Progress of Algal Protein Replacement of Fishmeal in Aquaculture Feed Application

Abstract

Algae, with their high nutritional value and balanced amino acid profile, are increasingly emerging as a potential key protein source to replace fishmeal in the aquaculture feed industry. Previously, fishmeal was considered the go-to high-quality protein source for aquaculture, but supply shortages and rising prices have limited its use, driving the development of algae protein as a new solution. Research indicates that appropriately replacing fishmeal with algae protein (less than 80%) can benefit fish growth, enhance intestinal digestion and absorption, improve immunity, and enhance muscle quality; however, excessive replacement (80% or more) may inhibit physiological metabolism and damage tissue structure. Currently, the application of algae protein still faces challenges such as high production costs at scale, low digestibility, and anti-nutritional factors in algae sources. This paper comprehensively analyses the characteristics of several common algal proteins, their current application status, and technical challenges and summarizes relevant solution strategies, aiming to provide theoretical support and practical guidance for the research and application of fishmeal alternatives in aquaculture feed.

蛋白质是鱼类有机体结构和功能不可或缺的营养物质之一,在生命过程中发挥着关键作用[1]。鱼粉作为水产养殖长期依赖的优质蛋白质,具有丰富的蛋白质含量、良好的适口性和较低的抗营养因子(ANFs)。然而,随着全球渔业资源枯竭,鱼粉有限的供给和不断攀升的价格致使自身在水产养殖中的使用严重受限。因此,开发新型可持续蛋白质源减少鱼粉使用成为水产行业迫切需求。一些常见的动植物蛋白质源曾广泛应用于水产饲料中,但随着研究的深入发现,动物性蛋白质源会表现出氨基酸失衡、脂肪酸含量过多等问题;植物性蛋白质源普遍存在适口性差、纤维素和ANFs含量高等缺陷,长期过量使用会降低水产动物对营养物质的吸收利用率,进一步影响水产品的风味[2]。而藻类蛋白质不仅蛋白质含量丰富、氨基酸组分均衡,还富含多种微量元素、不饱和脂肪酸和糖类等,其营养价值可与鱼粉等优质蛋白质媲美[3]。此外,藻类培养所需空间小且适应能力强的特征减少了其生产过程对环境的压力,上述优点为藻类蛋白质替代鱼粉成为新型蛋白质源提供了可能[4]。本文主要就藻类蛋白质种类及特点、应用现状及其在替代鱼粉作为饲料蛋白质源的过程中面临的问题及解决措施等方面进行了总结,旨在为水产饲料中鱼粉替代的研发和应用提供依据。

1 藻类蛋白质种类及特点

藻类形态多种多样,可根据色素沉着划分为3个高级类群:红色(红藻门,约6 131种)、棕色(褐藻门,约1 792种)和绿色(绿藻门,约4 548种)[5];藻类的营养价值正随着科研人员深入的研究而逐渐被人们所重视。已知水产养殖中使用的藻类超过40种,应用较多的藻类主要包括小球藻(Chlorella vulgaris)、螺旋藻(Spirulina)、拟微绿球藻(Nannochloropsis gaditana)和栅藻(Scenedesmus)等。藻类被认为是比大多数动植物性蛋白质更安全、更环保、更经济的蛋白质源[6]。与大多数植物蛋白醇溶性不同,藻类蛋白质主要呈现水溶特性(如螺旋藻蛋白质全部为水溶性),这一特征与水生动物消化系统的渗透压环境高度适配,可显著提升蛋白质的消化吸收率;大量不饱和脂肪酸成为了鱼类营养不可缺少的一部分;各种天然色素的存在能有效改善鱼肉品质。目前,藻类作为蛋白质来源已在大量研究中进行了评估,发现适量比例藻类添加能显著提高鱼类生产性能和饲料利用率;但藻生物质规模化生产成本对于饲养需求而言仍偏高,且藻类过量替代对鱼类生长发育存在一定有害影响。因此,除了成本及鱼类生长和营养利用的问题外,仍需针对长期使用藻类蛋白质源饲料喂养对鱼类生理代谢和组织形态等存在的潜在不良影响进一步探究[7]

1.1 小球藻

小球藻是单细胞淡水绿藻的一种,也是我国最早实现人工培养的微藻。小球藻中的各项营养成分含量较其他常见动植物蛋白质源很占优势,其中蛋白质含量为63.36%~63.98%(干物质基础),与鱼粉蛋白质水平接近;氨基酸组分均衡且包含鱼类生长所需多种必需氨基酸,其中谷氨酸、天门冬氨酸和亮氨酸含量较高,而蛋氨酸+胱氨酸与苏氨酸含量相对较低,分别作为第一、二限制性氨基酸出现[8]。因此,在使用小球藻蛋白质替代鱼粉时还应注意适量补充蛋氨酸、胱氨酸和苏氨酸,以满足水生生物生长需求。藻类蛋白质的营养价值还受到其脂质种类和含量的影响,小球藻属的脂质比例在14%~22%[9]。不同种类的小球藻脂质含量各不相同,且处于特定生长环境中的小球藻油脂含量偏高,并富含多种对水产动物生长至关重要的多不饱和脂肪酸(PUFA)。水产饲料中适量添加小球藻,有助于补充n-3系列PUFA,显著减少养殖鱼类应激反应和慢性疾病的发生风险;藻多糖和多种天然色素(如虾青素)存在能够提高鱼体生长性能并有效清除自由基,增强抗氧化及免疫能力[10]

1.2 螺旋藻

螺旋藻是“节螺旋藻”的通用名称,是一类由丝状多细胞组成,能进行光合自养的低级原核蓝细菌,自然生活在高pH和高碱度的热带和亚热带水体中[11]。螺旋藻作为一种古老的藻水生藻类植物,在营养价值、消化吸收率、适口性和安全性方面均优于其他类别,是当前常用藻类蛋白质中最受关注的品种之一。极大螺旋藻和钝顶螺旋藻是水产养殖中研究最多的2种替代鱼粉的螺旋藻[12],其蛋白质含量丰富且均衡,通常可达60%~75%;与小球藻属相比,螺旋藻不具有纤维素性细胞壁,且其蛋白质均为水溶性,这使得螺旋藻细胞成分能够被水生动物高度消化吸收[13]。螺旋藻中不仅富含水生动物机体无法自主合成的8种必需氨基酸和多种矿物质,还含有丰富的酚类物质、藻蓝蛋白和藻多糖,这些成分具有抗氧化、抗炎以及免疫调节的功能[14]。螺旋藻可用作多种动物饲料鱼粉蛋白质源的替代品,能提升幼鱼存活率、促进其生长发育、增强免疫力并改善肌肉色泽品质。

1.3 拟微绿球藻

拟微绿球藻是一种归属于褐藻门(Ochrophyta)的海洋单细胞藻类。拟微绿球藻适温范围广泛,易于培养且繁殖迅速,能够积累高浓度的天然色素增强水生动物机体免疫屏障和抗氧化能力[15]。与许多其他藻类相比,拟微球藻含有更高浓度的类胡萝卜素,这些类胡萝卜素化合物很容易被鱼类吸收并转化为可用的色素,能够有效帮助肌肉增色,提高鱼肉品质。拟微绿球藻中的多种多不饱和脂肪酸,特别是二十碳五烯酸(EPA),是鱼类神经发育和抗炎的关键功能脂质来源,能有效提高鱼类苗种的发育和存活率[16]。拟微绿球藻表现出较高的生物量积累率和丰富的脂质(16.6%)、蛋白质(43%),被认为是最有前途的鱼类微藻原料[17]

1.4 栅藻

1976年Trainor等[18]提出,广义的栅藻包含了2个属(或亚属),以有无芒刺为区别性特征分为链带藻属(Desmodesmus)和栅藻属(Scenedesmus)。其中,前者包含有细胞具刺的种类,而后者均不含芒刺等附属结构。目前,狭义的栅藻是一种常见的淡水绿藻,一般由4~8个(或16~32个)细胞组成定性群体。栅藻细胞通常为椭圆形或纺锤形,呈线性或交替排列形成扁平状的共生体。栅藻对恶劣环境条件的适应能力很强,营养要求简单,生长速度快,是一种适合在实验室培养的藻类。栅列藻属蛋白质含量十分丰富,几乎包含生物体生长所需各种必需氨基酸,其氨基酸结构模式可与鸡蛋和大豆等优质动植物蛋白质相媲美[19]。此外,栅藻中还含有大量亚麻酸脂类、维生素和类胡萝卜素等营养物质。其中,维生素不仅有助于增强生物体免疫屏障,还能促进肠道对铁的吸收转运;类胡萝卜素中的虾青素能保护细胞、脂质及膜脂蛋白免受氧化应激损害;β-胡萝卜素和叶黄素作为天然着色剂能显著改善动物体肌肉色泽[7,20-21]。而部分研究表明,藻类中碳水化合物常以多种形式存在(如纤维素、淀粉和多糖),而纤维素与多糖正是细胞壁的主要成分,这可能致使栅藻蛋白质消化率降低[22]。与螺旋藻和小球藻等种类相比,栅藻蛋白质虽含有较高的营养价值和生物活性,但目前用于研究栅藻属作为饲料鱼粉替代物的水产动物仍然较少,具体效果仍需进一步探究。

2 藻类替代鱼粉在水产养殖中的应用

藻类是一类多样的放氧光合生物,具有分布范围广、生长繁殖速度快及对极端环境耐受性强等特点。高蛋白质含量和一些共同组分(表1)是部分微藻能够替代鱼粉作为水产饲料蛋白质源之一的主要原因[23]。藻类通常以干粉形式添加并与其他原料混合搭配制成鱼类饲料,不仅能够为鱼类提供生长所需各种营养要素,还能帮助提高机体组织、细胞抗氧化能力,增加采食量的同时降低投喂过程中饲料损耗,从而提高水产养殖经济效益。研究表明,藻类部分替代鱼粉时,可通过显著增加饲料中生物活性化合物含量,包括功能性蛋白质和抗氧化剂等具有抗病毒和抗菌特性的物质,对鱼类生长性能、肠道健康、免疫状态及肌肉品质等方面产生不同程度的影响[24]

2.1 藻类替代鱼粉对水产动物生长性能的影响

藻类富含多种营养成分,添加藻类蛋白质有利于促进养殖鱼类的生长和饲料利用率的提高。适量小球藻替代斑马鱼(Danio rerio)饲料中的鱼粉,幼鱼体重显著增加,生长性能明显优于对照组[28];饲喂大口黑鲈(Micropterus salmoides)15.03%~15.43%的藻粉替代鱼粉制成的饲料,饲料利用率呈线性增长[29];15%的螺旋藻替代鱼粉制成饲料投喂鲶鱼(Ompok pabda),鱼体增重幅度和特定生长率均显著高于对照组[30]。此外,鱼类生长性能的提高与摄食量的增加有一定关联性。虹鳟(Oncorhynchus mykiss)饲料中添加5%的小球藻,其摄食率、特定生长率及增重率得到显著提升[31];25%~75%的螺旋藻替代饲料鱼粉,可显著增加罗氏沼虾(Macrobrachium rosenbergii)的摄食率和增长率,提高幼虾成活率[32]。藻类蛋白质部分替代鱼粉后水生生物摄食率的增加,可能与藻类中富含的多种矿物质、微量元素和不饱和脂肪酸改善饲料的适口性有关。有研究表明,大多数水生植物(包括藻类在内)中含有40%及以上结构性碳水化合物,其结构复杂且稳定,一定程度上限制水产动物对营养物质的消化吸收[33]。黄颡鱼(Pelteobagrus fulvidraco)饲料中螺旋藻替代鱼粉比例超过80%时,显著降低了鱼体终末体重、摄食率及特定生长率[34]。47.45%的小球藻粕完全替代饲料鱼粉显著降低大口黑鲈饲料利用率,导致生长发育迟缓,肝脏遭受损伤[29]。虽然藻类中丰富的营养和生物活性成分有助于改善水产饲料应用现状,但应注意饲料中藻类蛋白质添加量与鱼类生长效果并不成正比,只有适当比例藻类替代鱼粉才能对鱼体生长性能及健康产生积极作用。

2.2 藻类替代鱼粉对水产动物肠道消化能力的影响

藻类中丰富的藻多糖能为肠道有益菌提供反应所需底物,在一定程度上能够起到调节水生动物肠道微生态平衡的作用。小球藻和螺旋藻中的纤维消化活性酶及β-葡聚糖能够促进肠道有益菌群如乳酸菌(Lactobacillus)和双歧杆菌(Bifidobacterium)的增殖,显著改善肠道微环境[35]。其中,β-葡聚糖能被肠道微生物降解生成短链脂肪酸(SCFAs),这些SCFAs不仅为肠道表层细胞提供能量,还可调控蛋白质合成相关基因,增强肠道保护功能。此外,SCFAs能激活G蛋白偶联受体41/43(GPR41/43),促进抗炎信号通路,减少肠道炎症、保障肠道健康[36-37]。藻类中富含的大量游离氨基酸和矿物元素,能刺激肠道并改善微生物组成,促使体内生成更多的内源性消化酶,进而对水产动物肠道消化能力产生影响。窄爪小龙虾(Pontastacus leptodactylus)饲料中部分鱼粉被小球藻替代后,肠道乳酸菌数量显著增加同时大肠杆菌数量减少,有助于维持胃肠道良性微生态平衡,增强肠道消化吸收功能[38];花鲈(Lateolabrax japonicus)饲料添加4%~5%螺旋藻,其生长性能及饲料利用率显著提升,肠道蛋白酶活性增加,促进营养成分消化吸收[39]。然而,当饲料中过量藻类蛋白质替代鱼粉时,由于藻类刚性细胞壁的存在,可能会限制消化酶对机体细胞内成分的获取与利用,阻碍细胞膜内营养分解吸收[40]。因此,在藻类作为蛋白质源替代饲料鱼粉的过程中,应注意对藻类原料进行前处理,通过破壁粉碎工艺增加藻类蛋白质的吸收利用率。

2.3 藻类替代鱼粉对水产动物免疫能力的影响

水产动物机体免疫是其抵抗病原体侵袭的重要防线,当藻类蛋白质替代鱼粉时,其丰富的多酚化合物、β-胡萝卜素和藻类多糖含量对鱼类免疫屏障起到积极作用[41]。其中,藻多糖被认为是免疫调节剂,能够通过激活先天免疫受体(TLR)信号通路,提高巨噬细胞和淋巴细胞活性的同时增强抗炎因子的表达,从而强化机体免疫屏障[42];栅藻及其他藻类中富含的β-胡萝卜素和多酚化合物能够激活机体核因子-E2相关因子2(Nrf2)通路,诱导下游抗氧化酶活性增加,并清除自由基和氧化应激产物,保护细胞组织免受损伤[43]。饲料中藻类的添加使尼罗罗非鱼肝脾脏肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)等免疫反应基因的表达量显著增加[44];而对于褐牙鲆(Paralichthys olivaceus),饲料中小球藻添加比例达10%~15%时,其血浆过氧化氢酶活性显著升高[45]。研究证明,上述免疫和抗氧化信号的激活显著上调了闭合蛋白1(Claudin1),封闭蛋白(Occludin)和紧密连接蛋白-1(ZO-1)等基因的表达,使组织上皮细胞紧密连接功能加强,有效防止有害物质和病原菌的穿透[46]。藻类中的活性成分能够直接或间接激活水产动物免疫系统,因此,饲料中适量藻类蛋白质替代鱼粉能使水产动物获得较强的抗氧化能力,同时维持机体免疫稳态提高抗病性。

2.4 藻类替代鱼粉对水产动物肌肉品质的影响

藻类中富含鱼体自身无法合成的类胡萝卜素,是促进色素沉积的天然补充剂,能帮助改善肌肉色泽同时提高肉质[47]。起初,Liu等[34]发现,黄颡鱼腹部和背部皮肤中黄度(b*)值与叶黄素含量呈正相关,并认为叶黄素可能是判断黄颡鱼色素沉着的关键指标。Xi等[29]证实,适量藻类蛋白质作为饲料添加剂,鱼体背肌中的叶绿素含量成正比增长,显著增加了背体、腹体和背肌的b*值;Yu等[48]认为,饲料中藻类蛋白质的添加使大口黑鲈的皮肤亮度(L*)值和肌肉品质得到显著提升,且不影响肌肉纤维的发育。此外,肌肉脂肪和蛋白质含量也是评估肉质的2个重要参数。研究表明,添加适量藻类的饲料,不仅显著降低鱼体肌肉脂质过氧化程度[49],还能有效增加基质部胶原蛋白含量,刺激肌肉蛋白质合成改善肉质[50]。由于部分藻类中富含的EPA可诱导过氧化物酶体增殖物激活受体(PPAR)家族中受体α(PPARα)和受体γ(PPARγ)基因的表达,促进肌肉细胞脂肪酸β-氧化,抑制过量沉积并调节脂肪酸合成酶(FAS)和脂肪酸结合蛋白(FABP)的表达[51]。上述相关基因的表达有助于优化脂肪酸组分,提高肌肉鲜味和营养价值。因此,藻类蛋白质部分替代鱼粉,能够通过改善肉色、降低肌肉脂质过氧化速度和加快蛋白质合成的方式,达到提高肌肉品质的目的。

3 藻类替代鱼粉的应用前景

藻类作为水产饲料领域中的新型蛋白质源,展现出不同维度的应用价值。其不仅可有效缓解鱼粉资源短缺危机,还具备独特的环境调控功能。研究表明,藻类细胞壁多糖与重金属离子具有高亲和力,能在较短的时间内快速建立吸附平衡,显著降低养殖水体中重金属离子含量[52]。藻类蛋白质中含多种活性物质,杀灭病原菌抑制细菌繁殖同时,增强鱼类疾病防控。这一特性使得藻类蛋白质应用打破了传统饲料替代范畴,为水产健康养殖开辟方向。但值得关注的是,大规模将藻生物质应用到水产饲料中的先决条件仍是廉价的生产成本和高效的蛋白质利用率等问题,目前藻类想要替代鱼粉作为饲料蛋白质源还面临着诸多不确定因素。

3.1 藻类替代鱼粉过程中面临的问题

3.1.1 供需失衡及成本制约

据报道,目前全球自养藻生物质年产量约为2万t(干物质),远低于水产养殖业所需的鱼粉缺额[53]。此外,藻类规模化培养面临经济与技术双重难题:一方面,光合自养模式下藻类生长依赖外源二氧化碳(CO2)作为碳源,但工业级CO2(纯度≥99%)供应途径较少且成本高昂;另一方面,藻类生产过程中脱水和干燥环节耗能较高,约占总生产能耗的40%。目前大多数技术仍处于试验开发阶段,尚未形成产业化发展规模。以上原因均使藻类生产成本大幅增加[10]。研究表明,小球藻和螺旋藻等藻类生产成本在10~30美元/kg,而历年来鱼粉最高价格仅为1.74美元/kg[54]。因此,巨大的供求差及工业化生产工艺不成熟造成的替代成本过高等问题,使得大规模利用藻类替代鱼粉无法实现。

3.1.2 消化率低

相关研究证实,过量的藻类蛋白质替代鱼粉,会抑制鱼类生长,降低饲料利用率。当螺旋藻替代40%的鱼粉时引发尖吻鲈(Lates calcarifer)肠道微绒毛萎缩及肝脏损伤,阻碍肠道消化吸收,减缓鱼体生长发育[55]。20%的栅藻替代饲料鱼粉,显著降低了大西洋鲑(Salmo salar)的消化率和饲料转化率[19]。这可能与藻类细胞壁过厚及碳水化合物含量高有关[56],刚性细胞壁阻碍蛋白质释放,降低鱼类消化率[57];碳水化合物摄入过量,引发鱼类糖代谢紊乱[58]。高水平拟微绿球藻替代鱼粉,较高的纤维素(3.7%)和半纤维素水平(43.3 μg/mg)显著减缓尼罗罗非鱼的生长速度[59]。饲料藻类蛋白质高水平替代使碳水化合物含量过高(24%~36%),导致大口黑鲈葡萄糖耐受不良、代谢异常进一步致使鱼体生长迟缓[29]。水产动物肠道缺乏非淀粉多糖降解酶系,加剧了藻类营养利用障碍[60]

3.1.3 ANFs

ANFs是一种存在于食物和饲料中的内源性物质,当被人和动物过量摄入时,会对健康产生不利影响。藻类含单宁、凝集素等ANFs,高替代比例可能抑制摄食与生长[61-62]。尽管藻类ANFs普遍低于动植物蛋白质,但其与水产动物生长抑制的剂量效应关系尚未明确,需进一步探究作用机制及阈值。

3.2 解决措施

为了使藻类蛋白质更适合水产饲料,需尽可能降低藻类生产成本、分解其细胞壁以释放细胞内营养成份并清除有毒化学物质。
藻类替代鱼粉的供求缺口问题,降低藻类生产成本的同时提高产量成为当务之急,而异养培养可能成为解决这些问题的有效方法。与传统的藻类自养模式相比,异养培养是在没有光照的情况下,利用有机碳源进行微藻生长的封闭发酵过程。异养培养成本低且细胞生长密度更高;设施建设要求简单,更易于大规模维护[63]。此外,改进藻类培养设施及选育抗逆新品种也能够在一定程度上降低微藻粉的生产成本[64]
藻类消化率低的问题可采取多种方法,通过物理珠磨和冷冻或化学酶解等方法破坏细胞壁结构并释放胞内有机化合物,同时利用双水相系统(ATPS)将微藻中的淀粉与其他细胞组分分离,以此减少因碳水化合物和非淀粉多糖造成的低消化率等不利影响[65-66]
部分藻类蛋白质源中存在的ANFs,可采用物理(制粉、浸泡和高压蒸煮)、化学(碱)和生物(微生物发酵)等方式处理,也可选育一些ANFs含量较低的藻类作为蛋白质源候选,以缓解单宁、生物碱、凝集素和蛋白酶抑制剂等有害物质的作用[67]。但应注意在处理藻类的过程中需更谨慎,尽量减少藻类营养物质的损失。

4 小结

为了实现水产养殖可持续发展,减少鱼粉依赖并探索开发适宜的替代蛋白质源成为关键,藻类作为潜在新型蛋白质源已受到广泛关注。研究表明,饲料中以一定比例的藻类蛋白质替代鱼粉,能有效提高鱼类生产性能及饲料利用率,增强鱼体抗氧化能力及免疫屏障功能,并改善肌肉品质。然而,藻类蛋白质源的大规模开发和利用仍面临诸多挑战,如产量、成本、消化率及抗营养因子等问题。为了充分挖掘藻类蛋白质的潜力并提升其在水产养殖中的应用价值,未来研究需聚焦于几个关键方向:开发高蛋白质含量、生长快速、抗逆性强的新型藻类品种;优化藻类培养工艺以提高产量并降低成本;深入研究藻类蛋白质在水产动物体内消化吸收机制,并通过加工处理等手段提高其利用率、降低抗营养因子的影响等。这些深层探索将进一步推动藻类蛋白质替代鱼粉走向产业化之路。
