Isolation, Identification and Safety Evaluation of Probiotic Bacillus Derived from Pigeons

Abstract

This study aimed to isolate and screen a probiotic Bacillus strain derived from pigeons with excellent performance and safety, so as to provide strain resources for the development of probiotic additives derived from pigeons. Fresh fecal samples and intestinal contents from healthy Columba were collected. The strains were isolated and identified by methods of bacterial isolation culture, staining microscopic examination, biochemical identification and molecular biological identification. Subsequently, the in vitro probiotic properties of strains, including acid tolerance, bile salt tolerance, growth curve, hemolytic activity, drug sensitivity and antibacterial activity, were detected to preliminarily assess the in vitro probiotic potential of isolated strains. Finally, the in vivo safety of isolated strains was evaluated through acute and subacute toxicity tests in mice. The results showed that: 1) three Bacillus strains (F21, F31 and F54) were isolated from fresh fecal samples and intestinal contents of Columba. After biochemical identification and 16S rRNA sequencing analysis, they were all Bacillus subtilis, respectively. 2) The survival rates of three isolated strains were all above 60% under pH 2.0 conditions and above 70% under bile salt concentration of 0.3%; all three isolated strains could enter the logarithmic growth phase within 2 to 4 hours and the stationary phase after 12 to 16 hours; none of the three isolated strains had hemolytic activity; the order of drug sensitivity of three isolated strains was F31>F21>F54; both F21 and F31 strains had inhibitory effects on three common pathogenic bacteria (Staphylococcus aureus, Escherichia coli and Salmonella). 3) Bacillus subtilis F31 with better in vitro probiotic performance was selected and its in vivo safety was evaluated. In the acute toxicity test (7 d), the mice showed normal physical signs and no obvious pathological changes were observed in the dissected organs. In the subacute toxicity test (28 d), compared with the control group, the body weight of mice in the middle-dose group (1.1×109 CFU/mL, 0.2 mL/mouse) increased significantly (P<0.05), the liver index of mice in the low-dose group (1.1×108 CFU/mL, 0.2 mL/mouse) decreased significantly (P<0.05), the spleen index of mice in the high-dose group (1.1×1010 CFU/mL, 0.2 mL/mouse) increased significantly (P<0.05), and the numbers of white blood cells, lymphocytes, monocytes and neutrophils in blood of mice in the low-dose group decreased significantly (P<0.05). In conclusion, Bacillus subtilis F31 has good in vitro probiotic potential and favorable in vivo safety, which can be used as a candidate strain for probiotic preparations derived from pigeons for subsequent development and application.

鸽子已成为我国第四大家禽[1],在鸽子养殖过程中,滥用抗生素会导致细菌耐药性的产生及抗生素残留等问题,甚至危害公共卫生安全,因此,对抗生素替代品的研究显得尤为重要。常见的抗生素替代品有酶制剂、酸化剂和益生菌制剂等[2],其中,益生菌制剂因具有促进宿主代谢、抵抗病原菌感染、增强宿主免疫力等功能而被广泛关注[3-4]。益生菌制剂作为当前畜禽业养殖中替代饲用抗生素的制剂之一,在饲用效果、安全性等方面均有显著优势。芽孢杆菌是益生菌制剂中的常用菌种[5],可以用于预防或治疗细菌性疾病以及提高畜禽生长性能[6-8]。研究发现,在饲粮中添加枯草芽孢杆菌能够有效抑制肉鸽肠道中有害菌群的生长,具有较强的抗菌作用[9];还能够提高肠道内容物和肠上皮细胞刷状缘酶活性,增强肠道对营养物质消化利用的能力[10];并能提高乳鸽的免疫力和抗病力[11-12]。同源益生菌由于天然存在于宿主防御系统中,更容易在原生环境中发挥益生作用[13],且对致病菌的拮抗能力更强[14]。目前关于鸽源益生性芽孢杆菌的相关研究较少,因此分离筛选出益生性能优良的鸽源益生性菌种愈发重要。本试验采集了新疆喀什某规模化鸽厂健康白羽王鸽的新鲜粪便及肠道内容物,对其进行益生性芽孢杆菌的分离鉴定,通过体外益生潜能筛选优秀菌株并对其进行小鼠体内安全性试验,旨在筛选出益生性能优良且安全的鸽源益生性芽孢杆菌,为鸽源益生菌添加剂的研发提供菌种资源。

1 材料与方法

1.1 试验材料

1.1.1 样品采集和试验动物

无菌采集18只健康白羽王鸽的新鲜粪便样品36份及肠道内容物18份,共54份样品。采集后,将上述样品放入无菌的4 mL LB肉汤培养基中,4 ℃保存并运回实验室。试验动物为84只昆明小鼠,公母各占1/2,体重(24±2) g,购自新疆医科大学实验动物中心,动物合格证号为:SCXK2018-0002。动物试验方案经新疆农业大学伦理委员会批准,批准号为2017013。

1.1.2 主要试剂及供试菌株

LB琼脂培养基、LB肉汤培养基、HBI芽孢杆菌生化鉴定条、绵羊血平板购自青岛海博生物技术有限公司;牛胆盐、革兰氏染色液购自北京索莱宝科技有限公司;细菌药敏试纸片购自杭州滨和微生物试剂有限公司;4%多聚甲醛、中性树胶为Biosharp品牌产品;石蜡购自上海懿洋仪器有限公司;金黄色葡萄球菌、牛源大肠杆菌K99、鼠伤寒沙门氏菌菌株由新疆农业大学临床产科实验室提供。

1.2 试验方法

1.2.1 芽孢杆菌的初筛

1.2.1.1 芽孢杆菌的培养及形态学观察

参照孟维珊等[15]的方法,用灭菌生理盐水将样品稀释,80 ℃水浴15 min后均匀涂布于LB琼脂培养基上,37 ℃培养36 h,挑取菌落形态一致的单菌落于LB平板上进行纯化,纯化3次后于LB肉汤培养基中增殖培养,对分离菌株进行革兰氏染色后镜检及观察。

1.2.1.2 生化鉴定

取分离菌株接种至HBI芽孢杆菌生化鉴定条,根据说明书进行操作,观察并记录结果。

1.2.1.3 分子生物学鉴定

将纯化好的菌株送至生工生物工程(上海)股份有限公司进行16S rRNA测序,测序结果在NCBI上进行BLAST比对。

1.2.2 分离菌株的体外益生性能筛选

1.2.2.1 耐酸能力试验

将磷酸盐缓冲液(PBS)的pH分别调整至7.0、5.0、3.0、2.0,高压灭菌。取1 mL浓度为1×108 CFU/mL的分离菌株菌悬液,离心后弃上清加入上述PBS,振荡培养2 h后,取50 μL菌悬液接种于5 mL LB肉汤培养基中,37 ℃培养12 h后测定其在600 nm处的吸光度(OD600 nm)值[16]。每种分离菌株进行3次重复试验。按照如下公式计算分离菌株的存活率:

存活率(%)=100×Nx/N0。

式中:Nx为pH为2.0、3.0或5.0时的OD600 nm值;N0为pH为7.0时的OD600 nm值。

1.2.2.2 耐胆盐能力试验

将PBS的胆盐浓度分别调整至0、0.1%、0.2%、0.3%,高压灭菌。取1 mL浓度为1.1×108 CFU/mL的分离菌株菌悬液,离心后弃上清加入上述PBS,振荡培养2 h后,取50 μL菌悬液接种于5 mL LB肉汤培养基中,37 ℃培养12 h后测定OD600 nm值[17]。每种分离菌株进行3次重复试验。按照如下公式计算分离菌株的存活率:

存活率(%)=100×Ny/N0。

式中:Ny为胆盐浓度为0.1%、0.2%或0.3%时的OD600 nm值;N0为胆盐浓度为0时的OD600 nm值。

1.2.2.3 生长曲线测定

取分离菌株菌悬液,以2%的接种量接种于LB肉汤培养基中,37 ℃培养28 h,从0 h开始每隔2 h测1次OD600 nm值,每种分离菌株重复测定3次,收集并统计所有测量数据,绘制分离菌株的生长曲线。

1.2.2.4 溶血活性测定

取分离菌株菌悬液100 μL,用灭菌生理盐水稀释后均匀涂布于绵羊血平板上,于37 ℃恒温培养箱培养24 h,观察菌落周围有无溶血环。

1.2.2.5 药物敏感性试验

参照Hou等[17]的方法,选取头孢噻呋、青霉素G、庆大霉素、卡那霉素等13种常用抗生素对分离菌株进行药物敏感性试验。先用无菌棉签将稀释至0.5号麦氏标准管浊度的分离菌株菌悬液(1×108 CFU/mL)涂布于LB固体培养基上,然后将药敏试纸片放置在培养基上,37 ℃培养24 h,测量抑菌圈直径。结果判定参照美国临床和实验室标准化协会(CLSI)标准。

1.2.2.6 抑菌活性测定

参照王琦等[18]的方法,用无菌棉签分别蘸取浓度为1×108 CFU/mL的金黄色葡萄球菌、牛源大肠杆菌K99、鼠伤寒沙门氏菌菌悬液,均匀涂布于LB固体培养基表面,将无菌的牛津杯放置在培养基上,向牛津杯中加入200 μL浓度为1.1×108 CFU/mL的新鲜分离菌株菌悬液,37 ℃培养24 h。每种分离菌进行3次重复试验。测量抑菌圈直径,根据抑菌圈直径判定菌株的抑菌活性。

1.2.3 分离菌株体内安全性评价

参照农业农村部制定的《直接饲喂微生物和发酵制品生产菌株鉴定及其安全性评价指南》和巨敏莹等[19]的报道,对分离菌株进行安全性评估。

1.2.3.1 急性毒性试验

28只昆明小鼠适应环境7 d后随机分为2组,每组14只,公母各占1/2。小鼠自由采食和饮水。在第1天,对照组小鼠腹腔注射生理盐水(0.2 mL/只),试验组小鼠腹腔注射浓度为1.1×1010 CFU/mL的枯草芽孢杆菌F31菌悬液(0.2 mL/只),持续观察小鼠的体重变化、毛色、整体精神状态及分泌物、排泄物状况,7 d后安乐处死所有小鼠,解剖观察各组织器官是否存在病理变化。

1.2.3.2 亚急性毒性试验

56只昆明小鼠适应环境7 d后随机分为4组,每组14只,公母各占1/2。对照组灌胃生理盐水(0.2 mL/只),其他3组分别灌胃浓度为1.1×108(低剂量组)、1.1×109(中剂量组)、1.1×1010 CFU/mL(高剂量组)的枯草芽孢杆菌F31菌悬液(0.2 mL/只),持续灌胃28 d。每日灌胃前记录小鼠体重,观察小鼠精神状态、饮食饮水及排泄物情况。灌胃结束后,采集小鼠血液,进行血细胞指标分析;安乐处死所有小鼠,取心脏、肝脏、脾脏、肺脏、肾脏和胸腺,无尘纸擦干表面水分后称重,计算脏器指数[脏器指数(%)=100×脏器重(g)/体重(g)];采集部分肝脏、脾脏组织,将其置于4%多聚甲醛中固定,制作石蜡切片,然后进行苏木精-伊红(HE)染色,在显微镜下观察组织病理变化情况。

1.3 数据统计与分析

试验数据采用SPSS 17.0软件进行统计分析,对小鼠急性毒性试验结果进行t检验,亚急性毒性试验结果进行单因素方差分析(one-way ANOVA),P<0.05表示差异显著。结果以平均值±标准差表示。使用Graphpad Prism 8.0软件绘图。

2 结果与分析

2.1 芽孢杆菌的初筛结果

2.1.1 芽孢杆菌的培养及形态学观察

从LB琼脂培养基上分离得到3株菌,分别命名为F21、F31、F54。F21、F31菌株的菌落特征均为表面干燥,边缘粗糙,不透明,呈乳白色,有隆起;F54菌株的菌落特征为表面略湿润,边缘粗糙,呈乳白色,有隆起(图1-A)。革兰氏染色后,在油镜下观察到这3株菌均为蓝紫色的革兰氏阳性杆菌(图1-B)。

2.1.2 生化鉴定

分离菌株的生化鉴定结果见表1,参照《伯杰细菌鉴定手册》,初步判定分离菌株F21、F31、F54均符合枯草芽孢杆菌的生化特性。

2.1.3 分子生物学鉴定

对分离菌株的16S rRNA测序结果进行BLAST比对,结果显示,F21、F31和F54菌株与枯草芽孢杆菌的同源性均在99%以上。用MEGA 7.0.14软件将这3株菌与BLAST比对结果中同源性最高的菌株进行核苷酸序列比对,绘制系统发育进化树(图2)。通过对菌落形态特征、革兰氏染色结果、生化鉴定结果以及分子生物学鉴定结果进行综合分析,判定这3株分离菌均为枯草芽孢杆菌。

2.2 分离菌株的体外益生性能筛选结果

2.2.1 耐酸能力

由表2可知,3株分离菌经pH为2.0的PBS处理后存活率都有所降低,但均在60%以上。其中,F21菌株对酸的耐受能力最强,在pH为2.0条件下,其存活率仍高达80.97%。

2.2.2 耐胆盐能力

由表3可知,3株分离菌经胆盐浓度为0.3%的PBS处理后,存活率均在70%以上。其中,F54菌株的耐胆盐能力最强,在胆盐浓度为0.3%条件下存活率仍可达84.81%。

2.2.3 生长曲线

3种分离菌的生长曲线如图3所示,F21菌株在2~12 h为对数生长期,12 h后进入稳定期;F31菌株在2~18 h为对数生长期,16 h后进入稳定期;F54菌株在4~12 h为对数生长期,12 h后进入稳定期。

2.2.4 溶血活性

如图4所示,F21、F31、F54菌株在绵羊血平板上培养后,在菌落周围均未观察到溶血环。

2.2.5 药物敏感性

由表4可知,F21菌株对青霉素G等11种药物敏感,对链霉素和多黏菌素耐受;F31菌株对青霉素G等12种药物敏感,对万古霉素耐受;F54对链霉素、庆大霉素、四环素及卡那霉素敏感,对其他药物均耐受。选择药物敏感性相对更好的F21、F31菌株进行抑菌活性测定。

2.2.6 抑菌活性测定

由表5可知,F21、F31菌株对金黄色葡萄球菌均有抑菌活性,抑菌圈直径分别是14.13和11.69 mm。F21菌株对大肠杆菌无明显抑制效果,抑菌圈直径为8.23 mm;F31对大肠杆菌抑制效果明显,抑菌圈直径为14.41 mm。F21、F31菌株对沙门氏菌均有抑菌活性,抑菌圈直径分别为12.44和17.14 mm,可见F31菌株对沙门氏菌的抑菌活性最佳。

2.3 枯草芽孢杆菌F31的安全性评价结果

通过体外益生特性试验结果筛选出F31菌株的耐酸耐胆盐能力好、不具有溶血活性、药物敏感性强、抑菌效果明显,对F31菌株进行小鼠急性和亚急性毒性试验,探究其剂量安全性。

2.3.1 小鼠急性毒性试验

2.3.1.1 小鼠剖检观察

小鼠腹腔注射枯草芽孢杆菌F31 7 d后,对照组与试验组小鼠均未见异常的身体活动变化、精神状态和毒性的临床体征,2组小鼠剖检腹腔内脏器无明显眼观病理差异。

2.3.1.2 生长性能

由表6可知,试验组小鼠的终末体重、平均日增重、体重增长率与对照组均无显著差异(P>0.05)。

2.3.2 小鼠亚急性毒性试验

2.3.2.1 小鼠剖检观察

灌胃枯草芽孢杆菌F31过程中,4组小鼠无中毒及死亡现象且毛色、精神状态、食欲、排便情况均正常,剖检时小鼠口、鼻、耳及肛门处无异常分泌物,各脏器眼观无明显异常。

2.3.2.2 体重

由图5可知,低剂量组、高剂量组小鼠28日龄体重与对照组均无显著差异(P>0.05),中剂量组小鼠28日龄体重显著高于对照组(P<0.05)。

2.3.2.3 脏器系数

由表7可知,与对照组相比,低剂量组小鼠的肝脏指数显著降低(P<0.05),高剂量组小鼠的脾脏指数显著升高(P<0.05)。灌胃不同剂量灌胃枯草芽孢杆菌F31对小鼠的心脏指数、肺脏指数、肾脏指数和胸腺指数均无显著影响(P>0.05)。

2.3.2.4 血细胞指标

由表8可知,低剂量组小鼠血液中白细胞数、淋巴细胞数、单核细胞数及中性粒细胞数与对照组相比均显著降低(P<0.05)。中剂量组、高剂量组小鼠各血细胞指标与对照组相比均无显著差异(P>0.05)。

2.3.2.5 组织病理学观察

通过观察4组小鼠肝脏、脾脏组织切片(图6),未见小鼠肝脏和脾脏出现病理变化。

3 讨论

3.1 基于体外益生性能筛选分离菌株

益生菌想要通过胃在肠道中定植并发挥益生作用,需要具备对低pH环境和胆盐环境的良好耐受能力。健康动物胃液的pH一般为2.0~3.0,肠道中的胆盐浓度一般为0.1%~0.3%。有研究发现,从滇南小耳猪粪便中获得的1株枯草芽孢杆菌在pH为3.0及胆盐浓度为0.3%条件下的存活率分别为50.9%和48.1%[20]。本试验中,从白羽王鸽新鲜粪便和肠道内容物中分离出的枯草芽孢杆菌F21、F31、F54在pH为2.0条件下的存活率均在60%以上,在胆盐浓度为0.3%条件下的存活率均在70%以上,这表明3株枯草芽孢杆菌均有良好的耐酸、耐胆盐能力,可以抵抗胃肠道的恶劣环境,满足开发为微生态制剂的潜在条件。此外,益生菌在肠道内增殖到一定数量时,才能发挥调节肠道菌群平衡、促进健康的作用,所以用于微生态制剂的菌株应具备良好的生长性能[21]。生长曲线能够反映细菌的生长情况,细菌在生长对数期时繁殖速度最快,进入稳定期后生长趋于平衡[22]。本试验中,枯草芽孢杆菌F21、F31和F54均可快速进入生长对数期,并较快到达稳定期,展现出较好的生长性能。
细菌性溶血素是一种能够溶解红细胞的毒素,可使细菌在血平板上呈现溶血现象。由于产生溶血素的细菌可引起肝炎、溶血、肾病综合征及菌血症等多种疾病,因此溶血性常被用作评估菌株体外安全性的指标[23]。唐萍等[24]在对1株贝莱斯芽孢杆菌进行生物安全评价时未发现其有溶血活性。本试验分离的3株枯草芽孢杆菌在血平板培养过程中均未出现溶血现象,表明其不会产生溶血素,具有一定的安全性。
益生菌制剂的候选菌株必须满足以下条件:能够在胃肠道环境中生存;能够产生抗菌物质,对病原菌有抑制作用;不具有耐药性[25]。由于细菌的耐药性可以发生转移,因此菌株是否具有耐药性是评价其能否作为益生菌制剂的条件之一[26]。在本试验中,枯草芽孢杆菌F21、F31对大多数常见抗菌药物均表现出敏感性,而枯草芽孢杆菌F54对大多数抗菌药物不敏感,可能具有耐药性,所以枯草芽孢杆菌F54不符合作为益生菌制剂的条件。这一结果与樊丹等[27]分离到的鳟鱼源枯草芽孢杆菌及杜汉宇等[28]分离到的猪源枯草芽孢杆菌的耐药性研究结果不同,这可能是由菌株本身的差异性导致的。枯草芽孢杆菌F54具有耐药性可能是由于携带了耐药基因[29],其是否携带耐药基因可在后续进一步研究。
益生菌可以通过竞争排斥作用和产生抗菌物质等方式抑制病原菌生长繁殖,这是评价菌株益生潜能的标准之一[30]。大肠杆菌、金黄色葡萄球菌、沙门氏菌是常见的肠道病原菌[31]。已有研究表明,枯草芽孢杆菌对大肠杆菌、金黄色葡萄球菌、沙门氏菌这类常见肠道病原菌具有抑制作用[32-34]。本研究结果同样表明,枯草芽孢杆菌F31对金黄色葡萄球菌、大肠杆菌、沙门氏菌均有较强的体外抑菌活性,其中对沙门氏菌的抑制效果最佳。鸽沙门氏菌病是常发于鸽群中的细菌性传染病之一,可通过垂直传播在鸽群中持续引发感染,给鸽养殖产业造成巨大风险[35-36]。因此,后续可将对沙门氏菌抑制效果最佳的枯草芽孢杆菌F31作为防治鸽沙门氏菌病的候选益生菌菌株。

3.2 枯草芽孢杆菌F31的安全性

在益生菌菌株进入临床试验之前,应先进行动物毒性试验,以确定其是否具有潜在毒性,并探究其安全剂量[37]。研究表明,动物毒性试验是判断益生菌菌株能否用于后续临床试验的重要标准[38-39]。本试验通过7 d的小鼠急性毒性试验发现,腹腔注射枯草芽孢杆菌F31后小鼠未出现异常精神状态或死亡等临床体征。在28 d的亚急性毒性试验过程中,灌胃枯草芽孢杆菌F31的小鼠均保持健康且无死亡状况,体重增长正常。然而,低剂量组小鼠的肝脏指数显著低于对照组,高剂量组小鼠的脾脏指数显著高于对照组。组织病理学检查结果显示肝脏、脾脏未出现明显损伤。由于脾脏与机体免疫功能密切相关,脾脏指数的升高可能与免疫力增强有关,但并无病理学意义[40]。此外,血细胞指标检测结果显示,灌胃枯草芽孢杆菌F31后低剂量组小鼠白细胞数、淋巴细胞数、单核细胞数及中性粒细胞数与对照组存在显著差异,但均处在小鼠血细胞检测值的正常范围之内,与巨大芽孢杆菌BM24具有同样的安全性[41],这表明灌胃枯草芽孢杆菌F31对小鼠无有害影响。

4 结论

本试验从白羽王鸽新鲜粪便和肠道内容物中分离得到的枯草芽孢杆菌F31有较好的耐酸和耐胆盐能力,对大部分抗生素敏感,不具有溶血活性,对3种常见致病菌均有抑制作用,具有良好的体外益生潜能。此外,小鼠体内安全性试验结果表明枯草芽孢杆菌F31的安全性良好,可作为鸽源益生菌制剂候选菌株用于后续的开发与应用。
