black soldier fly larvae, which RMC and available carbon and nitrogen ratio significantly affect the growth performance and body nutrient composition of black soldier fly larvae, and should be paid more attention in dietary ingredients of black soldier fly larvae.

黑水虻(Hermetia illucens L.)作为一种食腐性昆虫,能够通过食物链方法迅速将餐厨垃圾、畜禽粪便、变质疏果以及食品加工下脚料等有机废弃物转化为自身优质虫体蛋白质和脂肪,而虫粉可部分替代豆粕、鱼粉和鱼油作为动物饲料蛋白质和脂肪来源,是当下可持续循环经济的新方法以及解决蛋白质资源短缺问题的新途径[1-3]。黑水虻幼虫饲料化营养价值取决于饲粮的物理性质和化学成分[4],虫体营养组成及生长性能在不同饲粮下变化跨度大,研究报道虫体蛋白质含量为31.7%~63.0%[5],虫体脂肪含量为7%~49%[4,6-7],个体虫重为55 ~299 mg[8-9]
碳源不仅是黑水虻幼虫饲粮的重要组成部分,同时在其幼虫生长过程中提供营养,为细胞代谢提供能量以及构成细胞物质的碳骨架,显著影响着幼虫生长发育及虫体营养组成[10]。研究表明,易生物利用碳水化合物可以被黑水虻幼虫高度消化[11],但黑水虻幼虫自身肠道没有分解纤维素的酶,中等及难生物利用碳源(如纤维素、木质素等)难以被消化[12],从而导致黑水虻幼虫饲喂效率降低并产生低价值幼虫生物量。但在易生物利用碳源基础上,添加难生物利用碳源如牛粪,可以有效改善饲粮孔隙度,平衡饲粮营养组成,为其pH稳定提供缓冲,有效提高纤维素降解率和缩短黑水虻幼虫发育周期[13]。由此可见,黑水虻幼虫饲粮营养调配必须重视黑水虻幼虫对不同碳源原料的生物可利用性。
目前,黑水虻复合饲粮配料过程中,大部分研究主要考虑总碳(TC)和总氮(TN)绝对含量的比值,但研究发现即使在同一碳氮比(C/N)条件下,由于不同生物利用度碳氮素含量的不同,虫体营养组成及幼虫生长发育也差别较大,从而导致黑水虻幼虫饲养技术应用缺乏足够的稳定性、可靠性及高效性[11,14],且目前对黑水虻饲粮的碳氮素生物可利用性有关研究相对较少。因此,本研究将葡萄糖和木屑2种不同生物利用度碳源进行不同比例混合,同时添加花生粕作为氮源并配制碳氮比为10~30的饲粮饲喂黑水虻幼虫,结合酸水解法通过正交偏最小二乘(orthogonal partial least square,OPLS)法建模与变量投影重要性指标(variable importance in projection,VIP)分析,筛选出适合黑水虻幼虫生长的饲粮碳源,并针对不同处理虫体营养成分含量变化,探究各虫体营养组成对不同生物利用度碳源含量的响应,以期为黑水虻幼虫饲粮配料指标优化以及黑水虻高效转化技术进一步开发提供理论基础和实践依据。

1 材料与方法

1.1 试验材料

供试黑水虻为3日龄幼虫,由海南省北科生态农业开发有限责任公司提供。花生粕购自海南省海口市椰海粮油交易市场,木屑为橡胶树木屑,取自定安龙湖南科食用菌有限公司基地。葡萄糖为分析纯,购自佛山西陇化工有限公司。

1.2 试验设计

试验以花生粕为氮素原料,添加由易生物利用碳源葡萄糖和难生物利用碳源木屑组成的5种不同比例碳源(100∶0、75∶25、50∶50、25∶75和0∶100),制作碳氮比分别为10、15、20、25和30,共配制25种不同碳源和不同碳氮比的黑水虻幼虫饲粮,每种饲粮3个重复,调节初始水分含量为72%,按总重量(湿重)600 g/盒平铺于饲养盒(23 cm×15 cm×15 cm),一次性加料。各处理按葡萄糖和木屑干物质(dry matter,DM)基础质量比例,分别记为P100、P75M25、P50M50、P25M75和M100,其饲粮组成及不同生物利用度碳氮组分构成见表1

1.3 黑水虻幼虫饲养管理

试验在中国热带农业科学院环境与植物保护研究所废弃物转化温控室进行。试验随机选择接入遗传背景相同、同一批次、发育正常、体尺及体重(体重约为11.33 mg/头)一致的3日龄幼虫45 000头,随机分为25个处理,每个处理3个重复,每个重复600头。在饲养盒顶部盖上100目滤布封住(防止家蝇在其中产卵和幼虫逃走),置于温度为27 ℃、相对湿度为70%的恒温恒湿饲养室。期间每隔2 d用勺子搅动饲粮,以保证黑水虻充分食用,盒中出现10只幼虫化蛹时停止试验。

1.4 检测指标

1.4.1 有机碳氮组分及碳氮素活度指标

酸水解法通过不同浓度硫酸(2.5和13.0 mol/L)将有机碳库分为2个活性程度不同的组分[不稳定碳组分1(LCP1)和不稳定碳组分2(LCP2)]和1个稳定碳组分(RCP)共3个组分,其中LCP1通常指非纤维素易生物利用多糖,LCP2主要成分是植物源纤维素,这2种组分化合物具有较高生物可利用性;而RCP主要化合物是木质素等高度耐生物降解的稳定组分化合物。同样,有机氮库也可分为3个组分[不稳定氮组分1(LNP1)、不稳定氮组分2(LNP2)和稳定氮组分(RNP)],通过各有机碳氮组分含碳(氮)量可以计算黑水虻饲粮中多种碳氮素活度指标[15-16]。计算公式如下:
碳素活度(AC)=不稳定碳组分(LCP)含量/RCP含量;
氮素活度(AN)=不稳定氮组分(LNP)含量/RNP含量;
碳素活度指数(LIC)=[LCP1含量/总有机碳(TOC)含量]×3+(LCP2含量/TOC含量)×2+(RCP含量/TOC含量)×1;
氮素活度指数(LIN)=(LNP1含量/TN含量)×3+(LNP2含量/TN含量)×2+(RNP含量/TN含量)×1;
碳素有效率(ACC,%)=100×LCP含量/TOC含量;
氮素有效率(ACN,%)=100×LNP含量/TN含量。

1.4.2 虫体常规营养成分含量

收获分离后虫体放入烘箱105 ℃灭活10 min后,在65 ℃烘干至恒重,用研钵研磨后过60目筛,-20 ℃保存备用待测。虫体DM、粗蛋白质(crude protein,CP)、粗脂肪(ether extract,EE)、粗纤维(crude fibre,CF)和粗灰分(crude ash,CA)含量分别参照国标GB/T 6435—2006、GB/T 6432—2018、GB/T 6433—2006、GB/T 6434—2006和GB/T 6438—2007测定。

1.4.3 饲粮及虫体基础理化指标

饲粮pH、电导率(EC)、饱和含水量(saturated water content,SWC)、TOC和TN等基础理化指标测定参照《土壤农化分析》[17]。其中相对含水量(RMC)计算公式如下:
RMC=100×饲粮实际含水量(MC)/SWC。

1.4.4 生长性能指标

对黑水虻幼虫和虫粪进行分离称重,分别记录黑水虻幼虫重量和数量,测定含水量,计算终末干虫体重,生长性能指标(DM基础)计算公式如下:
幼虫产率(%)=100×终末干虫体重/基质总干重;
虫体增重率(g/d)=(终末干虫体重-初始干虫体重)/试验天数;
存活率(%)=100×终末虫数量/初始虫数量;
百头虫重(g)=100×终末干虫体重/终末虫数量。

1.5 数据处理及统计分析

采用Excel 2016处理试验基础数据,并用SPSS 25.0软件及LSD法进行单因素方差分析、相关性分析和组间多重比较。试验结果以“平均值±标准误”表示,且P<0.05表示差异显著。采用SIMCA 14.1软件进行OPLS和VIP分析。

2 结果与分析

2.1 复合碳源饲粮在不同碳氮比下对黑水虻幼虫生长性能的影响

表2可知,除P100处理外,其他处理黑水虻幼虫存活率在不同碳氮比条件下达76.91%以上,均处于较高水平。与其他处理相比,P100处理在碳氮比为20时,黑水虻幼虫存活率显著骤降至17.60%(P<0.05),并随着碳氮比继续升高下降至7.60%,而其他处理存活率随碳氮比升高基本不受影响,这说明饲粮中添加葡萄糖在一定程度上导致黑水虻幼虫死亡率上升,碳源类型对存活率的影响大于碳氮比。而在相同碳源条件下,碳氮比提高,黑水虻幼虫产率、虫体增重率和百头虫重均呈逐渐下降趋势。不同类型碳源在同一碳氮比条件下,除碳氮比大于20的P100处理由于黑水虻幼虫死亡率上升导致幼虫产率下降明显外,随着难降解碳源木屑比例的增大,黑水虻幼虫产率显著降低(P<0.05)。进一步通过计算黑水虻幼虫百头虫重,排除P100处理幼虫死亡所产生干扰,在同一碳氮比条件下各处理百头虫重均为相同趋势,由大到小分别为:P100>P75M25>P50M50>P25M75>M100。由此可见,虫体增重与碳源中的易生物利用碳源比例呈正相关关系。此外,虫体增重率由大到小分别为:P50M50>P25M75>P75M25>P100>M100,说明难易碳素等比例混合饲粮虫体增重最快,难易碳素复合碳源提高了幼虫生物量积累的速率。

2.2 复合碳源饲粮在不同碳氮比下对黑水虻幼虫虫体营养成分含量的影响

表3可知,随着饲粮木屑添加比例的提高,黑水虻幼虫虫体CP、CF和CA含量在各碳氮比条件下显著提高(P<0.05),相反,虫体EE含量在各碳氮比条件下显著降低(P<0.05)。由此可知,饲粮中的易生物利用碳源葡萄糖可显著降低虫体CF含量累积,提高黑水虻幼虫饲用价值。此外,P75M25和P50M50处理虫体EE含量在碳氮比为25时较高,分别为47.88%和46.57%,均显著高于其他处理(P<0.05),说明易生物利用碳源葡萄糖显著影响虫体脂肪累积,并且碳氮比为25时更有利于虫体脂肪积累。本试验各处理虫体营养成分含量分别为CP 36.19%~61.57%、EE 12.81%~47.88%、CF 6.70%~13.95%、CA 2.96%~11.55%,黑水虻幼虫属于典型高蛋白质、高脂肪饲料资源,饲粮中不同生物利用度碳源对黑水虻幼虫虫体营养成分含量影响巨大。

2.3 基于OPLS模型分析饲粮碳源组成调控黑水虻幼虫生长性能和虫体营养成分的作用因子

2.3.1 OPLS回归模型建立

以8个黑水虻幼虫生长性能和虫体营养成分含量指标作为因变量(y),饲粮理化性质指标及碳素活度指标作为自变量(x)进行OPLS回归分析建立模型(表4)。基于原始数据建立模型(M1~M8),除M8外各模型的自变量累积解释能力参数(R2x)、因变量累积解释能力参数(R2y)和因变量预测能力参数(Q2y)均大于0.5,R2yQ2y之差小于0.2;将模型M8的CF含量原始数据进行lg转换后建立模型M8.1,各诊断参数也均达到以上所述标准。对OPLS模型进行200次置换检验,R2小于0.2,Q2为负值,未出现过拟合,这说明模型M1~M7和M8.1构建成功,具有较好的拟合度和预测能力,可用于进一步辨识饲粮中调控黑水虻幼虫生长性能和虫体营养成分的潜在作用因子。

2.3.2 VIP分析

VIP是反映自变量对因变量解释能力的重要指标,其值越大表明该自变量对因变量的解释能力越强。当VIP>0.8时,饲粮指标在影响黑水虻幼虫生长性能和虫体营养成分含量方面起主导作用。
表5可知,在52个理化性质和碳氮素活度指标参数中,RMC、LCP/LNP1、LCP/LNP2、LCP1/LNP2、LCP2/LNP2、RCP/LNP2、TC/LNP、TC/LNP1和TC/LNP2共9个参数对黑水虻幼虫所有生长性能和虫体营养成分含量指标VIP值均大于0.8,表明其对黑水虻生长性能和虫体营养成分含量影响的贡献最为显著,可优先考虑作为饲粮配料指导指标。此外,不稳定碳组分(LCP1、LCP2)、碳素有效率(ACC、ACC1和ACC2)对总增重率影响不显著,不稳定氮组分(LNP1、LNP2)对黑水虻幼虫所有指标影响均不显著,说明黑水虻幼虫的生长性能和虫体营养成分含量受碳氮组分综合影响,仅单一考虑饲粮碳源或氮源本身活度特性并不全面。不仅如此,总碳氮素含量(TOC、TN)、总碳氮比(TC/TN)以及各碳组分与TN之比(LCP1/TN、LCP2/TN、LCP/TN、RCP/TN)同样对所有黑水虻幼虫指标影响不显著。由此可见,传统黑水虻养殖配料指标基于TC/TN以及TN含量作为黑水虻转化配料指导指标存在一定局限性,在黑水虻养殖配料中不建议作为主要参考指标。

2.4 调控黑水虻幼虫生长性能和虫体营养成分含量作用因子相关性分析

为进一步提出简化实用的配料指导指标,通过对显著影响幼虫生长性能和虫体营养成分含量的9个参数指标进行相关性分析(表6)发现:5个活度指标TC/LNP1、TC/LNP2、TC/LNP、LCP/LNP1、LCP/LNP2之间相关系数均接近为1.00,呈极显著相关,其中TC/LNP1、TC/LNP2、TC/LNP也与另外3个活度指标LCP1/LNP2、LCP2/LNP2、RCP/LNP2相关性较高,相关系数为0.67~0.70。这8个碳氮素活度参数可认为是一个相关性很高,能够代表碳氮素营养的指标群(记为“指标群Ⅰ”)。此外,RMC与TC/LNP1、TC/LNP2、TC/LNP、LCP/LNP1、LCP/LNP2等5个活度指标无显著相关性,与LCP1/LNP2、LCP2/LNP2、RCP/LNP2等3个活度指标的相关性较弱。因此,RMC和“指标群Ⅰ”中任一指标组合,可作为黑水虻生物转化配料指导指标。

3 讨论

3.1 不同生物利用度复合碳源饲粮对黑水虻幼虫生长性能的影响

本试验结果表明,饲粮中添加葡萄糖显著影响黑水虻幼虫虫体的存活率和生物量产量。当碳氮比在20以下时,P100处理幼虫产率和百头虫重(P75M25处理除外)显著高于其他处理,这与Cheng等[18]和Lalander等[19]研究黑水虻幼虫转化不同含水率餐厨垃圾结果类似。当碳氮比为20及以上时,P100处理黑水虻幼虫存活率显著降低至低于20%,幼虫产率接近或低于5%。这可能与葡萄糖的水溶性有关,由于葡萄糖为溶解性有机物,溶于基质水分后使总体RMC升高,基质透气性降低,导致黑水虻幼虫存活率下降,由此可见饲粮具有良好的透气性,有利于提高幼虫存活率。Barros等[20]研究指出,过高含水量会使基质失去透气性结构,从而导致黑水虻幼虫虫体前后两端的气门堵塞,呼吸困难。此外,本试验中复合碳源饲粮虫体增重率较快,显著高于单一类型碳源处理,其中以等比例复合碳源饲粮P50M50处理虫体增重率最快。Kinasih等[7]研究表明,黑水虻幼虫能够在含有难生物利用碳源的饲粮中,通过分泌激素调控自身摄取更多的食物,以此来弥补较低的食物品质,从而快速获得所需的特定营养使虫体增重率加快。本研究结果发现,黑水虻幼虫生长性能主要受易生物利用碳源及蛋白质含量的影响,葡萄糖添加比例越高,黑水虻幼虫生物量产量越高;碳氮比越低,蛋白质含量越高,幼虫生物量产量同样显著升高。上述结果与Tschirner等[21]报道结果类似,表明饲粮中添加简单碳水化合物和蛋白质更有利于黑水虻生长。因此,为快速获得较高生物量产量,选择易生物利用碳源作为外加碳源,饲粮碳氮比应小于20;而选择复合碳源时,难生物利用碳源占比不宜超过25%。

3.2 不同生物利用度复合碳源饲粮对黑水虻幼虫虫体营养成分含量的影响

黑水虻幼虫虫体营养组成高度依赖于饲粮理化性质,因此其饲用价值也随着饲粮的不同而改变。本试验中,饲粮中添加葡萄糖可以提高虫体EE含量,在同一碳氮比条件下P100处理虫体EE含量最多提高了27.54个百分点,且最高值高于黑水虻转化蔬菜与水果基质试验的虫体EE含量[7]。Pliantiangtam等[22]和Meneguz等[5]研究指出,昆虫具有将易生物利用碳源转化为自身脂肪的能力,饲粮中过量的碳水化合物可被转化为脂肪并储存在体内,这有助于虫体储存一定非吸收水分,同时可用于成虫后能量消耗。此外,M100处理虫体CP含量在各碳氮比条件下均大于50%,但虫体生物量产量、个体虫重及虫体EE含量较低,因此高比例的难生物利用碳源不利于黑水虻幼虫的生长发育及虫体营养组成。Tschirner等[21]通过蛋白质饲粮、纤维饲粮及混合饲粮等3种营养不同的饲粮开展黑水虻幼虫饲喂试验,结果与本试验结果一致。此外,本试验中,饲粮中添加葡萄糖降低了虫体CA和CF含量,除P25M75和M100处理外,其他处理虫体CA含量均低于5.6%。Jucker等[8]在黑水虻幼虫饲粮中添加水果,虫体CA含量为5.6%,这比Barroso等[23]报道通过标准饲粮饲喂的虫体CA含量降低了约4个百分点,与本研究结论一致。Gao等[24]研究表明,黑水虻幼虫饲喂标准饲粮,虫体可作为优质的畜牧及水产养殖饲料,其CP含量为42.74%,EE含量为34.26%,CA含量为8.28%。本试验中,P100、P75P25及P50M50处理(碳氮比小于20)黑水虻幼虫虫体CP含量为42.77%~49.51%,EE含量为35.58%~44.60%,CA含量为2.96%~4.82%,该3种处理虫体营养组成均衡,可作为优质饲料原料[25],结合幼虫生长性能,P100(碳氮比小于20)与P75P25处理表现更为突出。

3.3 饲粮配料指标与黑水虻幼虫生长性能、虫体营养成分含量的相关性分析

众所周知,传统黑水虻饲粮配料主要考虑碳氮比[26],本研究表明,在相同碳氮比条件下,不同饲粮处理黑水虻幼虫的生长性能和虫体营养组成差别较大,通过OPLS建模与VIP分析也进一步证明了饲粮的TC、TN及TC/TN并不能显著影响幼虫生长性能和虫体营养组成。而RMC、LCP/LNP1、LCP/LNP2、LCP1/LNP2、LCP2/LNP2、RCP/LNP2、TC/LNP、TC/LNP1和TC/LNP2等9个饲粮参数指标,为影响幼虫生长性能和虫体营养成分含量的主要因素。这9个参数指标(RMC和指标群Ⅰ),可以分为物理指标和化学指标2类,其中化学指标(指标群Ⅰ)均为不稳定碳氮组分的比值。Gold等[11]和Liu等[12]研究也指出,原料中非纤维碳水化合物(NFC)和易生物利用氮源蛋白质是影响幼虫生长发育和虫体CP、EE含量的关键因素。Lalander等[27]报道,果蔬中的高含量易生物利用碳源平衡了屠宰场废肉的高含量易生物利用氮含量,使混合饲粮营养趋于平衡。物理指标RMC体现黑水虻幼虫饲粮的保水透气性,其显著影响黑水虻幼虫的存活率及幼虫生长发育。本试验在相同含水量(72%)条件下,黑水虻幼虫存活率跨度从7.60%至99.10%。Lalander等[27]和Chen等[28]也报道了以餐厨垃圾和畜禽粪便为饲粮,即使饲粮含水量相同为85%,但RMC不同,黑水虻幼虫存活率同样跨度较大,分别为94.30%和28.00%,由此可见黑水虻幼虫饲粮配料以绝对含水量为配料指标存在不稳定性。本研究中,饲粮RMC大于139%时黑水虻幼虫存活率骤降,这与Bekker等[29]报道的使用鸡饲料为饲粮的RMC大于121%时幼虫无法存活的结果相近,说明过高RMC抑制了黑水虻幼虫的存活与生长发育。这是由于当RMC过高时,基质中观察存在可见水,一方面,导致基质缝隙空气被水填充,底部黑水虻幼虫呼吸困难;另一方面,上方黑水虻幼虫浮于表面,能量来源被切断,最终使黑水虻幼虫存活率显著降低[28,30]。不过,目前对于黑水虻幼虫饲粮配料研究较少,幼虫生长发育最适RMC仍无法确定。此外,Tschirner等[21]研究指出,基质透气性好,黑水虻幼虫存活率高,且基质保水透气性与难生物利用碳源有关。

4 结论

① 黑水虻幼虫饲粮添加复合碳源,难生物利用碳源占比不应超过25%。
② 黑水虻幼虫饲粮选择易生物利用碳源作为外加碳源,碳氮比应控制在20以下且RMC低于139%。
③ OPLS建模和VIP分析表明,RMC和TC/TN在黑水虻饲粮配料中存在一定局限性,RMC和有效的碳氮素比例显著影响黑水虻幼虫生长性能和虫体营养组成,在黑水虻饲粮配料时更应被关注。
