Effects of Rumen Protected Fat on Body Weight, Blood Indexes and Musk Production Performance of Forest Musk Deer

Abstract

This experiment was conducted to investigate the effects of diet supplemented with rumen protected fat on body weight, blood indexes and musk production performance of forest musk deer. Sixty-three healthy male musk deer [body weight: (8.0±0.5) kg] aged 2 to 10 years old were divided into 7 groups with 9 replicates per group and 1 musk deer per replicate. The control group (group A) was fed the basal diet, and the experimental groups were fed the basal diet supplemented with 4 (group B1, short-term), 4 (group B2, long-term), 8 (group C1, short-term), 8 (group C2, long-term), 12 (group D1, short-term) and 12 g/d (group D2, long-term) rumen protected fat, respectively. The experiment lasted for 121 days in the short term groups (groups B1, C1 and D1) and 195 days in the control group and long term groups (groups B2, C2 and D2), containing a 15-day pre-trial period. The results showed as follows: 1) at the end of the experiment, the body weight of forest musk deer in each experimental group was higher than that in the control group, but the difference was not significant (P>0.05). 2) The dry weight of musk in group C2 was increased significantly with the control group and groups B1, B2 and D2 (P<0.05), but there were no significant effects on the fresh weight, the contents of moisture and musk ketone of musk (P>0.05). 3) There were no significant differences in the contents of serum insulin, testosterone, total protein, albumin, triglyceride, total cholesterol, glucose and urea among all groups (P>0.05). Serum non-esterified fatty acids ( NEFA) content in groups B2, C1, D1 and D2 were significantly decreased compared with the control group (P<0.05). In conclusion, under the condition of this experiment, diet long-term supplemented with 8 g/d rumen protected fat can increase the body weight and dry musk yield of forest musk deer.

1958年,国务院提出“变稀有名贵中药材为家种家养”的指示精神,先后在四川省的马尔康、米亚罗,安徽省的佛子岭,陕西省的镇平等地建立了养麝试验场。由于“杀鸡取卵”式的盗猎活动和生境变化等因素,野生麝的数量急剧减少,我国麝资源总量已从20世纪50年代的二三百万头迅速下降至现在的不足十万头,分布范围急剧缩减,且基本为零散的“岛屿”状分布。2002年国家将麝类的保护级别调整为国家Ⅰ级。数据显示,我国人工繁育麝数量截至2020年约3万只,且以林麝为主。截至2021年底,四川省共有人工繁育林麝5 588只、马麝230只。我国天然麝香需求量为每年1 000 kg左右,但目前人工养麝年产麝香不足200 kg[1],远不能满足市场需求,如何提高麝香单产量是养麝业亟待解决的问题。麝香是四大名贵动物药材之一,它是成熟雄性麝科动物林麝(Moschus berezovskii Flerov)、马麝(Moschus sifanicus Przewalski)和原麝(Moschusmoschiferus Linnaeu)香囊分泌物的熟化品[2],其中以林麝所产麝香质量最优。林麝属偶蹄目麝科,主要栖于针阔混交林,也在针叶林和郁闭度较差的阔叶林的生境生活,是林栖类小型反刍动物。每年的5—7月份为林麝的泌香期,麝香分泌期是能量的消耗期,此期也是林麝的生理绝食期,如果此期能量不能满足泌香的能量需要,动物就会动用体组织,特别是体脂来供给能量,从而导致能量负平衡,引起代谢机能障碍,甚至疾病,因此前期的能量储备对于麝香的正常分泌至关重要。麝最初分泌的麝香是液体,液态麝香经过一系列生化反应生成成熟的麝香,因此,麝香熟化期的营养供给是麝香质量的重要保证。过瘤胃脂肪作为一种高能值能量饲料,在瘤胃中不易降解,而在瘤胃后消化系统被消化吸收后作为能量被动物利用,不影响瘤胃内环境的稳定,能防止因精料采食过多而引起的瘤胃酸中毒等问题,可以供给动物所需能量,促进动物健康、提高反刍动物生产性能、饲料利用率、改善产品质量等[3⇓-5]。研究发现,高能饲粮能提高育成雄麝体增重和干麝香产量,改善被毛光泽[6]。过瘤胃脂肪在麝中的应用鲜有报道,本团队前期对过瘤胃脂肪对林麝的表观消化率和血液指标的影响进行了研究,发现添加10 g/d过瘤胃脂肪提高了饲粮林麝粗脂肪和能量的表观消化率及血清睾酮的含量[7-8]。睾酮可以刺激麝香腺分泌香液,并形成成熟麝香[9⇓-11],但过瘤胃脂肪对林麝产香的影响未见报道。本试验在前期研究的基础上,收集林麝麝香、血液生化指标等数据,研究过瘤胃脂肪对林麝麝香产量和质量的影响,以及过瘤胃脂肪在林麝生产中的适宜添加时期和添加量,为林麝的正常生长发育、麝香产量的提高、预防林麝营养性疾病的发生、促进人工养殖林麝的发展打下基础,为林麝营养需要的研究和饲养标准的制定提供数据。

1 材料与方法

1.1 试验材料

试验所用过瘤胃脂肪是由棕榈脂肪分馏物加工而成的纯植物源脂肪粉,为自由流动的白色颗粒状,粗脂肪含量大于99%,是一种高熔点的饱和脂肪,有效能含量高,易消化,不含载体。

1.2 试验动物及试验设计

由于林麝泌香期一般集中在每年的5—7月份,本试验从泌香前1月开始饲喂,短期试验喂至泌香期结束,长期试验喂至取香。试验在四川养麝研究所马尔康养獐场进行,选取63头2~10岁体况良好的雄性林麝[体重:(8.0±0.5) kg],随机分为7个组,每组9个重复,每个重复1头林麝。对照组(A组)饲喂基础饲粮,试验组分别饲喂在基础饲粮中添加4(B1组,短期;B2组,长期)、8(C1组,短期;C2组,长期)、12 g/d(D1组,短期;D2组,长期)过瘤胃脂肪的试验饲粮。该试验经四川养麝研究所实验动物伦理委员会批准后开展(批准号:SCYS-E-2018006),短期组(B1、C1、D1组)试验期121 d(2018年4—7月份),对照组和长期组(B2、C2、D2组)试验期195 d(2018年4—10月份),其中预试期15 d。

1.3 试验饲粮及管理

试验所用基础饲粮为养殖场日常饲养用饲粮,由多汁料、精补料和干树叶组成,其中多汁料(南瓜+莴笋,鲜重基础)与精补料(风干基础)按照5∶1混合均匀,日喂量600 g;干树叶日喂量200 g(风干基础)。试验麝单圈饲养,专人负责,每日08:00和16:00各饲喂1次,先饲喂干树叶,再饲喂多汁料+精补料,自由饮水。基础饲粮组成及营养水平见表1。

1.4 样品采集与测定

1.4.1 体重

分别于试验前1 d和试验最后1 d早饲前对参试麝进行空腹称重,记录体重数据。

1.4.2 饲料养分含量

粗蛋白质含量参照GB/T 6432—2018采用凯氏定氮法测定,粗脂肪含量参照GB/T 6433—2006采用索氏抽提法测定,粗纤维含量参照GB/T 6434—2006采用过滤法测定,粗灰分含量参照GB/T 6438—2007测定,钙含量参照GB/T 6436—2018测定,磷含量参照GB/T 6437—2018测定,总能参照SCWT/QM03-L-37方法采用ZDHW-3全自动量热仪测定。

1.4.3 血液指标

正试期最后1 d早饲前空腹采集林麝静脉血5 mL,2 540×g离心15 min制备血清,于-20 ℃低温冰柜保存待测。血清生化指标采用BS-190全自动生化分析仪(深圳迈瑞生物医疗电子股份有限公司)测定;采用酶联免疫吸附测定(ELISA)方法测定血清中胰岛素、睾酮和非酯化脂肪酸含量,测定仪器为SpectraMax iD3多功能酶标仪(美国Molecular Devices公司)。

1.4.4 产香性能

在试验期的最后2 d将林麝保定后采集参试林麝的麝香样品并称重,分别装入带盖塑料盒中,冷藏保存,备测麝香酮含量。麝香酮含量采用《中华人民共和国药典》(2015年版一部)[12]方法测定,测定仪器为Agilent7980A气相色谱仪,对照品麝香酮由中国食品药品检定研究院生产,纯度为99.9%。

1.5 数据处理及统计分析

试验数据使用Excel 2013整理后,均使用SPSS 22.0统计软件进行单因素方差分析,并采用Duncan氏法进行多重比较,显著性水平为P<0.05。试验结果以平均值±标准误表示。

2 结果与分析

2.1 过瘤胃脂肪对林麝体重的影响

由表2可知,试验前各组林麝体重差异不显著(P>0.05);饲粮添加过瘤胃脂肪后,各试验组林麝体重较对照组均有所提高,但差异不显著(P>0.05),其中C1、C2和D1组林麝体重分别较对照组提高了16.67%、16.67%和14.33%;试验后各试验组林麝的体重均较试验前有所提高,而对照组林麝的体重较试验前有所降低(P>0.05)。

2.2 过瘤胃脂肪对林麝血液指标的影响

由表3可知,饲粮添加过瘤胃脂肪对林麝血清胰岛素和睾酮含量无显著影响(P>0.05);B2、C1、D1、D2组血清非酯化脂肪酸含量显著低于对照组(P<0.05),与其他组差异不显著(P>0.05);血清总蛋白、白蛋白、甘油三酯、总胆固醇、葡萄糖、尿素含量各组间差异不显著(P>0.05)。

2.3 过瘤胃脂肪对林麝产香性能的影响

由表4可知,饲粮添加过瘤胃脂肪对林麝麝香鲜重、水分和麝香酮含量没有显著影响(P>0.05);C2组麝香干重显著高于对照组以及B1、B2和D2组(P<0.05),与其他组差异不显著(P>0.05)。

3 讨论

3.1 过瘤胃脂肪对林麝体重的影响

脂肪能提供动物生长和生产所需的能量。研究表明,饲粮中添加过瘤胃脂肪能提高动物的体增重。肥育动物需要足够的能量来维持其基本活动和生长肥育,脂肪作为一种营养型能量添加剂,能为机体提供能量,并促进机体对其他营养物质的消化利用[13⇓-15]。研究表明,饲粮中添加保护性脂类可以提高动物的日增重、日采食量、屠宰率和饲料转化率[16⇓⇓-19]。水牛以臂形草为基础饲粮时,添加精料和4%的过瘤胃脂肪提高了日增重和干物质采食量[20]。在限制精料饲喂的断奶南江黄羊羔羊精料中添加1%~2%的植物脂肪粉,其胴体质量显著提高,获得了较好的饲养效益[21]。过瘤胃脂肪能提高干物质采食量和饲粮中有机物、脂肪和酸性洗涤纤维等营养物质的消化率和利用率,增加微生物氮流量,改善氮平衡,从而提高增重。也有一些研究表明,饲粮中添加脂肪对动物体重没有显著影响[22⇓-24]。Manso等[25]报道,在羔羊饲粮中添加棕榈油和棕榈油脂肪酸钙皂,羔羊的平均日增重不受处理的影响,但饲料转化率得到提高。在12~15月龄的Beetal羊饲粮中添加过瘤胃脂肪对其平均日增重没有显著影响,干物质采食量随过瘤胃脂肪添加量的增加呈二次曲线变化[26]。对奶牛的研究发现,添加过瘤胃脂肪对其采食量和体重没有显著影响,但提高了产奶量[27]。本试验中,添加过瘤胃脂肪后,各试验组林麝体重较对照组均有所增加,其中C1、C2和D1组林麝体重分别较对照组提高了16.67%、16.67%和14.33%,且各试验组林麝体重较试验前均有不同程度的提高,而对照组林麝体重较试验前降低,说明饲粮中添加过瘤胃脂肪能满足林麝对能量的需求,避免了体内贮存能量的动员,对于林麝体重的维持有很好的作用。本团队前期研究发现,饲料中添加过瘤胃脂肪能提高林麝的粗脂肪和能量表观消化率,有提高粗蛋白质表观消化率的趋势[7],促进了林麝对营养物质的吸收利用,有效保证了林麝的体况,为后续的生长和生产做准备。对照组林麝体重降低可能是由于在泌香前的营养储备不足以满足泌香期的营养需要,导致动用体脂供能,加之后期能量供给不能满足需要,这有待进一步验证。而与前人研究结果的不同,可能是由于研究所使用的试验动物、过瘤胃脂肪的类型、饲粮结构和生理阶段不同所致。

3.2 过瘤胃脂肪对林麝血液指标的影响

机体功能和营养代谢情况可以通过血清生化指标反映。葡萄糖是机体各组织细胞新陈代谢所需能量的主要提供者。多项研究结果表明,过瘤胃脂肪的添加对动物血液中葡萄糖含量的影响不显著[17,24,28⇓-30]。本试验中,饲粮添加过瘤胃脂肪后,各组间血清葡萄糖含量差异不显著,与前人研究结果一致。血清葡萄糖含量是血糖是否稳定的反映,并不能反映某一时刻机体葡萄糖的生成量,也不能反映葡萄糖从循环血液中的消失量[31]。因此,尽管血清葡萄糖含量无显著变化,但并不意味着添加过瘤胃脂肪对机体葡萄糖的代谢没有影响。Baumgard等[32]的研究结果表明,补充脂肪可以提高磷酸烯醇式丙酮酸羧激酶和丙酮酸羧化酶mRNA的表达量,提示脂肪的添加可以促进机体葡萄糖的合成。
作为机体蛋白质分解代谢主要产物之一的尿素,可以反映饲粮中蛋白质的利用情况和机体蛋白质的代谢水平[33]。血清总蛋白包括白蛋白和球蛋白,可以维持血管内胶体渗透压、血液酸碱度和运输功能。研究发现,补充瘤胃保护性脂肪后,奶牛对饲粮中各种氨基酸的摄入量、瘤胃氨基酸的通过量和进入十二指肠的总氮量未发生显著变化[34]。本试验结果发现,血清总蛋白、白蛋白和尿素含量不受过瘤胃脂肪的影响,表明饲粮中添加过瘤胃脂肪对林麝体内蛋白质和氨基酸代谢没有产生显著影响,这与张克春等[28]和Kang等[24]的研究结果一致。
非酯化脂肪酸是体脂肪分解的中间产物,当机体能量需求增加而肝糖原耗尽时,会分解体脂为机体供能,从而产生非酯化脂肪酸,血清中非酯化脂肪酸的含量与脂类代谢、糖代谢和内分泌功能有关。已有研究表明,饲粮中添加脂肪可以降低试验动物的血清非酯化脂肪酸含量[35⇓⇓⇓-39]。而张克春等[28]和Bines等[40]的研究发现,饲粮中补充瘤胃稳定性脂肪会引起血液中非酯化脂肪酸含量显著升高。Haddad等[22]研究发现,饲粮中添加过瘤胃脂肪对绵羊血清中非酯化脂肪酸含量没有显著影响。本研究中,在添加过瘤胃脂肪后,B2、C1、D1、D2组血清非酯化脂肪酸含量较对照组显著降低,各试验组间差异不显著,过瘤胃脂肪的添加减少了机体体脂的动员,从而降低了血清非酯化脂肪酸含量。与已有研究结果的不同可能是由于所使用的试验动物、脂肪类型、添加量和添加阶段、饲粮类型不同所致。
甘油三酯主要在肠黏膜上皮细胞和肝脏中合成,可以反映机体对脂肪的利用情况,其含量与机体对脂肪的利用率成反比[41]。胆固醇是动物机体中一类最重要的固醇类化合物,而血清中的胆固醇主要由肝脏合成,其次来自饲料。已有研究发现,饲粮中添加过瘤胃脂肪可以提高血清甘油三酯和胆固醇含量[17,42-43]。也有研究发现饲粮中添加过瘤胃脂肪对血清甘油三酯和胆固醇含量没有显著影响[24,44]。Ghasemi等[30]研究发现,不同形式的脂肪均提高了荷斯坦犊牛血浆胆固醇和高密度脂蛋白含量,但对甘油三酯含量产没有生显著影响。王哲文等[45]在牦牛饲粮中添加过瘤胃脂肪,随着添加水平的提高,血清甘油三酯含量呈线性降低。本试验发现,过瘤胃脂肪的添加对林麝血清甘油三酯含量的影响不显著,各试验组血清总胆固醇含量较对照组有提高趋势,其中D2组血清总胆固醇含量较对照组提高了33.94%,说明过瘤胃脂肪的添加对脂肪利用率没有产生影响,而D2组长期高剂量过瘤胃脂肪的添加促进了胆固醇的吸收和肝脏胆固醇的合成,进而提高了血清总胆固醇含量。
胰岛素是一种多肽类激素,可以促进血糖转化为糖原、蛋白质和脂肪等营养物质,并促进乳脂肪和乳蛋白相关基因表达[46-47]。对牦牛的研究发现,添加2.4%和3.6%过瘤胃脂肪组牦牛血清胰岛素含量显著高于对照组和添加1.2%过瘤胃脂肪组[46]。睾酮是生殖功能、内分泌稳态和能量调节的重要调节剂,它的失衡会导致血脂代谢异常、能量代谢失调及肥胖的发生。本研究发现,添加过瘤胃脂肪有升高林麝血清胰岛素和睾酮含量的趋势,但差异不显著,说明添加过瘤胃脂肪能刺激胰岛素和睾酮分泌,促进林麝脂肪合成。

3.3 过瘤胃脂肪对林麝产香性能的影响

动物可以将脂肪酸直接沉积到产品或体组织中,减少饲粮中碳水化合物转变成脂肪酸的很多中间代谢过程。乙酸氧化是反刍动物的重要能量来源,但乙酸氧化产生ATP的效率比长链脂肪酸低,因此,长链脂肪酸对于提高反刍动物氧化代谢效率具有重要意义。林麝的泌香期集中在每年的5—7月份,泌香是一个能量的消耗过程,同时泌香盛期也是林麝生理性采食减少甚至废绝的时期,一般为3~7 d。因此,泌香前期的能量储备对于泌香活动尤为重要。泌香期分泌的初香液经过后期一系列复杂的生化过程转变为成熟的麝香。麝香中除了含有芳香性成分麝香酮以外,还有胆固醇及其酯类、长支链脂肪酸酯类以及多种C19甾体雄性激素存在。此外,麝香中蛋白质的含量高达25%,其所含肽类和氨基酸是麝香抗炎的主要成分[48]。添加过瘤胃脂肪可以提高血浆尿素氮含量[49],促进蛋白质代谢沉积,提高营养物质的消化率,为泌香和香液熟化提供所必需的营养物质和能量。程世国等[6]对林麝能量需要量的研究发现,高能水平饲粮组林麝产香量最高。本研究中,随着过瘤胃脂肪添加水平的提高,麝香产量呈现出先上升后下降的趋势,可能是饲粮添加过瘤胃脂肪后提高了林麝对饲料的转化效率,给林麝提供了麝香合成分泌及熟化所需的能量,使得沉积到麝香中的营养物质增加,从而提高了麝香产量,而长期高剂量添加过瘤胃脂肪可能造成了代谢负担,使得麝香产量又出现下降,添加过瘤胃脂肪提高麝香产量的具体机制还有待进一步研究。
此外,有研究表明,林麝的泌香反应过程受性腺轴激素调控。毕书增等[50]研究发现,下丘脑-垂体-性腺轴激素能诱导林麝的泌香反应,且雄性激素刺激出现泌香反应所需的时间最短。洪沂生等[51-52]研究发现,给正常林麝注射外源睾丸酮可显著提高泌香量,并且,注射外源雄激素可以刺激切除生殖器的雄麝、非泌香期的雄麝和成年后无法自然泌香的雄麝重新正常分泌麝香。白康等[53]研究表明,非泌香期林麝粪样中睾酮的含量较低,进入泌香初期,粪样中睾酮含量急剧升高,并在泌香盛期达到峰值。Fan等[54]研究揭示,性激素在泌香早期起着决定麝香成分的作用。综合以上研究结果可知雄激素是调节林麝泌香的关键因素,并且林麝产香量与睾酮含量呈正相关(r=0.159)[55]。本试验中,饲粮中添加过瘤胃脂肪对林麝麝香鲜重、水分和麝香酮含量没有显著影响,C2组麝香干重高于对照组以及B1、B2和D2组,这可能是过瘤胃脂肪的添加提高了血清中胰岛素和睾酮含量,刺激麝香的分泌,从而提高了麝香产量。

4 结论

饲粮中添加过瘤胃脂肪对林麝麝香鲜重、麝香酮含量以及血清胰岛素、睾酮、总蛋白、白蛋白、甘油三酯、总胆固醇、葡萄糖、尿素含量均无显著影响,饲粮中添加8 g/d过瘤胃脂肪时林麝体重增加,长期添加还可显著提高麝香产量,而短期添加则会显著降低血清非酯化脂肪酸含量。综合以上试验结果,本试验条件下,林麝饲粮中过瘤胃脂肪的适宜添加水平为8 g/d,且宜在泌香期前1个月以上开始添加。
