Evaluation of Standardized Ileal Digestibility of Amino Acids in Rapeseed Meal from Different Sources for Medium-Growing Yellow-Feathered Broilers and Establishment of Their Predicting Equations

Abstract

This study was conducted to analyze the chemical component contents and amino acid composition in rapeseed meal from different sources, and evaluate the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids in rapeseed meal for medium-growing yellow-feathered broilers, to establish the predicting equations based on the chemical component contents. The chemical component contents and amino acid composition in rapeseed meal from different sources were analyzed. A total of 276 sixty-three-day-old medium-growing yellow-feathered broilers were used in a one-way completely randomized design, including 1 nitrogen-free diet group and 10 rapeseed meal diet groups, with six replicates per group. The nitrogen-free diet group and rapeseed meal diet groups contained 6 and 4 broilers per replicate, respectively. The experimental period was 5 days. At the end of the experiment, the ileal digesta were collected, the titanium dioxide (TiO2) was used as an indicator, the amino acid contents in raw materials, diets and digesta were determined, and the AID and SID of amino acids were calculated. The results showed as follows: 1) the mean contents of gross energy (GE) and dry matter (DM), crude protein (CP), ether extract (EE), crude ash (Ash), nitrogen-free extract (NFE) were 17.91 MJ/kg and 10.19%, 39.28%, 2.94%, 6.57%, 28.39%, respectively, the mean contents of crude fiber (CF), neutral detergent fiber (NDF) and acid detergent fiber (ADF) were 12.62%, 29.51% and 21.59%, respectively, the mean contents of phytic acid (PA), glucosinolates (GSL) and erucic acid (EA) were 13.86 g/kg, 9.37 mmol/kg and 5.43%, respectively; the mean contents of 18 amino acids in the rapeseed meal ranged from 0.47% to 7.39%. 2) The means of AID and SID of 18 amino acids of rapeseed meal from different sources for medium-growing yellow-feathered broilers ranged from 52.5% to 75.8% and 60.9% to 79.9%, respectively, and the arginine showed the highest AID and SID. The rapeseed meal from different sources significantly affected the AID and SID of 18 amino acids (P<0.05). 3) In this study, 15 predicting equations of amino acids SID of rapeseed meal for medium-growing yellow-feathered broilers were established, the predicted values of SID of 13 amino acids were accorded the actual measurement range (the measured value of predicted value was in mean±standard deviation), and most equations had coefficients of determination (R2)>0.9, indicated their good predicting responses. In conclusion, the contents of chemical components and AID and SID of amino acids of rapeseed meat from different sources exist difference, and the established predicting equations provide a valuable reference for rapidly predicting the SID values of most amino acids in rapeseed meal for medium-growing yellow-feathered broilers.

豆粕为优质的植物性蛋白质饲料原料。随着畜牧业的快速发展,我国豆粕产量难以满足需求,需大量依赖进口。近年来,受多种因素影响,豆粕价格居高不下[1-3],非常规饲料资源的开发及应用可大幅缓解豆粕的需求压力。菜籽粕作为油菜籽加工的副产物,替代豆粕具有独特的价格优势,其氨基酸种类丰富且含量平衡[4]。但菜籽粕因产地、品种及加工工艺的差异,导致其氨基酸利用率存在差异。因此,精准评价中速型黄羽肉仔鸡对不同来源菜籽粕的氨基酸标准回肠消化率(standardized ileal digestibility,SID),并根据其化学成分含量建立预测方程,以精准配制黄羽肉仔鸡饲粮,满足其特定氨基酸需求,对于促进饲料养分的高效转化、减少浪费、实现生产性能提升与氮减排的双重目标以及节约饲料资源具有积极意义[5-6]。
目前,针对肉仔鸡常用饲料原料有效养分的系统性认知与数据库构建仍显薄弱,加之有效的预测方程匮乏,这构成了制约肉仔鸡精准营养配方技术发展的主要瓶颈[7]。目前,国内外研究测定肉仔鸡饲料原料的氨基酸SID主要通过无氮饲粮法和指示剂法,已成熟应用于黄羽肉仔鸡常用饲料原料的氨基酸SID研究中[8-9]。刘小杰等[10]基于不同来源小麦的化学成分含量,成功构建了生长猪小麦氨基酸SID的预测方程。Yuan等[11]通过分析不同来源豆粕的化学成分含量及氨基酸组成,以及精准评定黄羽肉仔鸡对不同来源豆粕的氨基酸SID,成功构建了基于化学成分含量及氨基酸组成的黄羽肉仔鸡对豆粕的氨基酸SID的预测方程。然而,关于黄羽肉仔鸡对不同来源菜籽粕的氨基酸SID评价及其预测方程构建的研究报道十分有限。此外,尽管国内外学者已围绕饲料原料的氨基酸SID预测方程开展了一定研究,但是部分研究报道的氨基酸SID预测方程的决定系数(R2)准确度不高。潘泳霖等[12]测定育成期蛋鸡对10种不同来源玉米的氨基酸SID预测方程的拟合度进行比较,发现苯丙氨酸最高(R2=0.975),色氨酸最低(R2=0.449);而Hang等[13]研究发现,13日龄肉鸡玉米的氨基酸SID逐步回归方程的R2在0.403(组氨酸)~0.989(天冬氨酸),跨度逾1倍,表明同种饲料原料不同氨基酸的SID预测精度也存在差距。因此,本研究提出如下科学假说,即应用不同来源菜籽粕的化学成分含量及氨基酸组成可构建具有更高预测精度和稳定性的预测方程,从而实现对不同来源菜籽粕氨基酸SID的准确评估与动态预测。为了验证以上科学假说,本研究旨在通过分析不同来源菜籽粕的化学成分含量及氨基酸组成,并精准评定其对黄羽肉仔鸡的氨基酸SID,建立基于其化学成分含量及氨基酸组成的氨基酸SID预测方程,同时验证预测方程的准确性,为黄羽肉仔鸡生产中快速预测其对菜籽粕的氨基酸SID提供重要参考,并对于高效利用菜籽粕饲料资源、缓解我国蛋白质饲料资源短缺的压力以及降低对进口蛋白质饲料原料的依赖具有重要意义。

1 材料与方法

1.1 伦理声明

动物试验伦理申请已获得扬州大学动物科学与技术学院伦理委员会批准,许可编号:SYXK(苏)2021-0027。

1.2 试验材料和试验设计

本研究所用10个菜籽粕样品由本实验室前期采集的15个不同来源样品经系统分析后筛选确定,菜籽粕样品信息见表1。

本研究以63日龄中速型黄羽肉仔鸡公鸡为研究对象,采用单因素完全随机设计,共设11个组,包括1个无氮饲粮组和由不同来源菜籽粕配制的10个菜籽粕饲粮组。试验期5 d(63~67日龄)。
选用530只55日龄的健康黄羽肉仔公鸡,预饲喂至63日龄后空腹称重。筛选出276只体重相近的试验鸡,随机分为11个组,每组6个重复;其中,菜籽粕饲粮组每个重复4只鸡,无氮饲粮组每个重复6只鸡。因无氮饲粮组试验鸡采食量通常较低,为保证无氮饲粮组有足够的回肠食糜用于后续分析,特将每个重复试验鸡数量从4只增加至6只。试验鸡采用笼养(90 cm×70 cm×45 cm),整个试验期的环境控制均参照黄羽肉仔鸡的饲养管理要求设定。
55~62日龄为适应期,试验鸡自由采食相同的商品饲粮及自由饮用自来水。63~67日龄为试验期,试验鸡自由采食各组试验饲粮及自由饮用自来水。无氮饲粮以玉米淀粉、葡萄糖等原料配制而成。菜籽粕饲粮以待测菜籽粕为唯一蛋白质和氨基酸来源,并添加玉米淀粉、葡萄糖作为碳水化合物补充。无氮饲粮和菜籽粕饲粮组成及营养水平见表2。以二氧化钛(TiO2)为外源指示剂。

1)预混料为每千克饲粮提供 The premix provided the following per kg of diets:VA 10 000 IU,VD3 3 750 IU,VE 27.5 IU,VK 2.5 mg,VB1 2.5 mg,VB2 8.0 mg,VB6 3.75 mg,VB12 0.025 mg,泛酸钙 calcium pantothenate 12.5 mg,烟酸 niacin 45 mg,叶酸 folic acid 125 mg,生物素 biotin 0.125 mg,沸石粉 zeolite powder 1 347.8 mg,Cu (CuSO4·5H2O) 7 mg,Fe (FeSO4·H2O) 80 mg,Zn (ZnSO4·H2O) 55 mg,Mn (MnSO4·H2O) 60 mg,Se (Na2SeO3) 0.15 mg,I [Ca(IO3)2·H2O] 0.5 mg。

2)粗蛋白质和氨基酸为实测值,其他为计算值,参照NY/T 3645—2020。CP and amino acids were measured values, while the others were calculated values, according to NY/T 3645—2020.

1.3 样品采集与制备

68日龄时,试验鸡自由采食后实施二氧化碳窒息屠宰。无氮饲粮组和菜籽粕饲粮组每个重复分别解剖6和4只试验鸡。采用回肠末端食糜法,试验鸡屠宰后分离回肠,对折后截取后1/2(弃去最后2 cm),经蒸馏水冲洗2次获取食糜,注意避免挤压肠黏膜。收集后的食糜立即放置于-20 ℃冷冻保存,完全冻结后的食糜放入冷冻干燥机中冻干,置室温下回潮24 h。各重复内所有试验鸡的冻干食糜经充分混合后粉碎,待测。

1.4 指标测定

饲粮中粗蛋白质(CP)含量采用GB/T 6432—2018的方法进行测定。饲粮和食糜中15种氨基酸(中性氨基酸:甘氨酸、丙氨酸、缬氨酸、亮氨酸、异亮氨酸、脯氨酸、丝氨酸、苏氨酸、酪氨酸、苯丙氨酸;酸性氨基酸:天冬氨酸、谷氨酸;碱性氨基酸:赖氨酸、精氨酸、组氨酸)含量采用GB/T 18246—2019的方法进行测定,含硫氨基酸(蛋氨酸和胱氨酸)含量采用GB/T 15399—2018的方法进行测定,色氨酸含量采用GB/T 15400—2018的方法进行测定,TiO2含量采用GB 5009.246—2016的方法进行测定。10种菜籽粕中水分、CP、粗脂肪(EE)、粗灰分(Ash)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)和粗纤维(CF)含量分别采用GB/T 6435—2014、GB/T 6432—2018、GB/T 6433—2006、GB/T 6438—2007、GB/T 20806—2022、NY/T 1459—2022和GB/T 6434—2022的方法进行测定,并计算无氮浸出物(NFE)含量;硫代葡萄糖苷(GSL)和芥酸(EA)含量分别采用NY/T 1582—2007和GB/T 23890—2009的方法进行测定;植酸(PA)含量采用试剂盒(苏州科铭生物技术有限公司)进行测定;总能(GE)采用全自动量热仪测定。
黄羽肉仔鸡对菜籽粕中氨基酸表观回肠消化率(apparent ileal digestibility,AID)和SID的计算公式如下:
氨基酸AID(%)=100-[(A1×B1)/(C1×D1)×100];
E1(mg/kg)=(A1×B1)/D1;
氨基酸SID(%)=氨基酸AID+(E1/C1)×100;
式中:A1为食糜中氨基酸含量(mg/kg);B1为饲粮中TiO2含量(mg/kg);C1为饲粮中氨基酸含量(mg/kg);D1为食糜中TiO2含量(mg/kg);E1为无氮饲粮中内源性氨基酸含量(mg/kg)。

1.5 数据统计分析

通过SAS 9.4软件中的一般线性模型(GLM)程序对10种菜籽粕的氨基酸AID和SID数据进行单因素方差分析,对于方差分析显著的指标,进一步采用最小显著性差异(LSD)法进行多重比较。通过RANDEN子程序随机抽取1个样品作为外部验证样本(样品9),剩余9个样品先采用皮尔逊相关性分析评估原料化学成分含量与氨基酸SID的关联强度,随后采用逐步回归分析,构建相应氨基酸的最佳预测方程。以随机抽取的样品9为验证材料,对所得预测方程的准确性进行检验。所有统计分析均以重复为基本单元,以P<0.05作为差异显著性检验水平。

2 结果与分析

2.1 不同来源菜籽粕的化学成分含量及氨基酸组成

由表3可知,10个不同来源菜籽粕的GE及水分、CP、EE、CF、NDF、ADF、Ash、NFE、PA、GSL和EA含量分别在17.24~19.37 MJ/kg、8.03%~12.70%、37.75%~41.73%、1.23%~6.99%、8.87%~16.53%、23.23%~45.63%、16.33%~38.15%、5.42%~7.86%、24.31%~32.90%、10.77~15.91 g/kg、0.96~54.68 mmol/kg和0~27.70%;其中,GE(4.39%)和CP(4.21%)的变异系数较小,而GSL(175.39%)和EA(181.38%)的变异系数较大。氨基酸含量平均值在0.47%~7.39%;其中,必需氨基酸的变异系数在2.57%~20.35%,以蛋氨酸最大,组氨酸最小;非必需氨基酸的变异系数在3.38%~22.89%,以脯氨酸最小,半胱氨酸最大。

2.2 不同来源菜籽粕对中速型黄羽肉仔鸡回肠氨基酸消化率的影响

由表4可知,不同来源菜籽粕对肉仔鸡18种氨基酸AID均有显著影响(P<0.05)。

18种氨基酸AID的变异系数在3.1%~13.7%,以半胱氨酸最小,赖氨酸最大。18种氨基酸AID的平均值在52.5%~75.8%,其中必需氨基酸中以精氨酸最高(75.8%),苏氨酸最低(52.5%);非必需氨基酸中以谷氨酸最高(74.5%),丝氨酸最低(56.4%)。
由表5可知,不同来源菜籽粕对肉仔鸡18种氨基酸SID均有显著影响(P<0.05)。18种氨基酸SID的变异系数在2.4%~11.4%,相较于AID,其变异程度更低,说明采用SID来评估氨基酸消化率具有更好的稳定性;其中,以半胱氨酸最小,赖氨酸最大。18种氨基酸SID的平均值在60.9%~79.9%,普遍高于AID,其中必需氨基酸中以精氨酸最高(79.9%),苏氨酸最低(60.9%);非必需氨基酸中以半胱氨酸最高(77.8%),甘氨酸最低(64.8%)。

2.3 中速型黄羽肉仔鸡对菜籽粕的氨基酸SID与菜籽粕化学成分含量的相关性分析

由表6可知,蛋氨酸、苯丙氨酸、组氨酸、精氨酸、天冬氨酸、谷氨酸、半胱氨酸SID与GSL含量呈显著负相关(P<0.05),组氨酸和谷氨酸SID与GE呈显著负相关(P<0.05),苯丙氨酸和组氨酸SID与EE含量呈显著负相关(P<0.05),丙氨酸SID与CF含量呈显著负相关(P<0.05),赖氨酸SID与NDF和EA含量呈显著负相关(P<0.05)。

2.6 中速型黄羽肉仔鸡对菜籽粕的氨基酸SID预测方程构建

由表9可知,以菜籽粕的化学成分和氨基酸含量为自变量,进行了逐步回归分析,构建的中速型黄羽肉仔鸡对菜籽粕的氨基酸SID预测方程均有效(P<0.05),且多数预测方程的R2>0.9,预测效果良好。

由表10可知,利用菜籽粕样品9对黄羽肉仔鸡菜籽粕氨基酸SID预测方程的准确性进行验证,所建方程能准确预测除甘氨酸和组氨酸外的其余13种氨基酸SID(预测值在实测值的平均值±标准差内),表现出良好的适用性。

3 讨论

本试验结果显示,不同来源菜籽粕的化学成分含量平均值分别为:水分10.19%、GE 17.91 MJ/kg、CP 39.28%、EE 2.94%、Ash 6.57%、CF 12.62%、NDF 29.51%、ADF 21.59%、NFE 28.39%、PA 13.86%、GSL 9.37%、EA 5.43%。不同来源菜籽粕的18种氨基酸含量存在差异,平均值在0.47%~2.90%。氨基酸消化率测定结果显示,SID平均值(60.9%~79.9%)高于AID平均值(52.5%~75.8%)。基于菜籽粕化学成分构建的15个预测方程中,多数预测方程的R2>0.9,预测能力良好,尤以蛋氨酸方程的拟合度最优。本研究提出的假说得以验证,并为快速预测中速型黄羽肉仔鸡菜籽粕氨基酸SID提供了新的思路和依据。
本研究分析了10个不同来源菜籽粕的化学成分含量,结果显示,所选10个菜籽粕样品的CP含量平均值为39.28%,且除GE及CP、NFE和PA含量外,其余指标变异系数均大于10%,表明样品覆盖面广、具有代表性。这与杨新拓等[14]研究中所选用的10种不同来源菜籽粕样品CP含量平均值(43.77%)相差不大。本研究中菜籽粕GE平均值为17.91 MJ/kg,与郑仕文[15]测定的菜籽粕GE(18.78 MJ/kg)相似。菜籽粕的NDF和ADF含量平均值分别为29.51%和21.59%,而李双江等[16]在四川地区测定的菜籽粕的NDF和ADF含量平均值分别为52.65%和35.12%,与本试验所测结果有一定的差异,可能与菜籽粕的产地和加工工艺等有关。本研究中,不同来源菜籽粕的EE含量平均值为2.94%,与Wu等[17]报道的菜籽粕的EE含量平均值(4.31%)差别较大;不同来源菜籽粕的水分含量平均值为10.19%,与韩冉[18]报道的水分含量平均值(10.58%)相近;不同来源菜籽粕的Ash含量平均值为6.57%,与《中国饲料与营养价值表(2024年第35版)》[19]中报道的Ash含量(6.0%)相近;不同来源菜籽粕的CF含量平均值为12.62%,低于Ye等[20]测得的16.95%。出现上述差异的原因可能与油菜的品种、产地环境、加工工艺(压榨或浸出方式、脱毒处理)以及储存条件(温湿度控制)等因素有关[21]。本研究测定的18种氨基酸变异系数普遍较大,与已有文献相符;部分氨基酸测定值的差异可能受油菜籽生长季节、品种及加工工艺等因素影响[22-23]。
本研究中,中速型黄羽肉仔鸡对不同产地菜籽粕的氨基酸AID平均值介于苏氨酸的52.5%至精氨酸的75.8%之间。邓雪娟[24]在36日龄白羽肉仔鸡上的研究表明,菜籽粕氨基酸AID中精氨酸最高,苏氨酸最低,这与本试验结果一致。类似地,张露等[25]在10种不同来源小麦次粉中及李双江[26]在四川北部地区菜籽粕对肉兔的氨基酸AID的研究中分别测得苏氨酸的AID最低,精氨酸的AID最高。本试验测得黄羽肉仔鸡对菜籽粕丝氨酸的AID为56.4%,且不同产地菜籽粕对18种氨基酸的AID均有显著影响,这一变化规律与刘丹丹等[27]在科宝肉仔鸡上的研究结果相似;其中,苯丙氨酸、组氨酸等氨基酸的AID与李波[28]在生长猪上菜籽粕氨基酸AID的研究结果高度吻合。但部分氨基酸AID略有出入,推测可能与动物品种、日龄、测定方法等因素有关[29]。
本研究中,中速型黄羽肉仔鸡对菜籽粕的氨基酸SID的变异系数显示,赖氨酸最大,与张开心[30]在不同来源菜籽粕对肉鸭氨基酸SID的研究中赖氨酸变异系数最大的结果一致;但半胱氨酸SID的变异系数最小,与黄连宿[31]在不同来源菜籽粕对空怀母猪氨基酸SID的研究中半胱氨酸SID变异系数最小的结果一致;不同来源菜籽粕对黄羽肉仔鸡18种氨基酸SID均有显著影响;其中,色氨酸、甘氨酸和丙氨酸的SID与余晨昊[32]在母猪妊娠后期上测得的菜籽粕氨基酸SID结果接近,赖氨酸和组氨酸的SID与郑萍等[33]对生长猪菜籽粕氨基酸SID的报道值接近。本研究中,氨基酸SID平均值在60.9%~79.9%(苏氨酸最低,精氨酸最高),而杭振宇等[34]报道的白羽肉鸡玉米氨基酸SID在68.14%~89.56%(苏氨酸最低,亮氨酸最高)。这种差异可能与品种、日龄、环境及加工方式有关[35-36]。
当前,饲料原料回归预测方程的研究多见于代谢能,而对氨基酸利用率,特别是家禽菜籽粕SID的研究尚显不足[37]。本试验中,菜籽粕丙氨酸SID与CF含量呈负相关,与孙征[38]在13日龄雄性爱拔益加肉仔鸡上菜籽粕氨基酸SID的研究结果相符。此外,黄祥祥[39]在28日龄白羽肉仔鸡上菜籽粕中赖氨酸SID与NDF含量之间呈负相关的结论,也佐证了本文的发现。这些研究结果表明,饲粮中的纤维成分会促进肠道黏液分泌,从而加剧内源性氨基酸的流失,进而导致菜籽粕氨基酸SID降低[40-41]。本研究中,菜籽粕组氨酸SID与GE之间呈显著负相关关系,与孙久鹏[42]在13日龄白羽肉仔鸡上报道的菜籽粕组氨酸SID与GE呈显著负相关关系的结果一致。但是,菜籽粕甘氨酸SID与GE之间没有显著相关性。孙久鹏等[43]在13和28日龄白羽肉仔鸡菜籽粕氨基酸SID与GSL含量之间没有发现显著相关性,而本研究中菜籽粕蛋氨酸、苯丙氨酸、组氨酸、精氨酸、天冬氨酸、谷氨酸和半胱氨酸SID与GSL含量之间存在显著负相关,此与先前研究结果[44-46]一致,提示GSL可能干扰动物的消化、吸收和代谢过程,间接导致氨基酸的生物学效价降低。上述不同研究中出现的差异可能与肉仔鸡品种、日龄及菜籽粕加工方式等因素有关。
本研究还发现,中速型黄羽肉仔鸡对菜籽粕的氨基酸SID与其必需氨基酸及非必需氨基酸含量之间具有显著正相关关系。Wang等[47]在非妊娠母猪上也报道了相似的研究结果。这些结果表明,菜籽粕中的氨基酸有利于提高中速型黄羽肉仔鸡对菜籽粕的氨基酸SID。
本试验中,亮氨酸和甘氨酸SID的预测方程中预测因子的数量均为1个,其中亮氨酸的R2在0.9以上,而甘氨酸的R2在0.7以下,其余方程均含2个及2个以上预测因子,且R2>0.8(多数>0.9),证实了预测因子数量与预测效果之间的正相关关系[48-49]。最终,本研究成功构建了15个具有显著差异的氨基酸SID预测方程。经验证,13个方程的预测值处于实测样品氨基酸SID平均值±标准差的范围内,符合预期,仅甘氨酸和组氨酸的预测方程存在一定偏差,且多数方程R2>0.9,预测效果良好,可用于快速评定中速型黄羽肉仔鸡菜籽粕的氨基酸SID。但是,应该指出的是,本研究用于检验氨基酸SID预测方程的预测准确性的菜籽粕样品太少(仅1个),今后应用更多的菜籽粕样品检验上述氨基酸SID预测方程的预测准确性,以便上述氨基酸SID预测方程能在中速型黄羽肉仔鸡生产中更快推广应用。

4 结论

① 不同来源菜籽粕的化学成分含量变异程度不同。GE、CP、NFE、PA的变异系数较小(<10%),而水分、EE、CF、NDF、ADF、Ash、GSL、EA的变异系数较大(>10%)。18种氨基酸的含量平均值在0.47%~7.39%,变异系数在2.57%~22.89%,以半胱氨酸的变异系数最大,组氨酸的变异系数最小。
② 中速型黄羽肉仔鸡对不同菜籽粕的氨基酸SID平均值存在差异,在64.3%~79.9%;其中,精氨酸的SID平均值最高,而缬氨酸的SID平均值最低。
③ 采用逐步回归法构建了中速型黄羽肉仔鸡菜籽粕15种氨基酸SID的预测方程,除甘氨酸和组氨酸外,其余13种氨基酸SID的预测方程均表现出良好的预测准确性,且其中大部分方程的R2>0.9,表明预测方程具有较好的预测效果。
