Effects of Lactic Acid Bacteria Additives and Harvest Periods on Quality of Natural Forage Silage Based on Principal Component Analysis and Membership Function Analysis

Abstract

This experiment aimed at exploring the effects of three kinds of lactic acid bacteria (LAB) additives and three harvest periods on quality of natural forage silage. Three kinds of LAB addictive, Lactobacillus plantarum (LP) and L. buchneri (LB) cultivated by forage processing team of China Agricultural University and commercial LAB additive (SY), was inoculated to the natural forage silage, respectively, setting untreated as a control (CK). The natural forage silage was prepared on 12 July (early period), 22 July (middle period) and 1 August (late period), 2021. Thus there were 12 groups totally and each with 3 replicates. After 75 days of fermentation, fermentation quality and nutritional quality were tested. The results showed that the LAB additive and harvest period had significant interaction on butyric acid content (P<0.01) and ammonia nitrogen/total nitrogen (P<0.01), the contents of dry matter and ether extract (P<0.05). Compared with CK group, pH, contents of butyric acid, neutral detergent fiber and acid detergent fiber as well as ammonia nitrogen/total nitrogen in LAB groups were significantly reduced (P<0.05), while a significant increase of the contents of lactic acid, acetic acid and dry matter can be seen at the same time (P<0.05). As the harvest period passed, the contents of crude protein and hemi-cellulose in natural forage silage decreased evidently (P<0.05), while contents of acid detergent fiber and acid detergent lignin increased obviously (P<0.05). The pH, butyric acid content, ammonia nitrogen/total nitrogen and neutral detergent fiber content of silage in middle period were remarkably higher than those in early and late periods (P<0.05). According to principal component analysis and membership function analysis, the ranking of silage from early period was LP group=SY group>LB group>CK group, from middle period was LP group=LB group>SY group>CK group, and from late period was LB group>LP group=SY group>CK group. In conclusion, the harvest period can affect the improvement effects of LAB additives on the quality of natural forage silage. The addition effect of LP is better than other additives both in the early and middle periods, otherwise the addition effect of LB is best in the late period.

天然牧草广泛分布于中国的北部和西部,是中国畜牧业生产的重要饲草资源^[1]。将天然牧草调制成干草是一种传统的保存方法。但是天然牧草的收获和晾晒常在雨季进行,这造成干草的干物质(dry matter, DM)和粗蛋白质(crude protein,CP)损失显著增加^[2]。青贮可有效保藏牧草的营养物质,具有受天气影响小、提高牧草适口性和家畜的消化率等优点^[3],可有效解决冬季家畜饲料短缺的问题。然而,天然牧草含水量低、可溶性碳水化合物(water soluble carbohydrates,WSC)含量低、缓冲能值(buffer capacity,BC)较高、附着乳酸菌(lactic acid bacteria,LAB)数量较少,直接青贮很难成功。LAB是青贮饲料调制中常用的一种添加剂,可促进青贮原料充分发酵。Cai等^[4]评估了添加剂对牧草青贮发酵的影响,发现使用LAB可以提高牧草青贮的发酵品质。有报道称,在天然牧草青贮饲料中添加LAB可以改善其品质^[5]。同时,收获时期会影响青贮饲料的发酵品质和营养品质^[6-7]。Long等^[8]研究发现,收获期对王草的青贮品质和微生物群落有显著影响,于生长期110 d收获的王草的青贮品质最好,具有更高的LAB相对丰度。但是目前关于LAB和收获时期及二者的交互作用对天然牧草青贮品质影响的相关研究还鲜见报道。因此,本试验对不同收获时期和添加不同LAB处理的天然牧草青贮发酵品质和营养品质进行测定分析,采用主成分分析和隶属函数法评定LAB添加剂在不同时期收获的天然牧草青贮中的应用效果,以期为天然牧草青贮技术的应用和评价提供参考依据。

1 材料与方法

1.1 试验设计

研究区域位于内蒙古呼伦贝尔市陈巴尔虎旗的草地农业生态系统试验站(N49°20'~49°26'、E119°55'~120°9',海拔628~649 m),分别于2021年7月12日(早期)、7月22日(中期)和8月1日(晚期),在草甸草原样地中采用五点取样法收获天然牧草,牧草主要组成为羊草(Leymus chinensis)、无芒雀麦(Bromus inermis)、二裂委陵菜(Potentilla bifurca)、斜茎黄芪(Astragalus laxmannii)、艾蒿(Artemisia argyi)。将样品切短至2~5 cm,混匀后对天然牧草的混合样设置3种LAB添加剂的处理,分别是中国农业大学饲草加工团队筛选出的植物乳杆菌(Lactobacillus plantarum,LP)和布氏乳杆菌(Lactobacillus brucei,LB)以及商业使用的LAB添加剂(SY,活性菌为植物乳杆菌),以未添加LAB为对照(CK),接种量为≥1×10⁶ CFU/g。经充分混匀,取300 g天然牧草的混合样装入真空袋内,密封。共计12个组,每组重复3次,发酵75 d,打开后测定天然牧草青贮的发酵品质和营养品质。

1.2 指标测定

1.2.1 发酵品质测定

青贮开启后混匀,随机取20 g鲜样,加入180 mL蒸馏水,榨汁1 min后用4层纱布过滤,获取青贮饲料浸提液,用pH仪测定浸提液pH,使用岛津高效液相色谱仪测定青贮饲料的乳酸(lactic acid,LA)、乙酸(acetic acid,AA)、丙酸(propionic acid,PA)和丁酸(butyric acid,BA)含量,色谱柱为Shodex Rspak KC-811S-DVB gel Column 30 mm×8 mm,柱温50 ℃,检测器为SPD-M10AVP,流动相为3 mmol/L高氯酸,检测波长为210 nm,进样量为5 μL。用苯酚-次氯酸钠比色法测定氨态氮(ammonia-nitrogen,NH₃-N)含量^[9],并计算氨态氮占总氮(total nitrogen,TN)的比例(氨态氮/总氮)。

1.2.2 营养品质测定

取100 g的青贮料,65 ℃烘干48 h,用以测定DM含量。将烘干后的样品粉碎过40目筛,用范式洗涤纤维法测定中性洗涤纤维(neutral detergent fiber,NDF)、酸性洗涤纤维(acid detergent fiber,ADF)和酸性洗涤木质素(acid detergent lignin,ADL)含量,用凯氏定氮法测定CP含量,用蒽酮-硫酸比色法测定WSC含量,用AOAC(2000)^[10]中方法测定粗脂肪(ether extract,EE)含量^[10]。原料各指标的测定参照上述方法,用AOAC(1975)^[10]中方法测定原料粗灰分(Ash)含量,盐酸-氢氧化钠滴定法测定原料BC^[11],纤维素(neutral detergent fiber,CL)、半纤维素(hemi-cellulose,HC)含量通过以下公式计算:

CL=ADF-ADL;

HC=NDF-ADF。

1.3 综合评价

利用主成分分析将12个组的14个指标降维,简化评价指标数量,利用模糊数学隶属函数法计算各组选择指标的隶属函数值,并将所选指标隶属函数值求和,之后取平均值,以平均值大小进行综合排名,平均值越大,排名越靠前。隶属函数计算公式^[12]如下:

1)R(X_i)=(X_i-X_min)/(X_max-X_min);

2)R(X_i)=1-(X_i-X_min)/(X_max-X_min)。

式中:R(X_i)表示某指标的隶属函数值;X_i表示该指标的测定值;X_max表示该指标的最大值;X_min表示该指标的最小值。若测定指标与饲料品质呈正相关关系,则选择公式1)计算;若呈负相关关系,则选择公式2)计算。

1.4 统计分析

采用Excel 2010初步处理数据,采用SPSS 17.0软件进行多变量方差分析,并用Duncan氏法对各测定数据进行多重比较,测定结果用平均值±均值标准误表示。

2 结果与分析

2.1 天然牧草营养成分

各收获时期天然牧草营养成分如表1所示。早期DM、NDF和ADF含量和BC显著高于中期和晚期 (P<0.05),中期WSC含量显著低于早期和晚期 (P<0.05)。随着收获时期的延后,CP含量变化不显著(P>0.05)。

2.2 LAB添加剂及收获时期对天然牧草青贮饲料发酵品质的影响

由表2可知,LAB添加剂和收获时期对BA含量和NH₃-N/TN具有显著的交互作用(P<0.01)。与CK组相比,在中期收获时添加LAB可以显著降低BA含量(P<0.05)。在早期收获时,LP组和SY组的NH₃-N/TN显著低于CK组和LB组(P<0.05),LB组显著低于CK组(P<0.05)。LAB添加剂显著降低天然牧草青贮饲料的pH(P<0.001)、NH₃-N/TN(P<0.001)和BA含量(P<0.01),显著增加了LA和AA含量(P<0.01),对PA含量没有显著影响(P>0.05)。随着天然牧草收获时期的延后,天然牧草青贮饲料的pH、NH₃-N/TN和BA含量均表现为先升高后降低。收获时期对LA、AA和PA含量没有显著影响(P>0.05)。

2.3 LAB添加剂及收获时期对天然牧草青贮饲料营养品质的影响

由表3可知,LAB添加剂和收获时期对DM和EE含量存在显著的交互作用(P<0.05)。早期收获时,LP组的DM含量显著高于CK组和SY组(P<0.05),LB组显著高于CK组(P<0.05);LP组的EE含量显著低于SY组(P<0.05)。中期收获时,LAB组的DM含量显著高于CK组(P<0.05),3种LAB添加剂之间没有显著差异(P>0.05)。晚期收获时,LB组和SY组的DM含量显著高于CK组(P<0.05),LP组与CK组之间没有显著差异(P>0.05)。与CK组相比,LAB显著提高了天然牧草青贮饲料的DM含量(P<0.05),其中LP组和LB组显著高于SY组(P<0.05)。LAB对CP和EE含量没有显著影响(P>0.05)。随收获时期的延后,天然牧草青贮饲料的DM和CP含量逐渐降低,EE含量先升高后降低。

LAB添加剂和收获时期对天然牧草青贮饲料营养品质中的木质纤维组分不具有显著交互效应(P>0.05)。LAB显著降低了NDF(P<0.01)和ADF含量(P<0.001),对ADL、CL和HC含量没有显著影响(P>0.05)。随收获时期的延后,天然牧草青贮饲料的NDF含量先升高后降低,ADF和ADL含量先升高后不变,HC含量显著降低(P<0.05),CL含量没有显著变化(P>0.05)。

2.4 不同处理下的天然牧草青贮饲料的主成分分析

对12个组的天然牧草青贮饲料主成分分析结果见表4。基于特征值>1标准挑选出5个主成分。其中,第1主成分的特征值为5.338,贡献率为38.129%,其对应特征向量中绝对值较大的指标为DM、pH、NH₃-N/TN、ADF和CP含量;第2主成分的特征值为2.308,贡献率为16.488%,其对应特征向量中绝对值较大的指标为HC含量;第3主成分的特征值为1.672,贡献率为11.944,其对应特征向量中绝对值较大的指标为AA、ADL和CL含量;第4主成分的特征值为1.490,贡献率为10.640%,其对应特征向量中绝对值较大的指标为LA和AA含量;第5主成分的特征值为1.202,贡献率为8.588%,其对应特征向量中绝对值较大的指标为PA含量。这5个主成分的累积贡献率达到了85.788%,即保留了原有指标85.788%的信息。

2.5 各指标综合得分及其排名

根据主成分分析结果,在挑选出的5个主成分中分别选择pH、NH₃-N/TN及DM、LA、PA、ADF、CL、HC、CP含量这9个指标来替代其余指标进行模糊隶属函数分析并排名。结果如表5所示,各添加剂组的平均隶属函数值均大于CK组。3个收获时期中,中期收获的平均隶属函数值最低。早期收获时,LP组和SY组平均隶属函数值最高,均为0.69。中期收获时,LP组和LB组平均隶属函数值最高,均为0.50。晚期收获时,LB组的平均隶属函数值最高,为0.54。

3 讨论

3.1 LAB添加剂及收获期对天然牧草青贮饲料发酵品质的影响

pH是衡量青贮饲料发酵品质的重要指标。LAB利用碳水化合物产生LA是青贮过程中pH下降的主要原因。一般认为pH在4.2以下为发酵优良^[13]。本试验中,LAB组的pH均在4.2以下,发酵品质优良,说明3种LAB均可以改善天然牧草青贮发酵品质。青贮过程中,在植物蛋白酶和微生物酶的共同作用下蛋白质发生水解产生氨类物质^[14-15],青贮过程中蛋白质的水解程度由NH₃-N/TN反映^[16],通常良好发酵的青贮饲料NH₃-N/TN不超过10%。本试验中,LP组的NH₃-N/TN最低,说明LP有效抑制了蛋白质的水解,这导致LP组在LA较少的同时具有更低的pH。BA主要由梭状芽孢杆菌这一不良微生物代谢产生。添加LAB后,BA含量显著降低,这与Zhao等^[17]在苜蓿上取得的结果一致。这是因为LAB提升了天然牧草青贮过程中的LAB数量,在发酵前期大量产生LA,使pH快速降低,从而抑制梭状芽孢杆菌等不良微生物的繁殖。

青贮原料的化学组分和附着的微生物会直接影响青贮发酵品质^[18]。其中,WSC含量和附生LAB数量影响青贮早期LA的增加速度和pH的下降速度,是青贮成功的关键^[19]。青贮原料中的WSC含量在中期最低,导致青贮发酵过程中LAB不能快速产生大量LA,以抑制不良微生物的繁殖,使得该时期调制的天然牧草青贮饲料具有更高的BA含量和NH₃-N/TN。同时,不同生长阶段的植物表面附着的微生物会存在显著差异^[20],这也可能成为不同收获时期调制的天然牧草青贮发酵品质不同的原因。

3.2 LAB添加剂及收获期对天然牧草青贮饲料营养品质的影响

DM是评定青贮饲料营养品质的重要指标之一。研究表明,LAB可以有效降低DM损失,提高DM含量^[21],这与本试验结果一致。主要原因是添加LAB增加了LAB菌群数量,促进了发酵进程,从而降低DM损失^[22]。NDF和ADF是影响家畜采食和消化的重要因素。NDF含量越高,家畜的干物质采食量(DMI)越低;ADF含量越高,家畜的DM消化率越低。Li等^[5]和Xia等^[23]分别研究了添加LAB对天然牧草青贮和甜高粱和田菁混合青贮发酵品质和营养价值的影响,均发现使用LAB添加剂可以显著降低NDF和ADF含量,与本试验结果一致。这可能是因为LAB的添加增加了LAB的数量,促进了青贮发酵,增加了LA的积累量,使NDF和ADF等结构性碳水化合物发生分解^[24]。通常添加LAB后,在青贮过程中由于菌体蛋白的增加和对蛋白质水解的抑制作用,青贮饲料的CP含量会升高^[25]。但在本试验中,LAB添加剂对CP含量没有显著影响,这与Du等^[26]的试验结果一致。

随着收获时期的延后,植物体营养成分发生变化,细胞壁成分逐渐增加,细胞内容物逐渐降低,CP含量逐渐降低^[27-28]。CL、HC和ADL是植物细胞壁的主要组成成分。李诚等^[29]和章燕柳等^[30]分别发现饲草型小黑麦和水稻稻草的HC含量在各生育期之间会出现下降的趋势,这与本试验结果一致。Baba等^[31]发现在成熟期水稻茎秆中,半纤维素组分中的(1-3,1-4)-β-葡聚糖含量显著降低,说明(1-3,1-4)-β-葡聚糖含量的降低可能是本试验中HC含量降低的原因。HC含量的下降导致了晚期收获的NDF含量的下降。由于不同收获时期的草原植物群落组成不一致,加之LAB添加剂的效果受原料种类的影响^[32],这导致了LAB添加剂和收获时期对DM和EE含量的显著交互作用。

3.3 主成分分析及隶属函数分析综合评定天然牧草青贮品质

主成分分析和隶属函数分析广泛应用于综合评价作物品质^{[33⇓⇓⇓-37]}。本试验根据主成分分析结果挑选出的9个指标与评价青贮饲料质量的主要常规指标一致。隶属函数评价结果显示,使用LAB后,其平均隶属函数值均大于CK组,说明LAB可以改善青贮饲料质量。同时,随收获时期延后,LAB组得分表现为先降低后升高,这与发酵品质和营养品质结果一致。综上所述,说明基于主成分分析和隶属函数法可以有效评价青贮饲料质量。此外,晚期收获的CK组得分低于中期,但LAB添加剂组高于中期,说明LAB的使用效果受到了收获时期的影响。

4 结论

LAB添加剂可以显著改善天然牧草青贮饲料发酵品质,其效果会受天然牧草收获时期的影响。根据综合评价结果,早期和中期收获时添加LP的青贮效果更好,晚期收获时添加LB的青贮效果更好。
