Effects of Dietary Trans-Anethole on Antioxidant Capacity of Broilers in Vivo and in Vitro

Abstract

This study was conducted to explore the antioxidant capacity of trans-anethole (AN) of AA broilers in vivo and in vitro. In vitro test, the strong antioxidant butylated hydroxytoluene (BHT) was used as a positive control, and the antioxidant activity of AN was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH), diammonium 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonate) (ABTS), hydroxyl radical scavenging methods and ferric ion reducing antioxidant power (FRAP) method, the antioxidant capacity of AN was expressed as half maximal inhibitory concentration (IC50). In vivo test, a total of 256 healthy 1-day-old broilers were randomly divided into four groups, with eight replicates in each group and eight chickens in each replicate. The addition levels of AN in each group were 0, 200, 400 and 600 mg/kg (AN 0, AN 200, AN 400 and AN 600 groups). The experimental period was 42 days, which was divided into two stages:the early stage (1 to 21 days of age) and the late stage (22 to 42 days of age). The results showed as follows:1) the IC50 values of AN for DPPH, ABTS and hydroxyl radicals were 16.26, 6.98 and 7.82 mg/mL. The reduction ability for Fe3+ was weak. 2) Compared with AN 0 group, the average daily feed intake (ADFI) of AA broilers aged 42 days in AN 400 group significantly increased (P<0.05), and the ADFI of AA broilers in the whole growth period of AA broilers in AN 200 group significantly increased (P<0.05). AN had no significant effect on the average daily gain (ADG) and feed to gain ratio (F/G) of AA broilers in early and late stage (P>0.05). Effects of AN on the antioxidant capacity of AA broilers aged 21 days:compared with AN 0 group, the serum total superoxide dismutase (T-SOD) activity in AN 200, AN 400 and AN 600 groups significantly increased (P<0.05), and the serum malondialdehyde (MDA) content in AN 400 and AN 600 groups significantly decreased (P<0.05), the liver catalase (CAT) activity in AN 600 group significantly increased (P<0.05), and the T-SOD activity in AN 200 and AN 400 groups significantly increased (P<0.05), the relative expression level of catalase (CAT) gene in liver in AN 200, AN 400 and AN 600 groups significantly increased (P<0.05), and the relative expression level of superoxide dismutase 1 (SOD1) gene in AN 400 and AN 600 groups significantly increased (P<0.05), the relative expression level of heme oxygenase-1 (HO-1) gene in AN 600 group also significantly increased (P<0.05). Effects of AN on antioxidant capacity of AA broilers aged 42 days:compared with AN 0 group, serum total antioxidant capacity (T-AOC) in AN 400 and AN 600 groups significantly increased (P<0.05), the liver CAT activity in AN 200 and AN 400 groups significantly increased (P<0.05), the activity of glutathione peroxidase (GSH-Px) and T-AOC in breast muscle in AN 200 group significantly increased (P<0.05), the relative expression level of glutathione peroxidase 1 (GPx1) gene in breast muscle in AN 200 group significantly increased (P<0.05). In summary, AN can scavenge DPPH, ABTS and hydroxyl radicals to a certain extent, but the overall antioxidant effect is weaker than BHT. AN can improve the antioxidant capacity of serum, liver and breast muscle of AA broiler and reduce the production of MDA in serum and liver. Under the experimental conditions, the optimal dietary AN supplemental level is 400 mg/kg.
